Successful STR and SNP typing of FTA Card samples with low amounts of DNA after DNA extraction using a Qiagen BioRobot EZ1 Workstation

FTA Cards (GE Healthcare) have been used for more than 4 years in Denmark for the collection of buccal cells as reference samples in crime cases. Semi-automated protocols for STR typing of DNA on punches of FTA Cards are routinely used. In average, full STR profiles were generated from approximately 95% of the FTA Cards with a standard punching protocol, while partial or no STR profile were obtained from 5% of the samples. Here, the Qiagen BioRobot^® EZ1 Workstation (Qiagen) and the EZ1 DNA Investigator Kit (Qiagen) was used to extract DNA from 29 FTA Cards from which a complete STR profile was not generated with the standard punching protocol. All 29 samples were successfully typed with the AmpF?STR^® Identifiler™ PCR Amplification Kit (Applied Biosystems) and with the SNPforID 49plex SNP assay. The lowest amount of DNA that resulted in complete STR and SNP profiles was 80 pg. The STR and SNP profiles were identical to those generated from another sample collected from each of the 29 individuals.

以苏北S村为例对农村留守妇女上班族的时间配置、社会支持网络、社会身份认同等方面进行的调查发现,农村留守妇女上班族在时间配置上,工作和家务劳动占据了主要部分,闲暇时间她们的生活较为丰富;留守妇女上班族的社会支持网中,同事和工作中认识的留守妇女为她们提供了情感支持和信息支持,在工具性支持上,她们和一般农村留守妇女没有差别;留守妇女上班族对社会身份的认同是通过经济独立、家庭地位提高等维持的;留守妇女上班族在工作获得途径、安全保障和工作保障方面面临困境。

DNA指纹图在法医学鉴定中应用的研究(Ⅰ)

应用 MYO DNA 探针对中国人进行了 DNA 遗传指纹图检验。从两个家系16人及100个无关个体中取肘静脉血提取 DNA,用限制性内切酶 HinfⅠ或 HaeⅢ水解,1%琼脂糖凝胶电泳分离,经 Southern印迹转移,MYO DNA 探针杂交,获得了清晰可辨的 DNA 指纹图谱。结果每个个体在3.0Kb 以上均能检出10条以上杂交区带,个体间的相关概率<4×10~(-9),由杂交区带构成的图谱是个体特异的,杂交区带遵循孟德尔的显性遗传方式由亲代向子代遗传;具有 DNA Fingerprints 的特点。对两起亲子鉴定的案例进行指纹图检验,孩子所存在的杂交区带,除来自母亲外,其余可在嫌疑父亲带中找到,肯定了孩子与嫌疑人的父子关系。MYO DNA 探针在亲子鉴定与个人识别的法医学鉴定中有着重要的实用价值。     

根据Y-染色体特异DNA序列鉴定牙齿性别

人类牙齿的性别鉴定,是法医学个体识别中的一个重要部分。作者们应用限制性内切酶 HaeⅢ、Sau3A 水解60颗牙髓 DNA,琼脂糖凝胶电泳分离酶解片段,在紫外灯下直接观察。根据 Y-染色体特异 DNA 序列,3.4kb 长片段的有无,判断牙齿的性别。为牙齿性别的法医学鉴定提供了一种简单、准确的新方法。     

Expert Testimony Pertaining to Battered Woman Syndrome: Its Impact on Jurors' Decisions

Participants (N = 200) were presented with a criminal homicide trial involving a battered woman who had killed her abuser. Within the trial, both the response history (passive, active) and presence of expert testimony pertaining to battered woman syndrome (present, absent) were systematically varied. As well, half of the participants in each of these conditions were provided with a nullification instruction informing them that they were free to disregard the law and acquit should a strict application of the law result in an unjust verdict. Results indicated that, compared to the passive response condition, the mock jurors were no less receptive to the expert testimony in the active response condition. The impact of the testimony on participants' verdicts, however, was moderated by the nullification instruction. That is, although the presence of the testimony did result in greater verdict leniency, this only occurred when the mock jurors had been released from a strict application of the law. The implications of these findings are discussed.     

Social Support: A Mediator between Child Maltreatment and Developmental Outcomes

The purpose of the current study was to investigate the relationship between child maltreatment, social support, and developmental outcomes in first-year college students. Participants were 202 undergraduate students (137 female, 65 male) who completed surveys at two time points: once before entering college and once during their first year of college. It was hypothesized that child maltreatment would predict poorer developmental outcomes in adolescence and early adulthood, but that social support would mediate this relationship. Results indicated that child maltreatment related negatively to developmental outcomes and to perceived social support; adolescent and young adult development related positively to perceived social support. In addition, a mediational model in which social support mediates child maltreatment and developmental outcomes was supported.
