白介素21的研究进展及其在寄生虫学上的应用

白介素21(interleukin-21,IL-21)是近年来报道的一种新的细胞因子,主要由活化的CD4+T细胞、NK细胞和Th17细胞产生,其受体是Ⅰ型细胞因子受体家族成员之一,具有共同的γ链受体。IL-21与其受体结合后可调节B细胞的增殖,促进T细胞、NK细胞的增殖、分化并能提高NK细胞杀伤活性。全文介绍了IL-21的分子结构和生物学活性,阐述了IL-21在寄生虫感染中的作用,旨在为进一步研究IL-21在寄生虫学上的应用提供参考。     

Sudden Death Secondary to an Undiagnosed B‐Cell Lymphoma of the Hypopharynx and Infiltration of the Inferior Constrictor Muscle

The aim of this presentation was to share an uncommon form of sudden death, suffered by a 64‐year‐old woman, due to a mechanical obstruction of hypopharynx by an undiagnosed B‐cell lymphoma, infiltrating the inferior pharyngeal constrictor muscle. A forensic approach by means of scene investigation, circumstantial data collection, autopsy, and histological and toxicological investigations led to conclude that the cause of death was asphyxia, correlated with B‐cell lymphoma of the hypopharynx. The autopsy examination highlighted the presence of a wall thickening, infiltrating, and projecting into the hypopharynx lumen. The histological analysis showed the essential finding of a B‐cell lymphoma of the hypopharynx, diffusely infiltrating the inferior pharyngeal constrictor muscle. To conclude, this case demonstrates once more that in the absence of specific data, a thorough forensic investigation including autopsy, histological examination, and circumstantial data collection is mandatory to reach a correct cause of death.     

Opinion 1/15 of the Grand Chamber dated 26 July 2017 about the agreement on Passenger Name Record data between the EU and Canada

On 26 July 2017, the Grand Chamber of the European Court of Justice rendered its seminal Opinion 1/15 about the agreement on Passenger Name Record data between the EU and Canada. The Grand Chamber considered that the decision of the Council about the conclusion, on behalf of the Union, of the agreement between the EU and Canada about the transfer and processing of PNR data must be based jointly on Article 16(2) about the protection of personal data and Article 87(2)(a) about police co-operation among member states in criminal matters, but not on Article 82(1)(d) about judicial co-operation in criminal matters in the EU of the Treaty on the Functioning of the EU. The Grand Chamber also considered that the agreement is incompatible with Article 7 on the right to respect for private life, Article 8 on the right to the protection of personal data, Article 21 on non-discrimination and Article 52(1) on the principle of proportionality of the Charter of Fundamental Rights of the EU since it does not preclude the transfer, use and retention of sensitive data. In addition to the requirement to exclude such data, the Grand Chamber listed seven requirements that the agreement must include, specify, limit or guarantee to be compatible with the Charter.The opinion of the Grand Chamber has far-reaching implications for the agreement on PNR data between the EU and Canada. It has also far-reaching implications for international agreements on PNR data between the EU and other third states. Last, it has far-reaching implications for Directive 681 of 27 April 2016 on PNR data.     

新藤黄酸对人表皮癌细胞A431增殖和凋亡的影响

目的 观察新藤黄酸（gambogenic acid,GNA）对人表皮癌细胞A431增殖和凋亡的影响。〖JP〗方法 以体外培养的人表皮癌A431细胞株为研究对象,采用甲基噻唑基四唑检测GNA对A431细胞增殖活性的影响;倒置显微镜下观察GNA对A431细胞株细胞形态的影响;荧光显微镜下观察GNA对Hoechst 33342染色的A431细胞凋亡的影响;采用流式细胞仪检测膜联蛋白Ⅴ-异硫氰酸荧光素/碘化丙碇双染的A431细胞的凋亡率。结果 随着GNA剂量的增大,GNA对A431细胞增殖活性的抑制作用增强。倒置显微镜和荧光显微镜下观察显示,GNA作用的A431细胞形态发生明显的变化并出现显著的凋亡特征。流式细胞仪检测显示A431细胞凋亡率随着GNA作用剂量的增大而升高。结论 0.75~12.0 μmol/L GNA能够剂量依赖性地抑制A431的增殖。     

Workplace Assessment of Targeted Violence Risk: The Development and Reliability of the WAVR‐21

This study describes the development of the WAVR‐21, a structured professional judgment guide for the assessment of workplace targeted violence, and presents initial interrater reliability results. The 21‐item instrument codes both static and dynamic risk factors and change, if any, over time. Five critical items or red flag indicators assess violent motives, ideation, intent, weapons skill, and pre‐attack planning. Additional items assess the contribution of mental disorder, negative personality factors, situational factors, and a protective factor. Eleven raters each rated 12 randomly assigned cases from actual files of workplace threat scenarios. Summary interrater reliability correlation coefficients (ICCs) for overall presence of risk factors, risk of violence, and seriousness of the violent act were in the fair to good range, similar to other structured professional judgment instruments. A subgroup of psychologists who were coders produced an ICC of 0.76 for overall presence of risk factors. Some of the individual items had poor reliability for both clinical and statistical reasons. The WAVR‐21 appears to improve the structuring and organizing of empirically based risk‐relevant data and may enhance communication and decision making.     

Fetal Age Estimation Using Orbital Measurements: 3D CT-Scan Study Including the Effects of Trisomy 21

Abstract:  This study evaluates a new method of fetal age estimation based on orbital measurement and including the potential trisomy 21 of the fetus. Six orbital and one facial CT-scan measurements were taken on 71 fetuses ranging from 14 to 41 weeks of gestational age. Forty-eight fetuses were "normal" and 23 fetuses presented the Down syndrome (trisomy 21). The anatomy/imagery correspondence was evaluated comparing our results to those obtained by direct bone measurements on the same fetuses and revealed no significant differences between the two kinds of measurements. Moreover, a multiple linear stepwise regression was realized to estimate fetal age and showed that the "trisomy 21 parameter" is not conserved in the final determination model. Therefore, we conclude that the good radio-anatomical correspondence offers an interesting alternative to direct bone measurement (necessitating dissections) and allows a reliable fetal age determination, whatever is the trisomy 21 condition of the fetus.     

Concordance Between the AmpFℓSTR® MiniFiler™ and AmpFℓSTR® Identifiler® PCR Amplification Kits in the Kuwaiti Population

Abstract:  The AmpFℓSTR^® MiniFiler™ polymerase chain reaction amplification kit, developed and supplied by Applied Biosystems, complements the AmpFℓSTR^® Identifiler^® polymerase chain reaction amplification kit (Applied Biosystems, Warrington, U.K.) by improving the success rate when profiling DNA that is degraded or contains inhibitors. Before applying the MiniFiler™ kit to casework, the profiles from 200 unrelated Kuwaitis were compared to Identifiler^® profiles. Concordance was observed for 99.875% (1598 of 1600) of the compared STR loci. The two discordant profiles displayed allelic dropout: one at the D13S317 locus due to nonamplification of allele 10 in the MiniFiler™ profile, and one at the D18S51 locus due to nonamplification of allele 18 in the Identifiler^® profile.     

Validation of the AMPFℓSTR® MiniFilerTM PCR Amplification Kit for Use in Forensic Casework*

Abstract: The AmpF?STR^® MiniFiler^TM PCR Amplification Kit is designed to genotype degraded and/or inhibited DNA samples when the AmpF?STR^® Identifiler^TM PCR Amplification Kit is incapable of generating a complete genetic profile. Validation experiments, following the SWGDAM guidelines, were designed to evaluate the performance of MiniFiler. Data obtained demonstrated that MiniFiler, when used in conjunction with Identifiler, provided an increased ability to obtain genetic profiles from challenged samples. The optimum template range was found to be between 0.2 and 0.6 ng, with 0.3 ng yielding the best results. Full concordance was achieved between the MiniFiler kit and Identifiler kit except in a single case of a null allele at locus D21S11. Numerous instances of severe heterozygous peak imbalance (<50%) were observed in single source samples amplified within the optimum range of input DNA suggesting that caution be taken when attempting to deduce component genotypes in a mixture.     

Validation of the Short Amplicon Multiplex Q8 Including the German DNA Database Systems*

Abstract: We have developed a concept to enable the analyzing of degraded stains with limited DNA template quantity. Therefore we have constructed a short tandem repeat (STR) multiplex including the German DNA database systems (Q8). The amplicon lengths are smaller than 280 bp. For the validation of Q8 over 50 degraded samples were investigated. Amplifications were performed with “low copy number” PCR, the number of PCR cycles was increased to 33 and the reaction volume was decreased to 12.5 μL. Compared with the MPX2 and Nonaplex kit, the average success rate was increased using the Q8 kit by approximately 20% and 30%, respectively. The efficiency of a sensitive STR multiplex with reduced amplicon lengths was confirmed in comparing the success rates of Q8 for typing degraded samples and samples with limited amount of DNA template while partial profiles were observed with the majority of the samples using commercially available kits.
