Enzymatic detection of γ-hydroxybutyrate using aldo-keto reductase 7A2

Gamma-hydroxybutyrate (GHB) is a prescribed medication as well as a drug of abuse. Its detection in various matrices for in-field forensic scientists remains a challenge. We have developed an assay that uses aldo-keto reductase 7A2 (AKR7A2) for the specific determination of GHB in various drinks. AKR7A2 was purified using Ni-affinity chromatography. The Michaelis-Menten constant for the GHB oxidation reaction was 10 mM, and the minimum detection limit was 4 mM. Ethanol was not a substrate for AKR7A2. In a coupled reaction with NADP(+), phenazine methosulfate (PMS), and 2,6-dichlorophenolindophenol, various beverages (orange juice, milk, soda, and numerous alcoholic drinks) containing GHB turned from blue to light yellow. In a second coupled reaction where diaphorase replaced PMS, the presence of GHB also caused the expected change of color in various beers.     

尿素氮、肌酐和尿酸死后生化学检测的法医学应用

死后生化学是法医鉴定中重要的检查手段。研究表明含氮化合物中尿素氮、肌酐、尿酸是临床上常用的生化学检测指标,其在法医死后生化学检测中也有重要的价值。本文就体内上述三者的特征及其在法医学中的研究及应用加以综述,以期为法医死后生化学分析提供参考。     

水果中亚硝酸盐含量的测定及分析

采用盐酸萘乙二胺法对市场所售的苹果、香蕉、甜梨、橙子、桔子等15种水果中亚硝酸盐的含量进行分析测定。结果表明,15种水果中均含有亚硝酸盐,其中亚硝酸盐含量最低的是柚子,最高的是西瓜。由于国家尚未出台水果中亚硝酸盐残留量的标准,参考中华人民共和国国家标准GB/T18406.1-2001对蔬菜中亚硝酸盐含量做出的限量要求:NaNO2≤4.0(mg/kg),可知,15种水果中,西瓜、特小凤西瓜、甜梨、烟台梨亚硝酸盐超标,其它水果亚硝酸盐含量较低。15种水果中亚硝酸盐含量的特点是,南方水果中亚硝酸盐含量普遍低于北方水果,尤其是柑橘类水果亚硝酸盐含量都很低。     

Capillary electrophoretic analysis of phosphorus species in clandestine methamphetamine laboratory samples

The clandestine manufacture of methamphetamine is a spreading epidemic. Manufacturing methods are constantly changing, necessitating the implementation of new analytical tools to identify materials from these labs. Characterization of phosphate, phosphite, and hypophosphite ions is necessary to distinguish the various phosphorus-iodine manufacturing methods that are popular methods for reducing pseudoephedrine to methamphetamine. This work describes a capillary electrophoresis method to separate acetate, azide, bromide, carbonate, chlorate, chloride, chromate, fluoride, hypophosphite, iodide, nitrate, nitrite, perchlorate, phosphate, phosphite, sulfate, sulfite, and thiocyanate. The CElixerOA 8.2 dynamic coating system was modified by lowering the capillary temperature to 15 degrees C and using an acid flush between runs to remove adsorbed materials. This allows detection of ions down to between 10 and 30 parts per million with percent relative standard deviations of normalized migration times under 0.1%. This method is a valuable tool for the characterization of phosphate, phosphite, and hypophosphite in routine analysis of clandestine methamphetamine manufacturing evidence and has a broader application in other areas of forensic analysis.     

A new staining method for constriction marks in skin

A modified Poley's acid fuchsin-methyl green stain was used to demonstrate ligature marks in tissues taken at autopsy. This stain was compared in 30 cases with haematoxylin-eosin, Mallory's trichrome and Ogata's picro-indigocarmine stains. Using the modified Poley method, it was possible to demonstrate the compression mark in corpses even when post-mortem changes were advanced. The method was found useful in evaluating the cases when the force used was minimal or when the mark was atypical using the usual haematoxylin-eosin method.     

近期刑法热点问题的思考

“银广夏做假事件”、“裁判黑哨事件”以及“硫酸伤熊事件”所折射出的问题,也是我国目前法制建设极为关注的问题。本文通过法理分析,指出“银广夏做假事件”的实质在于有法不依,以至使证券市场遭受了不应有的损害;而“裁判黑哨事件”的关键在于我国法律尚有不完善之处;而“硫酸伤熊事件”就其行为性质而言,可以认定为毁坏公私财物的行为,但能够唤起的整个社会的人文关怀远远超出了对个案的如何处理。     

不同加工工艺对亳菊中绿原酸、木犀草苷和3,5-O-双咖啡酰基奎宁酸含量的影响

目的 比较不同加工方法对亳菊中绿原酸、木犀草苷和3,5-O-双咖啡酰基奎宁酸含量的影响。方法 样品用50%甲醇溶液提取,采用高效液相色谱法在同一色谱条件（色谱柱为Shimadzu VP-ODS,250 mm×4.6 mm,5 μm;流动相：乙腈-0.1%磷酸溶液;梯度洗脱;流速：1.0 mL/min;柱温：30 ℃）下对亳菊3种成分同时进行测定并比较不同干燥条件下各成分的含量。结果 不同加工方法对亳菊中3种有效成分含量有明显影响,对亳菊花的色泽、气味、质地有明显影响,胎菊和菊米无明显变化。结论 微波和蒸制干燥可作为亳菊规模化生产的加工方法。     

肝片形吸虫及其培养液内花生四烯酸代谢物水平和作用探讨

用体外培养的方法分析了肝片形吸虫及其培养液内花生四烯酸代谢物水平.结果表明,肝片形吸虫虫体匀浆中的PGE2、TBX2和6-keto-PGF1α水平分别为0.93pg/g±0.12 pg/g,0.26pg/g±0.02pg/g和0.43 pg/g±0.09 pg/g;培养液中分别为52.50 ng/L±2.14 ng/L,32.17ng/L±7.18 ng/L和124.81 ng/L±44.33 ng/L;均以PGE2和6-keto-PGF1α为高.提示肝片形吸虫成虫有PGE2、TBX2和6-keto-PGF1α等花生四烯酸代谢物存在,这类代谢物可能对宿主机体有作用.
