Validation of the InnoXtract™ DNA extraction method for bone,teeth, and rootless hair

Forensic casework samples often include human hairs, teeth, and bones. Hairs with roots are routinely processed for DNA analysis, while rootless hairs are either not tested or processed using mitochondrial DNA. Bones and teeth are submitted for human remains identifications for missing persons and mass disaster cases. DNA extraction from these low templates and degraded samples is challenging. The new InnoXtract DNA extraction method utilizes magnetic beads that are optimized to bind small DNA fragments, as small as 100 base pairs, to purify high-yield DNA from compromised samples. This validation study evaluates InnoXtract's ability to obtain amplifiable DNA from samples such as rootless hairs and skeletal remains. Studies performed include sensitivity, stability, repeatability, reproducibility, non-probative samples, and comparison to standard organic extractions. Sensitivity studies demonstrate average yield recoveries ranging from 53% to 100% and 73% to 85% for the InnoXtract hair and bone methods, respectively. Studies demonstrate consistent results across a range of sample types, such as insulted and un-insulted bone and teeth, as well as hair shafts from donors of various ages, gender, race, and hair characteristics. The InnoXtract bone method outperformed organic extraction. The method was successfully automated on a MagMAX™ Express-96, with recoveries over 70% relative to the manual version. InnoXtract has the potential as an automated high-throughput, high-yield bone extraction method with 6 h of total extraction time for up to 96 samples. The validation study results demonstrate that the InnoXtract kits produce high-yield and high-quality DNA from compromised bone, teeth, and hair shaft samples.     

Supporting farmer participation in formal seed systems: lessons from Tharaka,Kenya

This article examines contemporary challenges of formal seed sector participation for resource-poor farmers in Tharaka and engages in a wider discussion of national seed policy and formal seed sector development in Kenya. While many farmers reported utilising modern seed varieties developed by the formal seed sector, the majority of these were introduced through seed aid and maintained through seed saving, supporting seed system integration strategies. Building on these findings, the article discusses ways in which national seed policy in Kenya might be refined to better meet national and regional development goals focused on decreasing the incidence of hunger and poverty.

Productivity Improvement and Public Personnel Management： An Analysis of Presidents Reagan and Obama Administration

Considering the financial crises, all the recent presidents have emphasized the importance of productivity improvement in government, especially in public personnel management. Though there is a general agreement on the concept of productivity, the means to achieve productivity vary with president＇s ideology. President Reagan championed the cause of conservatism. On one hand, despite Obama＇s attempt to diffuse ideological question, his actions can be regarded as liberal. This study seeks to make a comparison between Presidents Reagan and Obama on their efforts in productivity improvement in public personnel management, based on their ideological preferences. President Reagan campaigned on an anti-Washington agenda. He promoted the idea that the government is the problem rather than the solution. He popularized the idea that the people are burdened with heavy taxation as a result of the big government. Therefore, the solution to heavy taxation was to cut big government, which can be accomplished by reducing unnecessary and expensive regulations. On the other hand, when President Obama came to office, the country was faced not only with a deep recession, but two difficult wars, the collapse of the banking and the auto industries, the housing crisis, and overall capacity crisis in the government. Obama resisted ideological level in his campaign speeches to diffuse the criticisms of the conservatives.     

Inferring the population of origin of DNA evidence within the UK by allele-specific hybridization of Y-SNPs

Marked differences in Y-SNP allele frequencies between continental populations can be used to predict the biogeographic origin of a man's ancestral paternal lineage. Using 627 samples collected from individuals within the UK with pale-skinned Caucasian, dark-skinned Caucasian, African/Caribbean, South Asian, East Asian or Middle Eastern appearance we demonstrate that an individual's Y-SNP haplogroup is also strongly correlated with their physical appearance. Furthermore, experimental evaluation of the Marligen Signet Y-SNP kit in conjunction with the Luminex 100 detection instrument indicates that reliable and reproducible haplogrouping results can be obtained from 1 ng or more of target template derived from a variety of forensic evidence types including, blood, saliva and post-coital vaginal swabs. The test proved highly male-specific with reliable results being generated in the presence of a 1000-fold excess of female DNA, and no anomalous results were observed during degradation studies despite a gradual loss of typable loci. Hence, Y-SNP haplogrouping has considerable potential forensic utility in predicting likely ethnic appearance.
