用分子克隆技术构建D12S375基因座等位基因分型标准物及汉、回、维族群体遗传学研究

采用分子克隆技术大量制备STR基因座等位基因分型标准物 ,解决长期困扰STR分型的准确性和标准化问题。PCR扩增出D12S375基因座的 7个等位基因片段 ,将其插入pUC重组质粒中 ,经DNA测序分析证实插入片段的结构及大小 ,按重复单位的重复次数对插入的等位基因片段进行命名 ,最后经转染、扩增及再鉴定后制备出标准D12S375基因座等位基因分型标准物。应用此分型标准物 ,调查D12S375基因座在成都汉族 ,甘肃回族及新疆维族群体中的基因型分布频率 ,评估其在法医学实践中的应用价值。     

中国武汉汉族群体STR D13S325基因座的多态性研究

应用PCR ,聚丙稀酰胺凝胶垂直电泳及银染技术对 2 16例中国汉族人群STRD13S32 5基因座进行了多态性研究 ,首次获得了中国武汉汉族人群基因频率分布资料。检出 11个等位基因 ,30个基因型 ,基因频率分布符合Hardy Weinberg平衡。DP值为 0 92 92 ,三联体PE值 0 6 137,二联体PE值为 0 4370 ,PIC值为 0 775 5。观察 2 0 0次减数分裂未发现突变基因。研究证实D13S32 5是一个高多态性STR基因座标记系统。     

Verbücherung einer Grunddienstbarkeit als Zubehör-WE zugunsten des jeweiligen Wohnungseigentümers eines WE-Objekts?

Falls Objekte zubeh?rtauglich sind (hier: 2 in einer Garage befindliche Kfz-Abstellpl?tze nach § 1 Abs 2 WEG 1975), k?nnen sie – die entsprechende Widmung und Erfassung im Rahmen der Nutzwertfestsetzung (hier durch Au?erstreitrichterbeschluss aus dem Jahr 1995) vorausgesetzt – als Grunddienstbarkeit zugunsten des jeweiligen Wohnungseigentümers eines WE-Objekts im Grundbuch einverleibt werden, wodurch die sachenrechtliche Zuordnung zum WE-Objekt erfolgt. Als Rechtsgrund für die Verbücherung des Zubeh?r-WE in Form einer Grunddienstbarkeit scheidet allerdings eine "Amtsbest?tigung" nach § 182 Abs 3 Au?StrG (früher § 178 Au?StrG aF) aus, da hiefür nur zu Lebzeiten des Erblassers bestehende und nicht erst nach seinem Tod zu begründende (beschr?nkte) dingliche Rechte in Frage kommen. Zum grundbücherlichen Zweck der "Amtsbest?tigung". Den Antrag nach § 182 Abs 3 Au?StrG kann nicht der Erbe, sondern nur der Verm?chtnisnehmer mit Zustimmung des/der Erben stellen. An allgemeinen Teilen der WE-Liegenschaft k?nnen entweder alle Wohnungseigentümer gemeinsam oder zumindest mit Zustimmung s?mtlicher Teilhaber zu Gunsten oder zu Lasten der jeweiligen Mindestanteile Grunddienstbarkeiten durch Einverleibung im Grundbuch erwerben. Die Dienstbarkeitsbelastung ist mit dem ausschlie?lichen Nutzungs- und Verfügungsrecht des einzelnen Wohnungseigentümers begrenzt.     

Creative internationalization: widening the perspectives on analysis and policy regarding international R&D activities

Internationalization in R&D is further growing; it is changing its geographical balance, as it is shifting somewhat to the Far East, and its nature, increasing the global quest for talent and good research conditions as well as for low cost R&D. This paper focuses on the European perspective, i.e. it discusses current challenges Europe faces vis-à-vis trends in industrial R&D, but the findings and arguments are more general ones. It argues that our perspective on internationalization is still shaped too much by a zero sum-rationale, whereby one location wins R&D capacity that another location loses. It develops a cost–benefit matrix in order to capture the overall costs and benefits of international R&D activities more broadly. The paper argues that more creativity is needed, that our perspective needs to be broadened to tackle all variables conditioning international activities in R&D (including local conditions of demand and discourse) and to stress the importance of the absorption of global knowledge by as many actors within an innovation system as possible. On the basis of recent survey data the paper furthermore concludes that public research should be thought of as a trans-national transmission belt of knowledge and as the prime factor that shapes the attractiveness and effectiveness of a location for business R&D. Finally, it is argued that policy schemes geared towards international R&D need to accept and tackle the issue of co-ordination of governance and to take advantage of the flexible possibilities offered at the European level, beyond the logic of the European Framework Programme.

Sequence analysis of a new short tandem repeat locus D17S2266E located in the human growth hormone gene cluster HumGH@

D17S2266E is a new, variable genetic marker exhibiting polymorphism of the number of repeats of four- and two-nucleotide motifs. This study, carried out on a group of 250 unrelated persons from various regions of Poland, revealed the presence of 24 different alleles ranging in size from 232 to 290 base pairs. Analysis of the sequenced fragments demonstrated that the alleles consisted of two flanking regions and two variable blocks that were separated by a consensus sequence. There were (AAAG)(5)(AG)(1)(AAAG)(3-4) repeats in the first block, and [(AAAG)(2)(AG)(1)](0-1)[(AAAG)(6)(AG)(1)](0-1)(AAAG)(n) repeats in the second block. On the basis of the allele frequencies in the population, we were able to do biostatistical calculations, which gave the following results: expected heterozygosity 0.8947 +/- 0.0137, power of discrimination 0.9793, polymorphism information content 0.8837, probability of exclusion (PE) 0.7859, PE for motherless cases 0.6473, and an average paternity index of 4.7470. These biostatistical parameters show that the marker D17S2266E can find a wide range of applications in forensic testing.

The use of isoelectric focusing (IEF) as a method for the combined phenotyping of erythrocyte acid phosphatase (EAP) and esterase D (EsD)

The simultaneous isoelectric focusing (IEF) in polyacrylamide gels (PAG) of erythrocyte acid phosphatase (EAP) and esterase D (EsD) allows the poor discriminating power (DP) of EsD to be usefully combined with a highly discriminating system EAP, such that a joint DP of 0.766 was achieved compared with PGM IEF DP 0.756. Focusing was carried out in a centrally flattened gradient containing ampholines (pH 4-6 and 6-8) and the chemical spacer 3-(N-morpholino) propanesulphinic acid (MOPS). It enabled the identification of six EsD phenotypes including the recently discovered EsD5 isozymes. The application of this method to casework bloodstains is discussed.

应用PCR、聚丙烯酰胺凝胶垂直电泳及银染技术对STRD3S1359基因座进行多态性调查,首次获得中国汉族人群的基因频率分布数据。检出19个等位基因,59个基因型,基因型频率分布符合Hardy－Weinberg平衡。观察352次减数分裂未发现突变基因,DP值为0．9380,三联体PE值0．6311,二联体PE值0．4568,PIC值0．7824。证实D3S1359是一个高信息含量的STR标记系统。
