口蹄疫病毒3C蛋白酶的T4噬菌体表面展示

对FMDV 3C基因克隆质粒p3C-T进行PCR扩增,获得了口蹄疫病毒3C蛋白酶基因片段,将此片段与T4噬菌体整合质粒pR的EcoRⅠ酶切片段连接,转化E．coli,DH5α工程菌,经EcoRⅠ酶切、质粒PCR扩增、插入方向筛选及测序鉴定,成功获得了T4噬菌体重组整合质粒pR- 3C。将其转化E．coli E2后,与SOC基因缺失的噬菌体φT4-Z1同源重组,经溶菌酶依赖性筛选,获得了快速溶菌型噬菌体φT4-3C。经噬菌体PCR扩增、SDS-PAGE分析和Western-bot分析,重组噬菌体可展示约26 ku的重组蛋白;其大小与预期相符,具有免疫原性。     

鸡肝微粒体细胞色素P450酶系检测方法的建立

为建立鸡肝微粒体细胞色素P450(CYP450)酶系活性的测定方法,应用超速离心法提取了12只60日龄依莎公鸡的肝微粒体,采用Bradford法测定肝微粒体蛋白含量,紫外分光光度法测定CYP450酶系活性。结果显示,肝微粒体蛋白含量为11.32 mg/g±1.43 mg/g,CYP450含量为0.71 nmol/mg±0.08 nmol/mg,NADPH细-胞色素C还原酶活性为3.01 nmol/(min.mg)±0.26 nmol/(min.mg),AND活性为2.36 nmol/(min.mg)±0.18 nmol/(min.mg),ERND活性为1.21 nmol/(min.mg)±0.11 nmol/(min.mg),AH活性为0.25 nmol/(min.mg)±0.03nmol/(min.mg)。结果表明,建立的鸡肝微粒体CYP450酶系的检测方法灵敏可靠,重复性较好。     

Public health measures against neonatal tetanus on the island of Vestmannaeyjar (Iceland) during the 19th century

The article presents a study of neonatal tetanus on the tiny island of Vestmannaeyjar (Iceland) during the 18th and 19th centuries. At an early date, Vestmannaeyjar was known for its high levels of mortality from neonatal tetanus. This appalling mortality is analysed, inter alia, on the basis of parish registers at the individual family level. During the late 18th and early 19th centuries, more than three out of four newborns on the island died during the first 2weeks of life. At the beginning of the 19th century, Icelandic and Danish authorities had already showed great interest in improving infant survival on Vestmannaeyjar. In 1827 a physician was appointed to the island and by the late 1840s the disease was successfully fought on the island. The achievement on Vestmannaeyjar is a good example of how the sanitary movement was able to bring about important improvements in infant survival long before the breakthrough of the bacteriological revolution.     

Penetrating Trauma: Representing Rape in the Historical Drama Series Feast of the Uninvited

ABSTRACT

In the drama series Feast of the Uninvited, director Katinka Heyns portrays some of the pain and trauma experienced by families during the South African War (also known as the Anglo-Boer War). Within the storyline, the character of Magrieta Van Wyk is raped by a Boer man. This is one of the first physical “on screen” representations of rape in an Afrikaans television production, and because of this, the MNET series ignited a lot of controversy among audiences. Through this representation, Heyns challenges the traditional notion of the “pure and heroic” Boer hero that has become a well-known archetype within films and drama series set against this time period. While witnessing Magrieta’s rape on screen, we only later find out that the black housemaid Siena was also raped in a concentration camp reserved for black women and children. It is one of the first audio-visual case studies that mention black women’s suffering during this war, but it is still only given a brief glance. The author concludes that the collaborative effort of Heyns and cinematographer Koos Roets, combined with powerful performances, recreates the harshness of the South African War, and more specifically the traumatic experiences of women during this war.

2007年爆发的美国次贷危机早已转变为席卷全球的国际金融危机。由于东盟四国的银行业正处在改革与调整时期,其银行业发展轨迹明显不同于其他国家和地区。东盟四国银行业系统的基本面良好,甚至在危机期间还逆势而行,主要得益于东盟四国在1997年东亚金融危机爆发之后所采取的一系列有效的改革措施及此次金融危机东盟四国政府所采取的积极应对举措。     

Comparative analysis of gamma-hydroxybutyrate and gamma-hydroxyvalerate using GC/MS and HPLC

This paper describes two analytical techniques used to separate and quantify gamma-hydroxybutyrate (GHB) and gamma-hydroxyvalerate (GHV). The first technique was a N,O-bis(trimethylsilyl)triflouro-acetimide-trimethylchlorosilane derivatization, followed by gas chromatography/mass spectrometry analysis using an HP-5 capillary column at a rate of 1.0 mL/min with a run time of 9.25 min. This technique was found to be sensitive (LOD 1 pg on column) and gave a low average error (5%) in a beverage study. When supplemented by a surrogate spike, the method yielded 97% analyte recovery from beverages. The second technique was high-performance liquid chromatography/UV (HPLC/UV) using a C-18 column with a (20:80% v/v) methanol:dibasic phosphoric buffer (10 mM, pH 3) at a rate of 1.00 mL/min with a run time of 7.5 min. UV detection occurred at 254 nm. This method was found to be less sensitive (LOD 0.05 microg on column) for direct analysis of aqueous samples. To remove interferences seen in the beverage study, a liquid-liquid extraction before HPLC analysis was tested. However, a decreased sensitivity (LOD 100 microg on column) and irreproducible peak profiles resulted.     

Bundesstaat und Gleichheitsgrundsatz

Gleichheit und Bundesstaatlichkeit stehen in einem Spannungsverh?ltnis. Nicht nur, dass die von der Bundesstaatstheorie geforderte "arithmetische Gleichheit" angesichts realpolitischer Disparit?ten zwischen den Gliedstaaten in der Praxis kaum durchsetzbar ist, sondern auch die im Wesen des bundesstaatlichen Prinzips gelegene f?derale Differenzierung der einzelnen Landesrechtsordnungen birgt Sprengkraft in sich. Der vorliegende Beitrag geht der Frage nach, in welchem Verh?ltnis Bundesstaatlichkeit und Gleichheitsgrundsatz zueinander stehen. Dabei wird auch untersucht, welche Auswirkungen die Einbindung ?sterreichs in die Europ?ische Union auf dieses Verh?ltnis hat.

五四运动前后，中国社会的思想变动是鸦片战争后东西文化冲突的反映，先进人士在中国社会向何处去的探索中，经历了由浅到深的思想演变过程，至五四运动前夕，进入了文化层面的思考。五四运动前后的思想变动集中表现为道路的重新选择。先进知识分子经过反复比较，最后选择走俄国十月革命的道路。思想变动的最后结果是马克思主义成为中国历史发展的必然选择，从而导致中国革命道路的方向性交化。     

Klagsanmerkung nach § 27 Abs 2 WEG 2002 beim Miteigentumsanteil des Bekl, der diesen im Wege bücherlicher Vormerkung bereits ver?u?ert hat

Die nach § 7 Abs 1 KO ex lege eingetretene Unterbrechung des Zivilprozesses wirkt sich zwar nicht auf Au?erstreitverfahren (hier: über die Klagsanmerkung nach § 27 Abs 2 WEG 2002 iVm § 8a KO und § 25 Abs 1 Z 4 Au?StrG) aus, so dass die Anmerkung grunds?tzlich zu bewilligen ist. Ist die Sechsmonatsfrist für die Klagsführung gem § 27 Abs 2 WEG 2002 gegenüber dem grundbücherlich vorgemerkten neuen Wohnungseigentümer aber bereits abgelaufen, ist der Anmerkungsantrag bei dessen Miteigentumsanteil abzuweisen.     

羊痘病毒P32蛋白编码基因的克隆及表达

为获得山羊痘病毒膜蛋白重组P32抗原,根据其基因序列设计合成了1对特异性引物, 对CaPV P32基因进行了PCR扩增,产物大小约为969 bp;产物回收纯化后,将其克隆至pBAD／ Thio-TOPO载体中,转化TOP10大肠埃希氏菌感受态细胞,与已报道的多株CaPV P32基因序列的同源性高达99％;相应地,氨基酸序列同源性为98％。经测序筛选出阳性克隆,该重组菌株经 L-arabinose诱导,可稳定、高效地表达CaPV P32抗原。表达蛋白为融合蛋白,分子质量约 51．53 ku。
