Sensitive Method for the Confident Identification of Genetically Variant Peptides in Human Hair Keratin

Recent reports have demonstrated that genetically variant peptides derived from human hair shaft proteins can be used to differentiate individuals of different biogeographic origins. We report a method involving direct extraction of hair shaft proteins more sensitive than previously published methods regarding GVP detection. It involves one step for protein extraction and was found to provide reproducible results. A detailed proteomic analysis of this data is presented that led to the following four results: (i) A peptide spectral library was created and made available for download. It contains all identified peptides from this work, including GVPs that, when appropriately expanded with diverse hair-derived peptides, can provide a routine, reliable, and sensitive means of analyzing hair digests; (ii) an analysis of artifact peptides arising from side reactions is also made using a new method for finding unexpected modifications; (iii) detailed analysis of the gel-based method employed clearly shows the high degree of cross-linking or protein association involved in hair digestion, with major GVPs eluting over a wide range of high molecular weights while others apparently arise from distinct non-cross-linked proteins; and (v) finally, we show that some of the specific GVP identifications depend on the sample preparation method.     

旋毛虫新生幼虫cDNA文库的构建

为了克隆和研究旋毛虫新生幼虫功能性抗原基因,采用酸性异硫氰酸胍-酚-氯仿一步法提取新生幼虫总RNA,Oligo(dT)纤维素柱纯化mRNA,反转录合成第一链cDNA及第二链cDNA,用CHROMA SPIN-400柱离心层析纯化后,与载体λZAP Express连接.体外包装后得到中国猪旋毛虫(Trichinella spiralis)分离株新生幼虫cDNA文库.文库容量为2.0×106,重组率为98.6%,插入片段长度在0.4×103-2.0×103bp.

Now you see it,now you don’t: digital images and the meaning of ‹possession’

The advent of digital technology has had a profound impact on the production and distribution of child pornography. The international trade in such material is a major focus of law enforcement agencies around the world. Central to these efforts is the offence of ‹possession.’ However, concepts of possession in the criminal law evolved in the context of tangible items such as drugs, and their application to intangible data presents significant challenges. Drawing upon the laws of Australia, Canada, England and the United States, this article analyses the concept of criminal possession in a digital context. While courts and legislatures are adapting to these new challenges, in some cases it is argued that the offence of possession is being stretched too far. It is suggested that some of these difficulties may be avoided by utilising an offence of ‹accessing’ child pornography. Senior Lecturer, Faculty of Law, Monash University. 高职院校图书馆是高职教育教学的重要组成部分,是教师科研、教学的参谋部,是学生学习的"第二课堂".然而其馆员的知识结构和服务技能等综合素质不能适应高职教育的需要.高职教育要发展,高职图书馆也要发展,提高其馆员素质是高职图书馆的一项迫在眉睫的任务.我们应采取注重职业道德培养、加强在职培训、定期组织学术交流等措施,大幅度提高高职院校图书馆员的综合素质.     

Information technology and education： Partnership vs. competition

Information technology is coming strong to have influential impact upon what is going on in the formal education sector. During the past ten years, information technology almost became the first job choice among all the new faces finding their jobs. Job opportunity also has its great impact upon what is going on in the k-12 education. Ideally, information technology is to enhance a teacher＇s teaching. However, on-site classroom observations as revealed here, identified several things that we might want to work more to provide better learning environment for the young generation on an individual basis. We have to admit that although memorization of the old day＇s factual knowledge and skill is important, but is going to provide little help for the generation to come. Information technology brings brand new era to human beings, and new format of schooling can take different forms from the ones most of us had been experiencing during our school lives.
