小反刍兽疫病毒H糖蛋白基因原核表达载体的构建及表达

参照GenBank中小反刍兽疫病毒(PPRV)H抗原基因序列,人工合成了PPRV H基因,将其克隆至pUC18-T质粒中,转化E.coliJM109感受态细胞,构建并选择PPRV H基因克隆重组质粒,经核苷酸序列分析正确,将其克隆至pBAD/Thio-TOPO载体中,转化E.coliTOP10感受态细胞,核苷酸序列分析证实,成功构建了PPRV H基因重组表达载体。经不同浓度L-阿拉伯糖诱导,可稳定、高效地表达PPRV H抗原。SDS-PAGE分析结果表明,用终浓度为0.2 g/L的L-阿拉伯糖诱导5 h的表达量最高,表达蛋白为分子质量约83 ku的融合蛋白;经薄层扫描分析,其表达产量约占菌体总蛋白的10%。Western-blotting检测表明,诱导的蛋白能与PPRV H蛋白单抗发生特异性反应,说明表达的融合蛋白中含有PPRV H糖蛋白抗原。     

过氧化氢诱导PC12细胞损伤的模型的建立

目的建立不同浓度的H2O2诱导PC12细胞凋亡的模型,研究组织缺血、自由基损伤的机制。方法分别以不同浓度的H2O2处理PC12细胞12h,用噻唑蓝(MTT)比色法和流式细胞术检测处理后细胞的生长活力和凋亡情况。结果以0浓度组作为对照,100、300nmol/L的H2O2处理的PC12细胞生存率分别为(76±7.69)%、(39±7.08)%(P<0.01);0、100、300nmol/L的H2O2处理的PC12细胞后凋亡率分别为2.47%、6.00%和55.54%。结论建立了以0、100、300nmol/L的H2O2诱导PC12细胞凋亡的模型。     

Dialectics and Transformation: Exploring the International Theory of E. H. Carr

This article examines the appeal of Carr's theory of international relations, which has enjoyed a longevity matched by few other bodies of work in a discipline characterised by faddism. I attribute Carr's success as an international theorist to his subtle use of philosophy, history, and political theory. Carr's holistic and interdisciplinary approach achieves its best expression in his employment of a critical dialectics of international theory in The Twenty Years' Crisis. The article does not confine itself to this element of Carr's writings as I also examine several works, such as The Future of Nations. Independence or Interdependence?, Conditions of Peace, and Nationalism and After,that consider the transformation of the international society. The latter works also employ a dialectic of power and morality in order to envision a world very different from the international system that Carr observed, an international order that approximates the emerging relations among states at the beginning of the twenty-first century. The article concludes with an examination of the role relativism played in Carr's theory scheme and the extent to which his dialectics of utopianism and realism is an important precursor of critical and postmodern theories of international relations.     

Do Industrial Tax Abatements Spur Property Value Growth?

Despite ongoing debate regarding the effectiveness of tax abatements, Michigan's Industrial Facilities Tax (IFT) abatement program has been widely and extensively used to boost local economic development. In this article, we estimate the effects of industrial property tax abatements on industrial, residential, and commercial property value growth in the context of regional competition for a panel of 152 communities in the five counties surrounding Detroit from 1983 through 2002. We find that: (1) offering tax abatements yields statistically significant positive impacts on industrial property value growth; (2) the impacts are larger in high tax than in low tax communities; (3) there are positive spillover effects of industrial tax abatements on residential and commercial property value growth; (4) tax abatements offered in competitor communities do not appear to influence own industrial property value growth; and (5) changes in the own as well as competitor property tax rates are important determinants of industrial property value growth. However, the fiscal benefits of tax abatements are quite small as compared with the costs of offering abatements even when spillover benefits to residential and commercial properties are considered.     

T.H. Marshall and his critics: reappraising ‘social citizenship’ in the twenty-first century

T.H. Marshall's concept of ‘social citizenship’, developed in the 1949 lecture ‘Citizenship and Social Policy’, remains a vital study of welfare in developed nations. However, Marshall's social citizenship has come under attack as undermining civil liberties, or falling short of offering real equality to marginalised groups. This article returns to Marshall's lecture to show that he was in fact aware of such problems, but nonetheless held the provision of social rights to be a valuable normative project. Furthermore, this article argues that a new social citizenship, incorporating collective rights claims, could present a strong challenge to neoliberalism in contemporary welfare debates.     

Quantified Explosives Transfer on Surfaces for the Evaluation of Trace Detection Equipment

Trace explosive test surfaces are often required for the evaluation of trace detection equipment to determine the equipment performance. Test surfaces of C‐4, Detasheet, Semtex‐H, TNT, and HMTD were prepared by transferring trace amount of explosive deposited on polytetrafluoroethylene (PTFE) transfer strips onto different surfaces (Kraft paper, hard plastic, woven fabric, and soft vinyl). The amount of explosive transferred was deduced from the amount of explosive remaining on the PTFE strips after transfer, as quantified by direct analysis using tandem mass spectrometry with thermal desorption. From the data set of over 2000 transfers, we experienced lower transfer efficiency for Semtex‐H and Detasheet, and for soft vinyl and hard plastic. However, the rapid quantification mass spectrometric method allowed the transfer efficiency to be determined for all test surfaces used in an evaluation of trace explosive detectors, thereby permitting only the test surfaces with desired transfer to be accepted for the assessment.     

中国朝鲜族H19基因上游差异甲基化区SNP分布

目的调查中国朝鲜族群体中H19基因上游差异甲基化区(differentially methylated region,DMR)SNP及单倍型分布,为法医学应用及群体遗传学研究提供基础数据。方法收集中国朝鲜族101份无关个体血样和14份来自5个亲缘关系已知的两代家系血样,用PCR-循环测序、McrBC消化DNA后PCR方法检测H19基因上游DMR的SNP,并用亲源印记等位基因(parentally imprinted allele,PIA)分型法进行单倍型检测,再计算相关遗传学参数。结果在H19基因上游DMR 1174bp的目的基因扩增产物中,共检出13个SNP(rs10840167、rs2525883、rs12417375、rs4930101、rs2525882、rs2735970、rs2735971、rs11042170、rs2735972、rs10732516、rs2071094、rs2107425、rs4930098)和5种单倍型,有9个SNP属于高鉴别能力的遗传标记,其单倍型具有较高的个人识别率,单倍型平均基因多样性(GD)为0.714。应用McrBC酶消化基因组DNA的PIA分型法确定了家系子代样本的母源单倍型。结论 H19基因上游DMR在中国朝鲜族群体中具有很高的遗传多态性,母源单倍型的确定进一步提高了印记基因的法医学鉴定效能。     

展呈H5N1亚型禽流感病毒M2e表位鞭毛蛋白的构建及其免疫原性

以重组质粒pET-fliC和pUC57-M2e2为模板,通过重叠PCR将M2e2基因插入fliC的高变区域,构建嵌合鞭毛基因片段fliC/M2e2。将fliC/M2e2片段插入原核表达载体pET30a+,获得重组质粒pET-fliC/M2e2,将其转化鼠伤寒沙门氏菌LB5000,获得重组菌LB5000(fliC/M2e2)。应用P22噬菌体转导技术,将fliC/M2e2基因导入都柏林沙门氏菌SL5928的基因组中,经生化血清学鉴定、动力学测定、透射电镜观察、Western-blot分析,将阳性转导菌命名为SL5928(fliC/M2e2)。提取重组菌鞭毛蛋白fliC/M2e2作用HEK293-mTLR5细胞,能刺激细胞分泌IL-8。将fliC/M2e2通过皮下注射途径免疫C3H/HeJ小鼠,50μg/只。结果显示,二免后第14天,fliC/M2e2鞭毛蛋白免疫组的血清抗体效价与对照组之间呈现显著性差异(P<0.05)。ELISPOT结果表明,在脾细胞中,fliC/M2e2鞭毛蛋白免疫组检测到较高水平的IFN-γ和IL-4分泌细胞,且免疫应答以Th1型为主。结果表明,构建的fliC/M2e2嵌合基因能够在沙门氏菌中表达,重组嵌合鞭毛蛋白fliC/M2e2能激发机体针对M2e表位的体液免疫应答和Th1型细胞免疫应答。     

His master’s voice: H.L.A. Hart and Lacanian discourse theory

H.L.A. Hart’s jurisprudence seems antithetical to Jacques Lacan’s psychoanalysis. Professor Schroeder argues that, in fact, Hart’s concept of law has surprising similarities to Lacan’s ‘discourse of the Master’. Both reject a command theory of law: subjects do not obey law out of fear. Moreover, both insist that the authority of law is completely independent from its content. Anyone seeking to develop a psychoanalytically sophisticated critical legal theory should reconsider Hart. As insightful as his concept of the symbolic is, Lacan has no expertise in legal systems and does not discuss positive law per se. Although he posited a theory of ethics in his Seventh Seminar and the seeds of a jurisprudence are implicit within his theory, he offers no account of legal right, justice or what Hart misleadingly calls ‘morality’. A Lacanian jurisprudence must, therefore, be supplemented by other sources. Moreover, legal positivists should not dismiss psychoanalysis. As insightful as Hart’s jurisprudence is, his theories of legal subjectivity and linguistics are simplistic and his concept of law too narrow. He describes only one aspect of legal experience: obedience to law. He ignores what most legal actors do: Hart’s concept of law excludes the practice of law. Although Lacan’s ‘master’s discourse’ surprisingly parallels Hart’s jurisprudence, Lacan does not restrict the symbolic to the master’s discourse. It requires three other ‘discourses’. Lacan, therefore, supplements Hart. Specifically, Lacan’s fourth discourse describes the excluded practice of law and provides the mechanism by which ‘morality’ can critique law.

重组鸡白细胞介素6对不同种类疫苗的免疫增强作用

为评价重组鸡白细胞介素6(rChIL-6)在商品鸡体内对不同种类疫苗的免疫增强作用,采用不同接种方式,应用rChIL-6分别对不同种类新城疫病毒(NDV)单联疫苗和新城疫病毒-传染性支气管炎病毒-禽流感病毒H9亚型三联疫苗(NDV-IBV-H9)进行了免疫增强试验以及传染性法氏囊病病毒(IBDV)单联油乳剂灭活疫苗的免疫增强田间试验,并进行了将rChIL-6与NDV Ⅳ系预混合乳化制备成的灭活疫苗的免疫试验.结果表明,将rChIL-6与NDVⅣ系、Ⅰ系活疫苗预混合后分别采用滴鼻点眼、肌肉注射接种方式的免疫增强效果优于其他方式;rChIL-6与NDV抗原预混后制备成油乳剂灭活疫苗的免疫增强效果优于将相同抗原量的NDV油乳剂灭活疫苗和rChIL-6同时分点免疫接种.rChIL-6对NDV油乳剂灭活疫苗的免疫增强效果优于活疫苗,对NDV单联灭活疫苗的免疫增强效果优于NDV-IBV-H9三联灭活疫苗.田间试验表明,rChIL-6对IBDV油乳剂灭活疫苗首免和二免都具有显著的免疫增强作用,二免的免疫增强效果优于首免.这些研究结果为进一步开展新型鸡基因工程免疫增强剂研究奠定了基础.
