Developmental validation of the novel six-dye GoldeneyeTM DNA ID System 35InDel kit for forensic application

Insertion/deletion polymorphisms (InDels) have been treated as a prospective and helpful genetic marker in the fields of forensic human identification, anthropology and population genetics for the past few years. In this study, we developed a six-dye multiplex typing system consisting of 34 autosomal InDels and Amelogenin for forensic application. The contained InDels were specifically selected for Chinese population with the MAF ≥ 0.25 in East Asia, which do not overlap with the markers of Investigator^® DIPplex kit. The typing system was named as Goldeneye^TM DNA ID System 35InDel Kit, and a series of developmental validation studies including repeatability/reproducibility, concordance, accuracy, sensitivity, stability, species specificity and population genetics were conducted on this kit. We confirmed that the 35InDel kit is precise, sensitive, species specific and robust for forensic practice. Moreover, the 35InDel kit is capable of typing DNA extracted from forensic routine case-type samples as well as degraded samples and mixture samples. All markers are proved to be highly polymorphic with an average observed heterozygosity (He) of 0.4582. The combined power of discrimination (CPD) is 0.999 999 999 999 978 and the combined power of exclusion in duos (CPE_D) and trios (CPE_T) are 0.978 837 and 0.999573, respectively, which are higher than those of the Investigator^® DIPplex kit. Thus, the Goldeneye^TM DNA ID System 35InDel kit is suitable for forensic human identification and could serve as a supplementary typing system for paternity testing.Supplemental data for this article is available online at https://doi.org/10.1080/20961790.2021.1945723 .     

Delayed elimination in humans after ingestion of colchicine: Two fatal cases of colchicine poisoning

Colchicine has been widely used in the treatment of acute gout over the years, but it has a narrow therapeutic index, and overdose can be life threatening. A method using ultra-high-performance liquid chromatography–tandem mass spectrometry system was applied in two fatal cases of colchicine poisoning in this study to the determination of colchicine in blood. In case 1, a 19-year-old man suffered from depression and ingested 160 colchicine tablets (each 0.5 mg). The concentration of colchicine in his blood samples showed a fluctuating trend and kept above the therapeutic steady-state concentration for 5 days. In case 2, a 70-year-old female patient with a history of gout and chronic colchicine intake ingested five times the usual dose of colchicine (5 mg) and died after 12 days of medical care, with 5 ng/mL of colchicine in her blood sample. Our findings suggest that the delayed elimination and accumulation in humans after colchicine overdose could keep the concentration of colchicine maintaining above the therapeutic steady-state concentration for many days before dying, probably along with a fluctuating trend.

The Greek Conundrum

Syriza lost the July 2019 election in Greece to the right‐wing New Democracy Party, though it was not a crushing defeat. This article explains that although Syriza is chiefly responsible for the return of New Democracy to power, its remarkable electoral performance is because its party elites, being in power for over four years, succeeded in appropriating the state machine, establishing caucuses of power, influence and clientelism. Thus, the demise of the Panhellenic Socialist Movement (PASOK), upon which Syriza capitalised in full, led to the establishment of a new two (state) party system dominated by New Democracy and Syriza. The extreme left and the extreme right ceased to play a major role in this new political scene.     

犬瘟热病毒SD(14)7株对比格犬的致病性研究

对3~4月龄比格犬经肌肉注射方式接种犬瘟热病毒(CDV)SD(14)7株1 mL(病毒含量为106.0 TCID50/mL),并进行体温、淋巴细胞数、眼鼻直肠排毒和组织病毒含量及分布等指标检测,评价变异株SD(14)7对比格犬的致病水平。结果显示,CDV感染后第2天比格犬出现CDV感染的典型特征,如眼和鼻分泌物增多,淋巴细胞明显减少,然而动物的体温变化无明显的“双相热”特征。CDV定量检测结果显示,比格犬攻毒后第10天开始通过眼鼻和直肠向外界排毒,CDV在脾中含量最高。病理剖检可见脾有坏死、肺炎症出血等,病理组织HE和免疫组织化学检测发现脾有严重的充血,淋巴细胞中有CDV抗原,证实该毒株主要感染淋巴组织和器官。此外,比格犬大脑部分神经细胞严重肿胀,在脑中可检测到CDV,表明SD(14)7株具有一定的神经嗜性。本试验结果为进一步鉴定犬瘟热病毒感染比格犬的毒力因子和免疫原性以及疫苗免疫效果的评价奠定了基础。     

Computer Self-Efficacy and e-Government Service Adoption: The Moderating Role of Age as a Demographic Factor

This article investigated the moderating role of age on the positive relationship between computer self-efficacy (CSE) and intention to use e-government services.The Technology Acceptance Model was used as the theoretical framework for this study while the data was analyzed with SPSS. The results showed that age as a demographic factor was significant and positively moderates the impact of CSE on the intention to use e-government services. Also, CSE was a significant predictor of the intention to use but age does not have a direct significant impact on the intention to use. The implications of these findings are discussed.     

Does women’s empowerment through policy intervention reduce food insecurity in South Africa?

This article determines the extent to which the food insecurity status of women in South Africa can be reduced by participating in gardening around the homestead, using survey data from 207 women farmers. A household food insecurity average score index and an empirical model that accounts for observed and unobserved factors contributing to food insecurity were employed. The findings reveal that participating in the programme worked to reduce the food insecurity of women significantly, and also show other policy factors that need to be taken into consideration to reduce food insecurity. We recommend that policy interventions towards promoting high participation by women are a sustainable strategy to address food insecurity and to achieve the 2030 agenda targeting an end to poverty and hunger.     

Integrating gender into index-based agricultural insurance: a focus on South Africa

Index insurance is an agricultural risk management tool that can provide a safety net for smallholder farmers experiencing climate risk. While uptake and scale-out of index insurance may be slow among smallholders, we can learn from experiences that demonstrate where crop insurance can protect smallholders’ livelihoods from climate risk. Integrating gender into climate risk management is necessary to ensure that the benefits of index insurance are experienced by both men and women. A dedicated intention to integrate gender may be required. Taking South Africa as a case study, the potential for gender-sensitive index insurance scale-out among smallholders is investigated.
