羊源巴贝斯虫未定种甘肃敦煌株的发现

采用动物接种法将采自甘肃敦煌的15只绵羊血液混合后感染1只除脾绵羊,感染后逐日做血涂片检查。结果显示,在感染后第10天的血片上出现了1种大型巴贝斯虫。虫体形态以双梨子形、单梨子形、圆形及卵圆形为主。在感染后第12天,感染羊体温升至41.1℃。染虫率达到0.125%,之后逐渐下降。无菌采集该羊血液,提取全血基因组,用梨形虫通用引物进行PCR扩增全长18SrRNA基因,得到长约1 700bp的片段,将其克隆并测序(登录号:HQ730762),与GenBank中的其他巴贝斯虫的序列进行比较,发现其与新疆巴贝斯虫未定种的同源性最高,达到99.8%。本研究发现甘肃敦煌又是一个新疆羊巴贝斯虫未定种的疫源地。     

The use of Hemastix® severely reduces DNA recovery using the BioRobot® EZ1

Abstract: The choice of reagents for presumptive tests for blood, and subsequent extraction methodologies, can significantly affect both the quantity and quality of purified DNA. Blood samples directly tested with Hemastix^® yielded <1% of the DNA recovered from untested samples when purified using the Qiagen BioRobot^® EZ1 and EZ1^® DNA Investigator kit. Full short tandem repeat profiles were obtained from both tested and untested samples, suggesting that the Hemastix^® reagent(s) affect DNA binding, rather than produce DNA damage. The Hemastix^® inhibition of DNA yield could be overcome by the addition of MTL buffer to the sample prior to extraction. Laboratories may wish to modify current procedures for extracting blood samples, utilize other extraction/purification methodologies, or inform their submitting agencies to avoid direct exposure of questioned bloodstains to Hemastix^® reagents.     

犬源韦氏巴贝斯虫的分子鉴定和系统发育研究

无菌采集疑似巴贝斯虫感染血样,进行全血基因组DNA的抽提,用保守引物5-22F和1661R进行巴贝斯虫18SrRNA基因的PCR扩增,扩增产物经克隆、测序后,获得大小为1 677bp的核苷酸序列。序列比对和系统发育分析发现,该序列与韦氏巴贝斯虫(AB083374)18SrRNA基因的相似性达100%,而与犬巴贝斯虫(AY072926)、罗氏巴贝斯虫(L19079)和吉氏巴贝斯虫(AF271081)18SrRNA基因的相似性分别为98.0%,95.2%和93.4%。从分子水平证实了韦氏巴贝斯虫在广东宠物犬的存在,同时也对巴贝斯虫的分类进行了讨论。     

The Territorial Scope of Employment Legislation and Choice of Law

Traditionally, the determination of the territorial scope of the statutory rights conferred by employment legislation forming part of English law has been regarded as an issue entirely disconnected from the choice‐of‐law process. Indeed, this view formed the basis of the key decision addressing the problem of territoriality, Lawson v Serco, decided by the House of Lords in 2006. After presenting the current state of the law with regard to the territorial scope of employment legislation, this article takes a critical look at Lawson v Serco. It is argued that the ‘European’ choice‐of‐law rules must have a greater importance for determining the territorial scope of employment legislation and, consequently, that the approach pursued in Lawson v Serco is no longer correct, if it ever was, and should not be followed in the future.     

Recognition of Early Myocardial Infarction by Immunohistochemical Staining with Cardiac Troponin‐I and Complement C9*

Abstract: The diagnosis of early myocardial infarction (MI) after death, especially in the first few hours (c. 6 h) after the onset of MI, poses a challenge to the forensic pathologists. During this time, the damaged myocardium does not show grossly identifiable morphological changes and may not be recognized even with routine histological microscopic examination. However, the infarcted cardiac tissue releases certain chemicals that can be detected microscopically, two of these being cardiac troponin‐I (CT‐I) and complement C9 (C9). This study utilizes the importance of these two biomarkers immunohistochemically in an attempt to identify this early phase of MI. This study reveals that the early phase of MI of <6 h duration may be detected through immunohistochemical staining with CT‐I and C9. The ischemic/infarcted cardiac myofibers in the <6 h group display reduced/absent CT‐I staining as well as positive C9 staining.

群众路线是党的工作的生命线,也是党的根本政治路线和根本组织路线,一直作为党的优良传统作风而不断继承和创新。但从党的历史进程来看,群众路线的践行往往受到不良工作作风和不正确的工作方法等主观因素的干扰,究其根源,在于群众路线的运行机制一直缺乏一种自下而上的动力机制和民主保障机制。通过发展人民民主,保障人民主体地位,确定人民当家作主的权利,无疑会为新形势下践行群众路线提供长效性运作的内在动力。
