微小隐孢子虫感染HCT-8细胞模型的建立及不同阶段虫体形态的观察

利用人回盲肠癌(HCT-8)细胞作为感染细胞,采用不同的接种细胞浓度和培养血清浓度进行培养,观察细胞的生长状况;在培养细胞中加入1×105个经活力检测的微小隐孢子虫卵囊,培养72h后用荧光标记的隐孢子虫卵囊、子孢子染色试剂盒Crypt-a-Glo和Sporo-Glo进行染色,观察不同发育阶段的隐孢子虫形态。结果显示,在六孔培养板中接种10×105个细胞后,24h后长成80%～100%单细胞层,可以用来接种微小隐孢子虫。培养72h后观察到了隐孢子虫各个阶段虫体的形态。结果表明,成功建立了微小隐孢子虫感染HCT-8细胞模型。     

Sudden Death Secondary to an Undiagnosed B‐Cell Lymphoma of the Hypopharynx and Infiltration of the Inferior Constrictor Muscle

The aim of this presentation was to share an uncommon form of sudden death, suffered by a 64‐year‐old woman, due to a mechanical obstruction of hypopharynx by an undiagnosed B‐cell lymphoma, infiltrating the inferior pharyngeal constrictor muscle. A forensic approach by means of scene investigation, circumstantial data collection, autopsy, and histological and toxicological investigations led to conclude that the cause of death was asphyxia, correlated with B‐cell lymphoma of the hypopharynx. The autopsy examination highlighted the presence of a wall thickening, infiltrating, and projecting into the hypopharynx lumen. The histological analysis showed the essential finding of a B‐cell lymphoma of the hypopharynx, diffusely infiltrating the inferior pharyngeal constrictor muscle. To conclude, this case demonstrates once more that in the absence of specific data, a thorough forensic investigation including autopsy, histological examination, and circumstantial data collection is mandatory to reach a correct cause of death.     

Opinion 1/15 of the Grand Chamber dated 26 July 2017 about the agreement on Passenger Name Record data between the EU and Canada

On 26 July 2017, the Grand Chamber of the European Court of Justice rendered its seminal Opinion 1/15 about the agreement on Passenger Name Record data between the EU and Canada. The Grand Chamber considered that the decision of the Council about the conclusion, on behalf of the Union, of the agreement between the EU and Canada about the transfer and processing of PNR data must be based jointly on Article 16(2) about the protection of personal data and Article 87(2)(a) about police co-operation among member states in criminal matters, but not on Article 82(1)(d) about judicial co-operation in criminal matters in the EU of the Treaty on the Functioning of the EU. The Grand Chamber also considered that the agreement is incompatible with Article 7 on the right to respect for private life, Article 8 on the right to the protection of personal data, Article 21 on non-discrimination and Article 52(1) on the principle of proportionality of the Charter of Fundamental Rights of the EU since it does not preclude the transfer, use and retention of sensitive data. In addition to the requirement to exclude such data, the Grand Chamber listed seven requirements that the agreement must include, specify, limit or guarantee to be compatible with the Charter.The opinion of the Grand Chamber has far-reaching implications for the agreement on PNR data between the EU and Canada. It has also far-reaching implications for international agreements on PNR data between the EU and other third states. Last, it has far-reaching implications for Directive 681 of 27 April 2016 on PNR data.     

新藤黄酸对人表皮癌细胞A431增殖和凋亡的影响

目的 观察新藤黄酸（gambogenic acid,GNA）对人表皮癌细胞A431增殖和凋亡的影响。〖JP〗方法 以体外培养的人表皮癌A431细胞株为研究对象,采用甲基噻唑基四唑检测GNA对A431细胞增殖活性的影响;倒置显微镜下观察GNA对A431细胞株细胞形态的影响;荧光显微镜下观察GNA对Hoechst 33342染色的A431细胞凋亡的影响;采用流式细胞仪检测膜联蛋白Ⅴ-异硫氰酸荧光素/碘化丙碇双染的A431细胞的凋亡率。结果 随着GNA剂量的增大,GNA对A431细胞增殖活性的抑制作用增强。倒置显微镜和荧光显微镜下观察显示,GNA作用的A431细胞形态发生明显的变化并出现显著的凋亡特征。流式细胞仪检测显示A431细胞凋亡率随着GNA作用剂量的增大而升高。结论 0.75~12.0 μmol/L GNA能够剂量依赖性地抑制A431的增殖。     

Local response to federal changes in the housing voucher program: A case study of intraregional cooperation

Abstract

The desire to increase residential choice for Section 8 voucher clients resulted in the adoption of portability by the U.S. Department of Housing and Urban Development. Portability allows voucher holders to move between the jurisdictions of local housing authorities (HAs). InterHA cooperation could ease the administrative and financial burdens imposed by portability and improve service to voucher recipients. However, voluntary regional cooperation is rare.

This article presents a case study of a successful, voluntary, intraregional cooperative agreement among HAs. Theoretical and empirical analyses suggest that a cooperative agreement is more likely to develop voluntarily if two conditions are present: rational self‐interest and shared norms and trust among the managers. Agreements can ease the burdens associated with portability, but it is important for the parties to regularly assess implementation issues to ensure the agreement's continuing effectiveness. The article concludes with policy implications based on the findings from this research.     

Response to Herman and Chomsky

Under what conditions can political elites influence elections to favor their preferred policy outcomes by strategically crafting the language printed on the ballot? Drawing on psychological and political theories of voter cognition, we design a survey experiment to assess the degree to which ballot text can influence voter behavior in direct democracy elections and identify factors that may moderate such effects. We show that the language used to describe a ballot measure does indeed have the potential to affect election outcomes, including measures dealing with contentious social issues affecting individual rights. We also find, however, that exposing individuals to basic campaign information—in our case, endorsements from prominent interest groups—greatly attenuates the framing effects of ballot text. Our results suggest that the extent to which ballot text matters depends on the vibrancy of the campaign environment and other information available to voters.     

Validation of the Short Amplicon Multiplex Q8 Including the German DNA Database Systems*

Abstract: We have developed a concept to enable the analyzing of degraded stains with limited DNA template quantity. Therefore we have constructed a short tandem repeat (STR) multiplex including the German DNA database systems (Q8). The amplicon lengths are smaller than 280 bp. For the validation of Q8 over 50 degraded samples were investigated. Amplifications were performed with “low copy number” PCR, the number of PCR cycles was increased to 33 and the reaction volume was decreased to 12.5 μL. Compared with the MPX2 and Nonaplex kit, the average success rate was increased using the Q8 kit by approximately 20% and 30%, respectively. The efficiency of a sensitive STR multiplex with reduced amplicon lengths was confirmed in comparing the success rates of Q8 for typing degraded samples and samples with limited amount of DNA template while partial profiles were observed with the majority of the samples using commercially available kits.

3种方法联合运用提取人体脱落上皮细胞DNA

目的寻求提高法医物证检验中附有人体脱落细胞载体检材的DNA检出率的方法。方法根据案件检材的具体条件．选择3种提取方法即Chelex-100法、Chelex-100联合有机法和Chelex-100联合磁珠法。结果采用了分步提取DNA法,可充分、有效地提取微量DNA,提高了人体脱落上皮细胞有效DNA的检出率。结论根据第一步提取方法的检测结果,来调整下一步的提取策略,如是否加以纯化浓缩,可增加人体脱落上皮细胞有效DNA的检出率。
