禽流感病毒A/Turkey/Canada/63毒株HA和NA基因的克隆及序列分析

用SPF鸡胚增殖禽流感病毒(AIV)A/Turkey/Canada/63毒株,提取病毒基因组总RNA,应用RT-PCR技术分别扩增该病毒分离株的HA和NA基因全片段,并克隆到pMD18-T载体上。测序结果表明,该毒株的HA基因全长为1745bp,含有完整的阅读框架,编码566个氨基酸；NA基因全长1467bp,编码469个氨基酸。BLAST序列比较结果显示,该毒株HA基因属于H6亚型,NA基因属于N2亚型。将获得的HA和NA基因与AIV数据库中H6和N2基因进行遗传演化分析,表明该毒株HA基因与其他H6基因核苷酸的同源性为80.5%～87.3%,氨基酸的同源性为88.0%～93.1%；NA基因与其他N2基因核苷酸的同源性为86.0%～93.5%,氨基酸的同源性为90.6%～96.3%。     

个人言语风格的司法鉴定

言语是运用语言表达思想的结果 ,书写的文字 (笔迹 )是它借以存在的物质基础 ,笔迹鉴定和言语风格鉴定成为统一于文书鉴定中的两个有机组成部分。个人之间不同的言语风格、语言特征 ,主要表现在词汇、句式、句法等语言材料和表达方式的各个要素 ,以及言语内容与个人属性和案件情况的关系等诸方面。对案件中涉及的书面言语材料个人言语风格的鉴定 ,必须将言语、笔迹与具体的案件情况和其它物证结合起来进行综合判断。     

加入WTO后我国行政审批制度的改革

我国社会主义市场经济调控体系框架经过20多年的改革开放已初步建立,政府对经济的直接管理逐步转向间接调控,越来越多地注意运用经济手段、经济政策、经济法规调控经济运行。但是,我们也应看到,我国的行政守批制度还存在着项目多、程序繁、责权划分不清及腐败现象,这已经成为我国加入WTO后面临的一个最大的障碍。因此,必须通过把行政审批与其他方面改革相结合、清理规范行政审批项目、对不同的行政审批项目采取不同的对策及积极转换行政审批部门的观念等手段进行改革。     

赤羽病病毒核衣壳蛋白基因的表达纯化及活性鉴定

为获得赤羽病病毒(Akabane virus,AKAV)核衣壳蛋白重组抗原,经RT-PCR扩增了OBE-1株的S节段,将其插入到pMD 18-T载体,然后用带有酶切位点的引物从该重组载体亚克隆出表达核衣壳蛋白的N基因,用BamHⅠ和XhoⅠ双酶切后与同样处理的表达载体pET-28a( )进行连接,转化感受态细胞BL21(DE3)。将筛选出的阳性克隆进行测序、IPTG诱导表达和融合蛋白纯化及活性鉴定。结果表明,N基因含有1个全长702 bp的ORF,编码233个氨基酸,通过SDS-PAGE和Western-blotting分析,证实重组菌株可高效表达有免疫活性的分子质量约27 ku的可溶性融合蛋白,用薄层扫描仪分析,其最高表达量占可溶性菌体蛋白总量的58.5%。工程菌经超声波裂解高速离心后,上清液在ProBondTMPurification System(含Ni2 )天然状态下纯化,可获得高纯度的融合蛋白。     

Availability of target odor compounds from seized ecstasy tablets for canine detection

The aim of this study was to compare seized samples of 3,4-methylenedioxy-N-methylamphetamine (MDMA) pills, used to train law enforcement detection canine teams, to determine what differences exist in the chemical makeup and headspace odor and their effect on detectability. MDMA solutions were analyzed by liquid chromatography-mass spectrometry. Analysis of these samples showed a wide variance of MDMA (8-25%). Headspace SPME-GC/MS analysis showed that several compounds such as 3,4-methylenedioxyphenylacetone and 1-(3,4-methylenedioxyphenyl)-2-propanol are common among these MDMA samples regardless of starting compound and synthesis procedure. However, differences, such as the level of the various methylenedioxy starting compounds, were shown to affect the overall outcome of canine detection, indicating the need for more than one MDMA training aid. Combinations of compounds such as the primary odor piperonal in conjunction with a secondary compound such as MDP-2-OH or isosafrole are recommended to maximize detection of different illicit MDMA samples.     

Y-chromosome and autosomal STR diversity in four proximate settlements in Central Anatolia

Due to the longstanding human presence in the region and the influence of social traditions, the genetic make-up of populations currently inhabiting Turkey (Anatolia) is quite complex. To characterize the patterns of genetic diversity in rural Central Anatolian villages, we analyzed samples collected at four local settlements for variation at 17 Y-chromosome STR and 15 autosomal STR loci. The resulting data reveal considerable diversity within these settlements, as well as some structure in the paternal genetic variation, with a limited number of haplotypes being shared between the communities. These findings have important implications for forensic studies of Turkish populations.     

视觉刺激事件相关电位及其研究进展

事件相关电位(ERP)是人对客体进行认知加工(注意、记忆、思维等)时,在头颅表面所记录到的大脑电位变化。ERP由多种与认知功能相关的成分构成,包括早期成分,失匹配负波(MMN),N2家族,P3家族,语言加工相关的ERP成分等。通过对ERP这些成分的分析,可以深入探究人类大脑的认知功能,并且具有重要的法医学应用价值。     

H5N1亚型禽流感病毒血凝素单克隆抗体的制备及其应用

采用活病毒免疫,常规杂交瘤融合的方法,获得了4株针对H5N1亚型禽流感病毒(AIV)A/duck/Shandong/093/2004(A/D/SD/04)株血凝素(HA)的单克隆抗体A8E8、D2F8、A8C8和C3G4。其中A8E8可以与11株不同来源的H5N1AIV发生交叉免疫抑制性反应(HI),具有广谱性。采用间接免疫荧光试验(IFA),用A8E8单克隆抗体测定A/D/SD/04株对COS1和MDCK2种细胞的半数感染量(TCID50),结果显示,A/D/SD/04株在COS1上的TCID50为10-5.33/0.1mL,在MDCK上的为10-7.33/0.1mL。将编码A/D/SD/04株的HA基因质粒与PR8质粒共转染COS1细胞后,用A8E8进行IFA,能观察到特异性绿色荧光,这与鸡胚接种结果相符。表明,用此单克隆抗体能准确地检测到H5重组病毒。

Judges and CEOs: French Aspects of Corporate Governance

The debate about corporate governance has brought to the front stage the notion of social interest or interest of the company itself as distinct of its stakeholders. French Law, judges, and CEOs use this notion with different meanings and intentions. The judges refer to it mainly to guarantee the continuity of the firm, especially when the latter faces economic difficulties; the CEOs refer to it to keep a free hand in managing the company. Shareholders see the notion as ambiguous and mostly used against their own interest. Shareholders and company interest do not merge. They differ on the industrial relations policy they induce and in the management freedom bestowed on CEOs. A reform of French Company Law currently under discussion seeks conciliation while trying to comply with the principles of corporate governance best practices.
