Comparison of Commercial Kits for Recovery and Analysis of Bacterial DNA From Fingerprints

In forensic science, fingerprints are a common source of evidentiary information. However, latent examination is not always successful and trace human DNA cannot always be obtained. Thus, examining the fingerprint microbiome may offer a suitable alternative to more traditional methods of forensic identification. The Zymo Research ZR Bacterial/Fungal DNA MicroPrep™ Kit, Qiagen QIAmp^® DNA Mini Kit, Promega Wizard^® Genomic DNA Purification Kit, and the MPBio FastDNA^® Spin Kit were compared for their ability to yield a sufficient amount of bacterial DNA for next-generation sequencing in order to obtain a microbiome profile. Prints were deposited onto slides, allowed to sit for up to 1 month, and total DNA isolated and quantified using each kit. The kit from Zymo Research yielded the most concentrated DNA sample (0.0084 ng/µL) in the least amount of time as compared to other kits examined. Although this amount of DNA was far below the recommended DNA concentration threshold recommended for next-generation sequencing, a microbiome profile was successfully obtained. As interest in using the microbiome of an individual as a forensic tool continues to increase, there is the possibility that the microbiome of a fingerprint could complement traditional human DNA profiling in the future. This study provides evidence that trace amounts of bacterial DNA from fingerprints is quantifiable and sufficient for microbiome analysis.     

Twice-hyphenated: transnational identity among second-generation Ethiopian-American professionals in the Washington,DC, metropolitan area

This article examines the dynamics of transnational identity among second-generation Ethiopian-American professionals. Drawing on the experiences of 21 second-generation Ethiopian-American professionals, I analyze how they manage the dual challenges of maintaining Ethiopian identities while embracing American values and aspirations. Study participants indicated how their parents actively encourage them to embrace Ethiopian culture while they expect them to succeed in the US society. They expressed fear that if they become ‘too American’, they will disappoint their parents. Moreover, they find restricting their identity to one group or another too confining. They selectively choose defining ethnic characteristics from the cultural domains in which they operate –their families, social networks, and school environments; media images; popular culture; and the broader dominant culture. Most of them embrace individualism and autonomy in order to negotiate, create, and recreate their own transnational identities. The ethnic characteristics of second-generation Ethiopian-American can certainly be characterized as ‘hybrid identities’.

《红楼梦》与《源氏物语》三类线性时间叙写

线性时间叙事是中日古典文学叙事基本特征,文本撰作的线性时间可分为朝代纪年、世代时间、人物年纪,三者指向明确且不可回溯。《源氏物语》原文古典当中并没有明确标明这三类时间标识,《红楼梦》更是有意识地模糊、弱化、消除这三类时间标识,开篇采用倒叙,篇中多次打乱时间流变次序。但,三类时间标识的的确确涵括于两书文本,不加细辨,难以识别。本论通过文本细读与反复举证,明确两文本当中三类时间标识,勘校过往两书研究当中的相关舛误,并将这三类时间标识的叙写技巧进行逐类比较。     

Mitochondrial genome sequencing with short overlapping amplicons on MiSeq FGx system

With the development and maturation of massively parallel sequencing (MPS) technology, the mitochondrial genome (mitogenome) sequencing is increasingly applied in the forensic field. In this study, we employed the strategy of short overlapping amplicons for the whole mitogenome, library preparation with tagmentation using the Nextera® XT DNA Library Preparation Kit, sequencing on the MiSeq FGx^TM Forensic Genomics System and analyzing data using the mitochondrial(mtDNA) MSR Plug-in and the mtDNA Variant Analyzer. A total of 27 libraries and 56 libraries were sequenced in a run using MiSeq Reagent Kit v2 and v3, respectively. Results showed more than 1800 × of averaged depth of coverage (DoC) at each position. Concordant haplotypes of 9947 A and 2800 M were obtained at 32 variants. Cross-reactivity was observed with 1 ng primate DNA and 10 ng non-primate DNA but could be easily distinguished. Full and accurate variants were obtained from at least 50 pg input DNA and from minor contributors between 19:1 and 1:19 mixed ratios with known reference profiles. More than 86% variants were detected from ≥200-bp degraded samples but its haplotype was assigned to more ancestral haplogroup. Further, a total of 3 962 variants were observed at 613 nucleotide positions from 103 Xibe mitogenomes with 25:1 ratio of transitions to transversions. Two new transversions (C13735A and A14755C) and two tri-alleles at nps 9824 and 16092 were identified. There were 103 unique mitogenome haplotypes from 103 Chinese Xibe that were assigned to 79 haplogroups. Haplogroup D was the preponderant top-level haplogroup in Xibe followed by F, B, M, A, N, G, C, Z, Y, HV and J. Random match probability (RMP) and haplotype diversity (HD) of the whole mitogenome was calculated as 0.0097 and 1.0000, respectively. Compared with HVS-I only, RMP decreased 33.56%, while the number of haplotypes and HD increased 15.73% and 0.49%, respectively. Principal component analysis (PCA) showed that Xibe was clustered to East and Southeast Asian. As a whole, this MPS strategy is suitable for the whole mitogenome sequencing especially for degraded samples and can facilitate generating mitogenome data to support the routine application in forensic sciences. {"type":"entrez-protein","attrs":{"text":"EMP00726","term_id":"464458599"}}EMP00726 is the first whole mitogenome dataset from Xibe contributed to the EMPOP.Supplemental data for this article are available online at.
