Automated DNA extraction using the QIAsymphony platform: Estimation of DNA recovery from simulated forensic stains

The aim of this study was to evaluate the forensic protocol recently developed by Qiagen for the QIAsymphony automated DNA extraction platform. Samples containing low amounts of DNA were specifically considered, since they represent the majority of samples processed in our laboratory. The analysis of simulated blood and saliva traces showed that the highest DNA yields were obtained with the maximal elution volume available for the forensic protocol, that is 200 μl. Resulting DNA extracts were too diluted for successful DNA profiling and required a concentration. This additional step is time consuming and potentially increases inversion and contamination risks. The 200 μl DNA extracts were concentrated to 25 μl, and the DNA recovery estimated with real-time PCR as well as with the percentage of SGM Plus alleles detected. Results using our manual protocol, based on the QIAamp DNA mini kit, and the automated protocol were comparable. Further tests will be conducted to determine more precisely DNA recovery, contamination risk and PCR inhibitors removal, once a definitive procedure, allowing the concentration of DNA extracts from low yield samples, will be available for the QIAsymphony.     

Third-worldism: sensibility and ideology in Uruguay – from Third Position to the thought of Carlos Real de Azúa

This article examines the trajectory of the concepts ‘Third World’ and ‘Third-worldism’ in Uruguay, and attempts to prove that, although Third-worldism developed thoroughly as sensibility, it did not have the same success as ideology. The article examines authors and intellectual groups who reflected on the Third World, and especially on ‘tercerismo’ (Third Position) – understood as a set of ideas related to Third-worldism but not part of Third-worldism as such. It next explains the importance of the thought of Carlos Real de Azúa, identified as the main ideologist of Third-worldism in Uruguay. The research shows as a result that there was great concern about the Third World, especially in the 1960s and the 1970s, expressed in articles, reports and speeches, among others. Nevertheless, a full conceptualisation was never realised, except in the contribution made by Real de Azúa. The article concludes that, paradoxically, ‘tercerismo’ blocked the development of more elaborated third-worldist thought in Uruguay.

济南市住宅价格影响因素灰色关联度分析

住宅价格深刻影响经济社会发展,研究其影响因素可作为调控地区房价的参考和依据。以济南市作为具体研究区域,选取影响住宅价格的指标,运用灰色关联度方法进行分析。结果表明,国民生产总值、人均可支配收入、房地产开发完成投资额与住宅价格的关联度在0.9以上,房屋施工面积、住宅销售面积与住宅价格的关联度在0.8以上,其余指标的关联度则在0.8以下。通过调控房地产开发投资额、房屋施工面积及销售面积等可有效影响住宅价格,而住宅价格增长应与经济发展水平相适应。
