Enzymatic detection of γ-hydroxybutyrate using aldo-keto reductase 7A2

Gamma-hydroxybutyrate (GHB) is a prescribed medication as well as a drug of abuse. Its detection in various matrices for in-field forensic scientists remains a challenge. We have developed an assay that uses aldo-keto reductase 7A2 (AKR7A2) for the specific determination of GHB in various drinks. AKR7A2 was purified using Ni-affinity chromatography. The Michaelis-Menten constant for the GHB oxidation reaction was 10 mM, and the minimum detection limit was 4 mM. Ethanol was not a substrate for AKR7A2. In a coupled reaction with NADP(+), phenazine methosulfate (PMS), and 2,6-dichlorophenolindophenol, various beverages (orange juice, milk, soda, and numerous alcoholic drinks) containing GHB turned from blue to light yellow. In a second coupled reaction where diaphorase replaced PMS, the presence of GHB also caused the expected change of color in various beers.

国产试剂盒Goldeneye~(TM)20A在亲权鉴定中的应用评估

目的评估GoldeneyeTM20A试剂盒在亲权鉴定中的应用价值。方法应用GoldeneyeTM20A试剂盒对289宗亲权鉴定案例中的FTA卡血样本基因组DNA进行PCR扩增,扩增产物用ABI 3130xl遗传分析仪进行毛细管电泳,GeneMapper v3.2和GeneMarker HID软件进行基因分型及统计学分析,并与IdentifilerTM、SinofilerTM、PowerPlex16 3种试剂盒进行比较。结果采用GoldeneyeTM20A试剂盒,累积非父排除概率(CPE)为0.999 999 996,累积个人识别能力(CPD)达0.999 999 999 999 999 999 999 932 44,与目前常用的3种试剂盒相比较,GoldeneyeTM20A试剂盒在不排除的案例中具有更高的CPI值;在排除的案例中具有更多的排除指标。结论国产GoldeneyeTM20A试剂盒在亲权鉴定中有较高的应用价值。     

金融机构并购的法律支持路径

并购是当前解决我国金融机构市场退出难题的最为实际有效的办法。然而好的制度设计才能使金融机构的并购真正富有成效,金融机构并购的相关立法必须体现谁受益谁承担成本的原则,只有真正减少行政干预,让金融机构立足市场,引入民营金融机构参与竞争,并尽早出台针对金融机构切实可行的并购实施细则,金融领域的市场退出机制方能得以建立。     

DNA甲基化检测方法及其法医学应用研究进展

DNA甲基化是表观遗传标记的重要组成部分,参与基因表达的调控,在生物发育、衰老以及肿瘤学等领域受到广泛关注。由于具有相对稳定性、可遗传、含量丰富、随龄变化等特点,在法医学领域,DNA甲基化可以作为DNA序列相关经典遗传标记的有效补充,用于年龄推断、组织体液来源检测、同卵双生子的鉴定等。DNA甲基化的检测方法主要有基于甲基化敏感限制性内切酶、重亚硫酸盐转化以及甲基化CpG结合蛋白等原理而建立的一系列方法,近年研究表明,第三代测序技术也可用于DNA甲基化检测。本文就DNA甲基化检测方法及在法医学领域的应用研究进行回顾与综述,以期为法医学实践提供参考。     

Microbiota Composition and Pulmonary Surfactant Protein Expression as Markers of Death by Drowning

Pathological diagnosis of drowning remains a challenge for forensic science, because of a lack of pathognomonic findings. We analyzed microbiota and surfactant protein in the lungs for a novel diagnosis of drowning. All rats were divided into drowning, postmortem submersion, and control groups. The water, lungs, closed organs (kidney and liver), and cardiac blood in rats were assayed by targeting 16S ribosomal RNA of Miseq sequencing. Lung samples were analyzed by immunohistochemical staining for surfactant protein A. The closed organs and cardiac blood of drowned group have a lot of aquatic microbes, which have not been detected in postmortem submersion group. Furthermore, intra‐alveolar granular staining of surfactant protein A (SP‐A) was severely observed in the drowned group than the postmortem submersion and control groups. The findings suggested that the presence of aquatic microbiota in the closed organs and increased expression of SP‐A could be markers for a diagnosis of drowning.
