Laser microdissection can be used in forensic casework to isolate specific cell types from mixtures of biological samples. Extraction of DNA from selected cells is still required prior to STR amplification. Because of the relatively pristine nature of the recovered cells, laser microdissection is more sensitive than more traditional methods of DNA analysis, theoretically resulting in DNA profiles from less cellular material. A one-tube extraction and amplification method minimises loss of DNA through liquid transfers and reduces the potential for contamination events occurring. In this paper, the development of a one-tube method for the effective extraction of DNA from laser microdissected sperm and epithelial cells is described. The performance of the in-house method was compared to that of a commercial DNA extraction kit for extraction of DNA from sperm and the downstream compatibility with STR amplification was determined for both sperm and epithelial samples. Fiscal stress and decreasing government budgets have led to renewed interest in voluntary contributions for the funding of public goods. This paper experimentally examines the Provision Point Mechanism (PPM), a voluntary contribution mechanism for the funding of lumpy public goods. Previous research has demonstrated the effectiveness of this mechanism at providing public goods, however all were conducted in an environment of complete information, which fails to capture the uncertainties of the real world. This study tests the efficacy of the PPM in informationally limited settings. We find no significant differences in the rate of successful provisions or level of group contributions when subjects have limited information about the valuations of others than when they have complete information. 相似文献

Latent fingerprints were successfully visualized using fluorescence lifetime imaging (FLIM) on paper which emits strong fluorescence with a lifetime close to that of fingerprints and thus from which it is difficult for time-resolved spectroscopy to visualize fingerprints. Latent fingerprint samples on paper were excited using a 450 nm or 532 nm nanosecond pulsed-laser, and time-resolved fluorescence images were obtained at a delay time of 6–16 ns in intervals of 1 ns, to the excitation pulse. The excitation beam was expanded using a lens, and the fluorescence from the fingerprints was captured using an intensified CCD camera. Because of the large fluorescence intensity of the background paper of approximately two to four orders of magnitude larger than that of the fingerprint, the fingerprint was not visualized on each fluorescence image by time-resolved spectroscopy. However, the fingerprint was visualized in a FLIM image constructed using a series of the fluorescence images for the case with the fluorescence intensity of the background paper being four orders of magnitude larger than that of the fingerprint. The difference in fluorescence lifetime in the FLIM image of the visualized fingerprint and background paper was in the order of 0.1 ns, which was an order of magnitude smaller than the inherent fluorescence lifetime of a few nanoseconds for the fingerprints and paper. It was demonstrated that, at a background fluorescence intensity with a certain order of magnitude larger than that of fingerprints, FLIM has the potential to visualize latent fingerprints which cannot be visualized by time-resolved spectroscopy. 相似文献

Estimating the postmortem interval of skeletal remains is difficult, as few tools exist to do so. To address this problem, we conducted a field experiment to measure the chemistry of swine (Sus scrofa domesticus Linnaeus) gravesoil after 1 year and 3 years postmortem. Carcasses were placed on the soil surface of a pasture during June in a cold (Dfa) climate. Significant (p < 0.05) increases in ninhydrin‐reactive nitrogen, pH, total nitrogen, and nitrate in gravesoil were detected 1 year postmortem. Significant differences in gravesoil chemistry were not detected 3 years postmortem. These observations coincided with gaps in plant growth 1 year postmortem and the development of lush vegetation 3 years postmortem. We conclude that these phenomena can be used to assist the decision‐making process regarding the allocation of resources during the early stages of a death investigation. 相似文献

Currently, there is no standardized protocol for multi‐detector computed tomography (MDCT) measurement of juvenile remains. Using 33 juvenile clavicles, this paper investigates a protocol to allow MDCT measurements, comparable or supplemental with traditional osteometric measurements, to be acquired for application to previously published algorithms. The results illustrate that there is no significant difference between MDCT measurements and those taken by direct osteometric methods. By presenting such a protocol, this paper takes the first steps toward validation of the process of conversion from measurement of dry juvenile bone to MDCT compatibility and allows the forensic world to take a step forward in standardizing the way MDCT is used for forensic practice. This paper assesses the limitations and potential applications of this virtual approach and offers some suggestions for where further work might progress the conversion of these new approaches into legally admissible anthropological techniques of age estimation. 相似文献

Apparatus to produce a spatially and temporally uniform heat source is described and this is used to visualize latent fingerprints deposited onto thermal paper by raising the temperature of the paper. Results show an improvement over previous research when fingerprint deposits are aged or the developed fingerprints faint; visualization being enhanced by the use of a blue LED light source of 465 nm peak wavelength. An investigation of the components in fingerprint sweat likely to affect the solubility and hence color change of the dye present in the thermal paper has shown that polar protic solvents able to donate a proton are favored and a polar amino acid found commonly in eccrine fingerprint sweat (lysine) has been shown able to produce the desired color change. Aged fingerprint deposits on thermal paper from a variety of sources up to 4 years old have been visualized with this technique. 相似文献
