Several assays for gamma-hydroxybutyrate (4-hydroxybutyrate, GHB) have been developed based on the enzyme gamma-hydroxybutyrate dehydrogenase (GHB-DH). Enzymatic oxidation of GHB by NAD+ is coupled to diaphorase-mediated reduction of pro-dye to yield colored product. GHB-DH from Ralstonia eutropha was cloned and expressed as a stable fusion protein easily purified by affinity chromatography. Quantitative initial velocity and endpoint versions of the assay in solution are described. Michaelis-Menten parameters for oxidation of GHB and ethanol were estimated. A semi-quantitative "dipstick" version of the assay on paper also is described. Both solution endpoint and "dipstick" assays are sensitive to about 0.05 mg GHB/mL using 10 microL of sample. Ethanol at concentrations possible in urine and agents used to stabilize physiological fluids for forensics analysis do not interfere significantly. The "dipstick" assay also allows detection of GHB in alcoholic beverages after evaporation of about one-fourth drop of beverage before testing. The enzymatic assay for GHB is reliable, sensitive, inexpensive and rapid.  相似文献

Thirty-six male students, drawn from a sample of 1195, were interviewed to obtain a personal history. A battery of projective psychological tests (Rorschach and TATs) were also administered to them. The students were divided into four groups of nine each, Jewish radicals (JR), Christian radicals (CR), Jewish moderates (JM), and Christian moderates (CM), to test the significance of religious background as it related to political outlook. Eight significant psychological variables were found and defined. No differences were found between JMs and CMs. Radicals differed from moderates on three variables: negative identity, masochistic surrender, and treating people as concepts. In addition, JR subjects demonstrated consistently a wandering fantasy, flight from the mother, the mother as salient, and machismo as psychological variables. CRs were not characterized by any of these variables. As with both groups of moderates, the father of the CRs was psychologically salient, but unlike the moderates, CTs perceived their fathers as flawed. Abstract. This paper presents the results of interviews carried out in twenty urban centres in British Columbia, Alberta, and Ontario on the matter of education/training programs for municipal administrators. After a brief review of literature on this topic, a composite profile of the respondents is presented. The survey results are analysed under four separate items: the need and its dimensions; awareness of and attitudes toward existing programs; anticipated attitudes of civic civil servants and politicians; and factors to be considered in designing and initiating programs for senior and middle management personnel. The study, while indicating a positive demand for programs, suggests that further field research be conducted and that a trial program be mounted to assist in evaluation. Sommaire. L'auteur de cette communication expose les résultats d'entrevues faites dans vingt centres urbains de la Colombie-Britannique, de l'Alberta, et de I'Ontario sur la question des programmes d'éducation/formation pour les administrateurs municipaux. Après une revue rapide des ouvrages sur ce sujet, il présente le profil moyen des personnes interrogées. Les réultats de l'enquête sont analysés de quatre points de we & tincts:les besoins et leur dimension; la connaissance des programmes existants et les réactions qu'ils provoquent; les attitudes antidpées des fonctionnaires et des représentants municipaux; et les facteurs A considérer pour concevoir et lancer des programmes à l'intention des cadres moyens et supérieurs. The legislative process in the tax field and the administrative process for the issuance of regulatory and interpretative material by the Treasury Department are closely coordinated. The same persons and interests both inside and outside of Government are involved in both processes. There is thus continuity of expertise, understanding of what is at stake, adversary viewpoints, and Washington knowhow. The preparation of administrative material within the Treasury is a task for those technical personnel and policy officials who in varying degrees also have a voice in the preparation of the Treasury's legislative position and the legislative development of statutory language. The outside forces who watch legislative activity also closely watch administrative activity and seek to shape the latter just so they do the former. The response of the Treasury is much the same in both situations, and governed by the policies, attitudes and predilections of tax policy officials and career personnel.  相似文献
