目的建立一种能够在同一反应条件下鉴定多个具体物种.又满足简单、快速、特异、灵敏、准确等实用要求的种属鉴定方法。方法从GenBank中获取人、鸡、鸭、鹅、猪、兔、鼠、绵羊、水牛、狗、山羊等11个物种的12SrRNA基因序列.设计一对针对上述11个物种的通用引物和分别针对人、鸡和鸭的特异引物,同时扩增各物种12SrRNA基因。以通用引物扩增片段为内部对照.以特异引物扩增片段用于人、鸡和鸭的种属鉴定,并分别对人、鸡、鸭单一检材以及人和鸡、人和鸭、鸡和鸭等二元混合DNA进行鉴定。结果通用引物扩增,各物种均有400bp左右的扩增片段:特异引物只对各自目标种属有扩增产物,片段大小:人163bp、鸡286bp、鸭374bp;测序结果与GenBank既有序列比对,Identities分值:人100%、鸡99%、鸭100%;通用引物扩增人、鸡和鸭检材的灵敏度为2.5Pg;特异引物扩增灵敏度人为2.5pg,鸡、鸭均为200Pg;混合DNA中任一种DNA的含量只要高于检测灵敏度即可被准确检出.不受另一种DNA量的干扰:盲测结果准确。结论本方法可以利用同一种PCR反应条件鉴定多个物种的种属。 相似文献

Since the introduction in 2001 of a urine-based detection method for recombinant erythropoietin (rHuEPO), transfusion-doping practices have regained interest. To address this problem, an efficient antidoping test designed to obtain direct proof of allogeneic blood transfusion was developed and validated. This test, based on flow cytometry analysis of red blood cell (RBCs) phenotypes, was used to determine the absence or the presence of numerous RBCs populations in a blood sample. A such, it may constitute a direct proof of an abnormal blood population resulting from homologous transfusion. Single-blind and single-site studies were carried out to validate this method as a forensic quality standard analysis and to allow objective interpretation of real cases. The analysis of 140 blood samples containing different percentages (0-5%) of a minor RBCs population were carried on by four independent analysts. Robustness, sensitivity, specificity, precision and stability were assessed. ISO-accredited controls samples were used to demonstrate that the method was robust, stable and precise. No false positive results were observed, resulting in a 100% specificity of the method. Most samples containing a 1.5% minor RBCs population were unambiguously detected, yielding a 78.1% sensitivity. These samples mimicked blood collected from an athlete 3 months after a homologous blood transfusion event where 10% of the total RBCs present in the recipient originated in the donor. The observed false negative results could be explained by differences in antigen expression between the donor and the recipient. False negatives were more numerous with smaller minor RBCs populations. The method described here fulfils the ISO-17025 accreditation and validation requirements. The controls and the methodology are solid enough to determine with certainty whether a sample contains one or more RBCs populations. This study evaluates the abilities of the Integrated Ballistics Identification System (IBIS) and BulletTRAX-3D electronic imaging systems to identify bullets fired by the same weapon in a large database of images. Ten consecutively rifled handgun barrels were test fired to obtain reference sample and known match sample pairs for upload onto both bullet acquisition systems. Both copper-jacketed and lead bullets were uploaded, to account for variations in the manner in which markings are reproduced on the different metal compositions. Ranked correlation lists were examined and evaluated. For copper-jacketed bullet correlations, both IBIS and BulletTRAX-3D identified all reference samples to their known matches within the top 10 positions. For lead bullets, BulletTRAX-3D identified all reference samples to their known match in the top 10 positions while IBIS identified only 30%. For inter composition comparisons, BulletTRAX-3D was more successful than IBIS, identifying 100% of reference samples to their known match in the top 20 for copper-jacketed to lead comparisons and 90% for lead to copper-jacketed comparisons. These results suggest that BulletTRAX-3D is more effective than IBIS in the analysis of a wider range of bullet types and it was also found to produce images of superior quality. 相似文献

Transfer restrictions have a long tradition in professional sports but came under heavy attack in recent years (e.g. Bosman
ruling, Monti system). Based on a bargaining model with stochastic player productivity, we show that less restrictive transfer
rules reallocate ex post bargaining power from players to clubs. This reallocation is efficient and in the ex ante self-interest
of players. The right to charge transfer fees enables clubs to insure their players. The players, in turn, benefit by converting
risky future income into riskless current income. Overall, player utility is higher under more than under less restrictive
transfer rules.

应用 PCR复合扩增技术、变性聚丙烯酰胺凝胶电泳和银染法显色检测技术,对云南省汉族人群 100名个体的 D9S302、 D18S865和 PLA2A基因座进行了研究,分别检出 10、 7和 7个等位基因,基因型频率分布均符合 Hardy- Weinberg平衡,家系调查证实三基因座的传递遵循孟德尔遗传规律。三基因座累积个人识别率为 0.999 7,累积非父排除率为 0.992 9。证实了复合扩增 D9S302、 D18S865和 PLA2A三基因座是法医学个人识别和亲权鉴定中极有价值的检测方法。 相似文献
