目的应用mt DNA中COI(348bp)、COII(637bp)及16Sr DNA(513bp)基因序列鉴定贵阳地区常见尸食性蝇类的种属。方法收集贵阳地区常见尸食性蝇类标本,经形态学分类鉴定后提取胸肌DNA,扩增COI(348bp)、COII(637bp)及16Sr DNA(513bp)基因片段,测序后用DNAMAN 4.0序列分析软件进行序列拼接,NCBI中的BLAST、MEGA 5.2软件包对所得序列分析建立种内及种间进化分歧率,并构建系统发育树。结果 COI种间平均进化分歧率为12.55%,种间范围在3.0%~18.6%,种内范围在0%~0.7%;COII种间平均进化分歧率为13.73%,种间范围在5.3%~21.6%,种内范围在0%~0.6%;16Sr DNA种间平均进化分歧率为4.54%,种间范围在0.4%~7.6%,种内范围在0%~0.4%。结论 COI、COII及16Sr DNA可用于尸食性蝇类的种属鉴定,该方法可作为传统分类方法的补充手段。  相似文献

从临床病猪无菌采集抗凝血 ,抽提全血基因组DNA ,用能够扩增大多数真细菌 16SrRNA基因的通用引物 ,扩增出长度为 15 0 2bp的嗜麦芽窄食单胞菌的 16SrRNA基因 ;该基因与国外报道的嗜麦芽窄食单胞菌部分临床和环境分离株核苷酸序列的同源性达 99%以上。证实嗜麦芽窄食单胞菌可感染猪。  相似文献

POPULATIONS: This study reports the genetic polymorphism observed at 15 short tandem repeat loci D3S1358, TH01, D21S11, D18S51, D5S818, D13S317, D7S820, D16S539, CSF1PO, vWA, D8S1179, TPOX, D2S1338, D19S433, and FGA in four aboriginal populations of Bengal. The analysis was performed to decipher the suitability of CODIS as well as six other highly polymorphic and unlinked markers in Forensic Testing. A 38-year-old diabetic woman developed hyponatraemia and fatal non-ketotic coma after elective cholecystectomy. At the autopsy, it was revealed that the immediate cause of death was cerebral oedema with secondary pontine haemorrhage. The cerebral oedema was associated with severe hyponatraemia and atrophy of the endocrine organs, including the adrenal glands. Biochemical analysis of serum taken immediately before death indicated that the primary defect was pituitary insufficiency, a recognised but rare complication of diabetes.  相似文献

Abstract: The quality and efficiency of a standard organic DNA isolation method and a silica‐based method using the QIAGEN Blood Maxi Kit were compared to obtain human DNA and short tandem repeats (STRs) profiles from 39 exhumed bone samples for paternity testing. DNA samples were quantified by real‐time PCR, and STR profiles were obtained using the AmpFlSTR® Identifiler® PCR amplification kit. Overall, the silica‐based method recovered less DNA ranging from 0 to 147.7 ng/g (average 7.57 ng/g, median = 1.3 ng/g) than did the organic method ranging from 0 to 605 ng/g (average 44.27 ng/g, median = 5.8 ng/g). Complete profiles (16/16 loci tested) were obtained from 37/39 samples (95%) using the organic method and from 9/39 samples (23%) with the silica‐based method. Compared with a standard organic DNA isolation method, our results indicate that the published silica‐based method does not improve neither the quality nor the quantity of DNA for STR profiling.  相似文献

目的利用Maxwell 16裂解纯化法从保存8年以上陈旧精斑检材中获取精子DNA。方法 8份陈旧精斑检材采用Maxwell 16裂解纯化法提取精子DNA,并采用Powerplex○R21试剂盒进行复合扩增,产物用AB3130型遗传分析仪检测,结果与常规方法进行对比。结果成功获得8份陈旧精斑检材精子STR分型。结论差异裂解配合Maxwell 16裂解在陈旧精斑检材精子DNA检验中效果明显。  相似文献

The allocation algorithm of the Linux FAT32 file system driver positions files on disk in such a way that their relative positions reveal information on the order in which these files have been created. This provides an opportunity to enrich information from (carved) file fragments with time information, even when such file fragments lack the file system metadata in which time-related information is usually to be found.Through source code analysis and experiments the behaviour of the Linux FAT allocator is examined. How an understanding of this allocator can be applied in practice is demonstrated with a case study involving a TomTom GPS car navigation device. In this case, time information played a crucial role. Large amounts of location records could be carved from this device's flash storage, yielding insight into the locations the device has visited—yet the carved records themselves offered no information on when the device had been at the locations. Still, bounds on the records' time of creation could be inferred when making use of filesystem timestamps related to neighbouring on-disk positions.Finally, we perform experiments which contrast the Linux behaviour with that of Windows 7. We show that the latter differs subtly, breaking the strong relation between creation order and position.  相似文献
