Quantitative analysis of chlorpromazine by electron spin resonance (ESR) spectroscopy

A simple and sensitive method is described for quantitative analysis of chlorpromazine in blood, serum, urine and tissue homogenate. The chlorpromazine cation radical produced by adding perchloric acid and 2,3-dichloro-5,6-dicyano-p-benzoquinone to the sample can be detected by the ESR method at room temperature. The sensitivity limit is 10 ng, that is, 20 μl of the solution containing 0.5 μg chlorpromazine/ml. The time needed for the measurement is within 10 min. The chlorpromazine radical thus produced is very stable; for example, 95% of the radical was observed after 24 h. The advantage of this method is discussed by comparing with the ordinary spectrophotometry which requires the purification of the sample.     

Police officer attitudes toward peers, supervisors, and citizens: A comparison between field training officers and regular officers

This article assesses whether police Field Training Officers (FTOs) display patterns of attitudes that distinguish them from non-field training officers. The analysis focuses on attitudes toward four important groups: fellow officers, immediate supervisors, top managers, and neighborhood residents. Interview data used were collected from Indianapolis Police Department (IPD) during the summer of 1996. Findings show that FTOs are more critical of their immediate supervisors and district managers than non-FTOs, whereas FTOs and non-FTOs hold similar attitudes toward their co-workers and neighborhood residents. Implications for future research and policy are discussed.     

Detection of ABH blood group antigens in the saliva of Koreans and their stability according to storage of saliva samples

The purpose of the present study was to identify salivary molecules carrying the ABH blood group antigens in Koreans and to investigate the changes in these antigens according to processing and storage of saliva samples. Secretor or non-secretor phenotypes and salivary components carrying the ABH antigens were identified in 90 subjects, 30 subjects in each ABO blood group, by SDS-PAGE and immunoblotting. Saliva samples were then obtained from 12 secretors-two males and two females in each ABO blood group and aliquots of both fresh saliva samples and their supernatants after centrifugation were stored at room temperature, 4, -20 and -70 degrees C. The same experiments were performed after 1, 3 and 6 months to investigate changes in the blood group antigens. In all 68 secretors, high-molecular-weight salivary mucin (MG1) was found to be the primary carrier of the ABH antigens. A salivary component of approximately 80 kDa also carried H antigen in seven saliva samples of 22 blood type O secretors. The blood group antigens were better detected in centrifuged samples. In saliva samples preserved at room temperature and 4 degrees C, the blood group antigens were either not detected or detected as degraded molecules. No change was found in the blood group antigens in saliva samples preserved at -20 and -70 degrees C for 6 months.     

Analysis of rosin and modified rosin esters in adhesives by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS)

The optimum method for analyzing glyceryl rosinate by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was studied. The performances of dithranol, anthracene, and 2,5-dihydroxybenzoic acid, pyrene, trans,trans-1,4-diphenyl-1,3-butadiene, 9-nitroanthracene, indole-3-acrylic acid and retinoic acid as a matrix agent, and those of sodium trifluoroacetate (NaTFA) and silver trifluoroacetate (AgTFA) as a cationization agent were measured. Dithranol showed higher signal to noise ratio and lower base-line, and AgTFA showed better performance than NaTFA. In this study, however, NaTFA was chosen since the ester peaks with high signal to noise ratios were observed, and the spectra of sodium-cationized molecules are simpler than those of silver-cationized molecules. Rosin esters and modified rosin esters in 22 rubber-based pressure sensitive adhesives and a paper-cement were analyzed. Glyceryl rosinate, glyceryl disproportionated rosinate, pentaerythrityl rosinate and pentaerythrityl hydrogenated rosinate were easily detected despite the fact that rubber-based adhesives are complex mixtures of elastomers, tackifiers, antioxidants and other additives. The detection limit of this method was studied and 2% of glyceryl rosinate in a rubber-based pressure sensitive adhesive was detected. It has been proved that MALDI-TOF-MS is a useful analytical method to analyze rosin and modified rosin esters in adhesives.     

Detection and validated quantification of 21 benzodiazepines and 3 "z-drugs" in human hair by LC-MS/MS

A method for detection and quantification of 21 benzodiazepines and the pharmacologically related "z-drugs" in human hair samples was developed and fully validated using liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). After methanolic and methanolic/aqueous extraction, the analytes were separated using two different LC-MS systems (AB Sciex 3200 QTRAP and AB Sciex 5500 QTRAP). Separation columns, mobile phases and MS modes for both systems were: Phenomenex Kinetex, 2.6 μm, 50/2.1; 5mM ammonium formate buffer pH 3.5/methanol, total flow 0.75 mL/min; electrospray ionization (ESI), multiple reaction monitoring (MRM), information dependent acquisition (IDA), enhanced product ion scan (EPI). The assays were found to be selective for the tested compounds (alprazolam, 7-aminoclonazepam, 7-aminoflunitrazepam, bromazepam, chlordiazepoxide, clonazepam, N-desalkylflurazepam, diazepam, flunitrazepam, flurazepam, alpha-hydroxymidazolam, lorazepam, lormetazepam, midazolam, nitrazepam, nordazepam, oxazepam, phenazepam, prazepam, temazepam, triazolam, zaleplon, zolpidem and zopiclone), all validation criteria were in the required ranges according to international guidelines, except for bromazepam. Matrix effects, and process efficiencies were in the acceptable ranges evaluated using the post-extraction addition approach. Lower limits of quantification were between 0.6 and 16 pg/mg of hair. The LC-MS/MS assay has proven to be applicable for determination of the studied analytes in human hair in numerous authentic cases (n=175).     

From the Inside/Out: Greene County Jail Inmates on Restorative ReEntry

The application of criminal justice sanctions is often misguided by a failure to recognize the need for a comprehensive approach in the transformation of offenders into law-abiding citizens. Restorative justice is a growing movement within criminal justice that recognizes the disconnect between offender rehabilitative measures and the social dynamics within which offender reentry takes place. By using restorative approaches to justice, what one hopes of these alternative processes is that the offenders become reconnected to the community and its values, something rarely seen in retributive models in which punishment is imposed and offenders can often experience further alienation from society. In this study, the authors wish to examine factors that contribute to failed prisoner reentry and reintegration and explore how restorative reintegration processes can address these factors as well as the needs, attitudes, and perceptions that help construct and maintain many of the obstacles and barriers returning inmates face when attempting to reintegrate into society.
