目的建立了生物样品中丁丙诺啡的高效液相色谱-电喷雾串联质谱检测方法。方法样品经固相萃取提取净化、反相液相色谱分离后进行质谱检测,根据保留时间及特征离子进行定性分析,以母离子m/z468进行定量分析。结果在10-500ng/ml(ng/g)范围内峰面积与质量浓度的线性关系良好(r^2〉0.993)。在50、100、500ng/ml(ng/g)3个添加水平,尿、血、肝中丁丙诺啡的平均回收率为74%~94%,日内测定结果的相对标准偏差小于8%,日间测定结果的相对标准偏差小于10%。结论该方法简单、灵敏,特异性强,适用于生物样品中丁丙诺啡的分析检验。  相似文献

目的基于变性高效液相色谱技术,建立一种不需测序和杂交的新的mtDNA控制区多态性分析系统。方法mtDNA控制区序列(包括HVⅠ,HVⅡ和HVⅢ)被分为4个扩增片段,采用配对分析突变检测模式进行DHPLC分析。对DHPLC检测条件(包括柱温和洗脱梯度等)进行优化。对100个不同类型差异序列的组合配对以检验该方法的检测效力。结果10组序列相同的样本配对DHPLC图谱均只显示1个样品峰。对序列相差1个碱基~7个碱基、插入(/缺失)1个碱基、插入(/缺失)2个碱基等类型的90个扩增片段组合,用DHPLC进行分析,均得到≥2个样本峰的DHPLC图谱,序列差异检出率达100%。该技术可检测的异质性DNA成分的最小百分含量为10%。结论DHPLC-mtDNA控制区多态性分析系统快速、经济和实用,在检测mtDNA异质性方面较直接测序更灵敏。  相似文献

Abstract

This paper provides an analysis of data on school security measures which were obtained from a survey administered to a sample of 230 high school students. The majority of students indicated that the school police officers and security officers help keep the schools safe and that the drug-sniffing dogs help reduce drugs in the schools, but there was no clear consensus among the students on the issues of whether the video surveillance cameras increase safety, whether the police and security officers should search students with metal detectors, or whether there should be more police and security officers in the schools. The only security measure which the majority of students disliked was the policy that all backpacks be translucent. An examination of gender differences in student perceptions of school security measures shows that males were significantly more likely than females to negatively evaluate the school police officers and to oppose the use of metal detectors in the schools. Finally, the data indicate that the aforementioned security strategies have little impact on the presence of drugs and weapons in the schools. The policy implications are discussed.  相似文献

Abstract

From the late seventeenth century on the idea of culture underwent a gradual transformation. Originally this concept referred essentially to the “refined” way of life of the ruling social elite (which certainly included among others also such activities as listening and making music, reading works of literature, commissioning works of fine art). Popular culture, on the other hand, refers to the usually collective practices of groups of rural and urban workers taking the form of performance. They were not only excluded from refined culture, but it was regarded as completely unsuitable for them, potentially creating dangerous social aspirations. It is with the great social transformation from feudal to bourgeois society that the idea of refined culture was replaced by that of “high culture” encompassing both the arts and the sciences: works claiming universal human significance. This “high culture” for a considerable time coexisted with the remnants of popular culture. It has been only due to the great technical advances that its true opposite, “mass culture” emerged, at the turn to twentieth century, claiming an empirical universality: being understandable and truly interesting to everyone. In economic respect, there is a competitive relation between high and mass culture. However, it is argued that there can be no cultural competition between them. For they posit differing and potentially co-existing receptive attitudes. The characterisation of this difference and the discussion of the seeming exceptions to the so-articulated conceptual scheme occupies the concluding part of this essay.  相似文献

Abstract: GeneMarker® HID was assessed as a software tool for the analysis of forensic short tandem repeat (STR) data and as a resource for analysis of custom STR multiplexes. The software is easy to learn and use, and includes design features that have the potential to reduce user fatigue. To illustrate reliability and accuracy, STR data from both single‐source and mixture profiles were analyzed and compared to profiles interpreted with another software package. A total of 1898 STR profiles representing 28,470 loci and more than 42,000 alleles were analyzed with 100% concordance. GeneMarker HID was also used to successfully analyze data generated from a custom STR multiplex, with simplified and rapid implementation. Finally, the impact of the user‐friendly design features of the software was assessed through a time scale study. The results suggest that laboratories can reduce the time required for data analysis by at least 25% when using GeneMarker HID.  相似文献

In this paper, we use bloodstains to illustrate an approach for identifying latent evidence on dark cloth using near infrared (NIR) photography combined with high dynamic range (HDR) photography techniques. NIR photography alone has been used to capture latent evidence that cannot be seen in normal ambient light. HDR techniques combine multiple bracketed photographs of the same image to increase the dynamic range of the photograph which can provide greater contrast. Using NIR photography alone, we were able to detect a bloodstain up to a 1/16 dilution, an improvement over previous studies. Combining NIR photography with the HDR process resulted in a noticeable increase in visibility up to 1/16 dilution when compared to NIR photographs alone. At 1/32 dilution, we were able to detect bloodstains that were not visible using NIR alone. NIR is a useful tool for imaging latent evidence, and combining NIR with HDR consistently provides better results over NIR alone.  相似文献

Lubricating oil comparisons impact a variety of forensic investigations, including cases where oil was transferred from a suspect vehicle to the crime scene or victim. In this study, high-temperature gas chromatography/mass spectrometry was used to examine the influence of oil mixtures and oil changes over time on the comparison of known and questioned lubricating oils. Varying concentrations of oil mixtures were prepared and showed the potential for identifying individual components. Motor oils from 18 automobiles monitored over a 2-month period did not demonstrate significant changes in the chromatographic data. Chemometric analysis of motor oil mass spectral data provided little information regarding differentiation of, or changes in, the samples. Power steering fluid (PSF) from a naturally occurring leak collected from several locations was consistent with the PSF in the automobile's reservoir, and the PSF composition did not change over time.  相似文献
