The distribution of Y chromosomal haplotypes and haplogroups in two different population samples from the Romagna region (North Italy) was performed. One population sample was collected in the urban area of Rimini, an ancient port in Roman age and the other one in the near and geographically more isolated rural area of Valmarecchia. Fast and slow evolving markers have been studied to infer population history and to analyse the microgeographic heterogeneity of Y chromosome in a Northern Italian region. Haplotype variability values compared to those observed in a Central Italy sample from the other side of Apennine mountains and in an Austrian population sample were very similar. 相似文献

Short amplicon autosomal short tandem repeat (Mini-STR) assay has proved to be a highly useful tool in forensic applications, especially for highly degraded DNA samples that typically result in partial profiles and total loss of information from regular STR amplicons.In this study two new quadruplex systems were designed to get nuclear DNA profile from degraded forensic casework samples. In order to obtain PCR products less than 120 bp in size, primer pairs of eight STR markers, included in available commercially multiplex PCR kits, were redesigned and assembled in two PCR-multiplexes: D8S1179, D3S1358, TPOX, D16S539 and CSF1P0, TH01, D13S317, D5S818.After validation, these two Mini-STR quadruplex were employed in paternity testing case that involved DNA extraction from archival postmortem Bouin's fluid-fixed paraffin-embedded tissue where commercial kit yielded low success. The results obtained with the present Mini-STR PCR-multiplexes proved clearly demonstrating their usefulness in analyzing degraded DNA samples. 相似文献

ABSTRACT Since 2008, the international economic and financial crisis has been affecting the living and working conditions of European citizens in different ways and scope. Yet, the pattern is of rising unemployment, social deprivation and poverty, cuts in health, education and social security budgets. These negative socio-economic conditions have led to major transformations in collective responses, which, among others, take place through Alternative Action Organisations (AAOs). The specific organizations carry out non-mainstream activities that primarily target the economic and the social well-being of citizens, including their basic needs, health and lifestyles. Using quantitative data from the LIVEWHAT project and drawing on social origins theory and resource mobilization theory, the article investigates AAOs’ main characteristics across four European countries that have been differently affected from the recent recession as well as belong to different welfare state and third sector regimes, including Germany, Greece, Sweden and Poland. The findings stress the importance of considering the particular combinations of the welfare state and third sector regimes as well as the severity of the experienced economic crisis in understating the variation in AAOs’ main features under a comparative perspective. 相似文献

Consuming honey can result in adverse effects owing to poisoning by bacterial (botulism) or plant toxins. We have devised a method to extract polymerase chain reaction (PCR) amplifiable DNA of up to c. 400 bp in length based on dialysis of a 15-mL honey sample for 18 h against deionized water followed by sequential extraction using phenol, phenol/chloroform/isoamyl alcohol, chloroform/isoamyl alcohol, and ether. Sequence analysis of PCR products obtained using "universal" plant, fungal, and bacterial primers targeted to the ribosomal RNA genes has allowed us to identify six different orders of plants (Apiales, Fabales, Asterales, Solanales, Brassicales, and Sapindales), two orders of fungi (Entylomatales and Saccharomycetales), and six orders of bacteria (Sphingomonadales, Burkholderiales, Pseudomonadales, Enterobacteriales, Actinomycetales, and Bifidobacteriales) in a single honey specimen. 相似文献

The analysis of the genetic structure of regions with a complex demographic history shed light on the various topographic, linguistic and historical influences which form the present genetic landscape of Europe. In the Emilia-Romagna region (North of Italy) Ravenna is a geographical area with a historical complex background: it was an important seaport on the Mediterranean sea, the capital of the Ostrogothic kingdom of Italy and the seat of the Byzantine governor of Italy. The purpose of this study was to investigate the microgeographic variation of Y chromosome haplotypes of the area of Ravenna by analyzing 17 Y-chromosome short tandem repeats (Y-STRs) in 122 unrelated males. 100% of all haplotypes were different. A comparison with neighbouring Italian as well as with European and Levante root populations was done by AMOVA and visualized by a phylogenetic tree. The two main haplogroups found in this area were R1b and E3b1. The results of the present study add to the data for the forensic databases and can be useful also for anthropological studies. 相似文献

We report the results of an attempt to identify the supposed remains of a famous World War I (WWI) Italian soldier who was killed in battle along the Italian front in 1915.Thanks to the availability of offspring from both paternal and maternal lineage Y-STRs and mtDNA were analysed and both showed a clear exclusion scenario: the remains did not belong to the supposed war hero.This is the first effort of identification of the remains of soldiers who perished during World War I within a multidisciplinary project aimed at the retrieval of historical and cultural aspects linked to WWI, and the systematic study of the remains of soldiers and ultimately their identification. This last step involves both Italian and Austrian laboratories. 相似文献

Results from a collaborative exercise with proficiency testing conducted by 20 Italian laboratories on the 15 loci included in the Identifiler kit were analyzed by allele sharing methods and by standard population genetics tests. The validated database, including about 1500 subjects, was merged with that of a previous exercise conducted on nine loci, and the resulting allele frequencies, subdivided by Italian region, were published on-line. 相似文献
