Bacterial Degradation of Risperidone and Paliperidone in Decomposing Blood

The stability of two benzisoxazole antipsychotics was determined in vitro in decomposing porcine blood inoculated with bacteria, utilizing a high‐performance liquid chromatography with ultraviolet and fluorescence detection method for drug quantitation. Stability experiments for risperidone and paliperidone were conducted at 7, 20 and 37°C for 4 days using sterile and bacterially inoculated porcine blood. The drugs were stable in sterile blood at each temperature and in inoculated blood at 7°C, but degraded significantly in inoculated blood at 20 and 37°C. Complete loss occurred within 2 days when incubated at 37°C. The benzisoxazole‐cleaved degradation products for both drugs were identified as 2‐hydroxybenzoyl‐risperidone and 2‐hydroxybenzoyl‐paliperidone utilizing liquid chromatography quadrupole‐time‐of‐flight mass spectrometry and accurate mass measurements. The degradation products have been found in postmortem case studies, including one case where risperidone and paliperidone were not detected, indicating complete conversion can occur in situ.     

The normalizing role of rationalist assumptions in the institutional embedding of neoliberalism

The political economy of Britain over the past three decades provides an interesting example of the consolidation, normalization and institutionalization of a new economic paradigm – neoliberalism. As such, it serves as a potentially instructive focus for debate both about the conditions under which economic paradigms are replaced and consolidated and the evolution of such paradigms through the process of institutionalization. In this paper I suggest that the institutionalization of this new economic paradigm has been associated with the shift from a normative to a normalized and necessitarian neoliberalism. I examine the role played by rationalist assumptions in this extended process of normalization-institutionalization. After presenting a stylized account of the evolution of British neoliberalism, I show how New Labour's monetary policy regime is the heir to the legacy of monetarism and its agenda of labour-market reform is the heir to Thatcherism's supply-side economics. I suggest that the time-inconsistency thesis and the business school globalization thesis have played an equivalent role, for New Labour, to that played for the new right by monetarism and supply-side economics in legitimating neoliberalism. In this way neoliberalism has been normalized. In the final sections of the paper I reflect on the implications of the normalized and necessitarian character of neoliberalism in Britain for its contestability and for democratic economic governance more broadly.     

柔嫩艾美球虫pEtK2抗原基因的克隆表达及其表达产物的抗原性分析

应用RT-PCR技术从纯化的柔嫩艾美球虫孢子化卵囊子孢子中克隆了pEtK2抗原基因,并进行了原核表达和抗原性分析,同时用MTT法检测了重组蛋白对球虫耐过鸡淋巴细胞的刺激效果。结果显示,克隆的pEtK2基因全长1464 bp,具有一个完整的开放阅读框,编码487个氨基酸,具有多个T细胞表位。重组蛋白诱导5 h后表达量最多,分子质量约55 ku,占菌体总蛋白的32%。纯化的重组蛋白能够刺激球虫耐过鸡T淋巴细胞增殖,可作为球虫基因工程疫苗或DNA疫苗的候选基因。     

Simple and Sensitive Method for Identification of Human DNA by Allele-Specific Polymerase Chain Reaction of FOXP2

Abstract:  The forkhead box P2 ( FOXP2 ) gene is specifically involved in speech and language development in humans. The sequence is well conserved among many vertebrate species but has accumulated amino acid changes in the human lineage. The aim of this study was to develop a simple method to discriminate between human and nonhuman vertebrate DNA in forensic specimens by amplification of a human-specific genomic region. In the present study, we designed an allele-specific polymerase chain reaction (PCR) using primers to amplify smaller than 70-bp regions of FOXP2 to identify DNA as being of human or nonhuman, including ape, origin. PCR amplification was also successfully performed using fluorescence-labeled primers, and this method allows a single PCR reaction with a genomic DNA sample as small as 0.01 ng. This system also identified the presence of human DNA in two blood stains stored for 20 and 38 years. The results suggested the potential usefulness of FOXP2 as an identifier of human DNA in forensic samples.     

Concordance Between the AmpFℓSTR® MiniFiler™ and AmpFℓSTR® Identifiler® PCR Amplification Kits in the Kuwaiti Population

Abstract:  The AmpFℓSTR^® MiniFiler™ polymerase chain reaction amplification kit, developed and supplied by Applied Biosystems, complements the AmpFℓSTR^® Identifiler^® polymerase chain reaction amplification kit (Applied Biosystems, Warrington, U.K.) by improving the success rate when profiling DNA that is degraded or contains inhibitors. Before applying the MiniFiler™ kit to casework, the profiles from 200 unrelated Kuwaitis were compared to Identifiler^® profiles. Concordance was observed for 99.875% (1598 of 1600) of the compared STR loci. The two discordant profiles displayed allelic dropout: one at the D13S317 locus due to nonamplification of allele 10 in the MiniFiler™ profile, and one at the D18S51 locus due to nonamplification of allele 18 in the Identifiler^® profile.     

Validation of the AMPFℓSTR® MiniFilerTM PCR Amplification Kit for Use in Forensic Casework*

Abstract: The AmpF?STR^® MiniFiler^TM PCR Amplification Kit is designed to genotype degraded and/or inhibited DNA samples when the AmpF?STR^® Identifiler^TM PCR Amplification Kit is incapable of generating a complete genetic profile. Validation experiments, following the SWGDAM guidelines, were designed to evaluate the performance of MiniFiler. Data obtained demonstrated that MiniFiler, when used in conjunction with Identifiler, provided an increased ability to obtain genetic profiles from challenged samples. The optimum template range was found to be between 0.2 and 0.6 ng, with 0.3 ng yielding the best results. Full concordance was achieved between the MiniFiler kit and Identifiler kit except in a single case of a null allele at locus D21S11. Numerous instances of severe heterozygous peak imbalance (<50%) were observed in single source samples amplified within the optimum range of input DNA suggesting that caution be taken when attempting to deduce component genotypes in a mixture.     

Determining an Optimal Sequence for Chemical Development of Latent Prints on Cartridge Casings and Shotgun Shells*,†

Abstract: In developing latent prints on cartridge casings and shotgun shells, multiple chemical processes should be used in order to obtain the best results. In Phase I, this study established an optimal chemical sequence for both Brass and Nickel cartridge casings based on six sequences involving four chemicals: Cyanoacrylate, Black Powder, Rhodamine 6G and Acidified Hydrogen Peroxide. Phase II was a validation study of Phase I involving a random sample of both Brass and Nickel cartridge casings, which were processed according to the determined optimal sequences. In addition, ribbed shotgun shells were processed under Phase I results and determined to be dependent upon the utilization of a CrimeScope at 515 nm. Consideration should be given to the type of cartridge case being examined. Although limitations exist, some chemical sequences undeniably work better than others. All photographs were manipulated with Adobe^® Photoshop^®. All results were verified by a senior latent print examiner.

Auf Grund der Art 8 iVm 13 MRK und aus rechtsstaatlichen Erw?gungen muss neben einer Erteilung von Amts wegen auch ein Antragsrecht des in seinen subjektiven Rechten Betroffenen auf Erteilung eines humanit?ren Aufenthaltstitels vorgesehen sein.     

Alternative Verursachungskonkurrenz mit dem Zufall – Die Wahrscheinlichkeit als Haftungsgrund?

Der Arbeitsgruppen-Entwurf eines neuen ?sterreichischen Schadenersatzrechts sieht vor, dass die Verursachungskonkurrenz mit dem Zufall zu einer Anteilshaftung führt. Die Verfasser des Entwurfs konnten sich dabei an der hM orientieren. Deswegen wird im Folgenden die alternative Konkurrenz mit dem Zufall auf der Grundlage der lex lata beurteilt, um den Ausgangspunkt des Entwurfs bewusst zu machen. Dabei wird gezeigt werden, dass die hM mit dem geltenden Recht nicht vereinbar ist. Die ge?u?erten Bedenken sprechen überdies gegen eine Normierung der Anteilshaftung im Zuge der Reform des Schadenersatzrechts.
