目的建立自动化工作站同步提取不同种类涉案法医生物检材DNA的新方法。方法选用TECAN Freedom EVO100.4、75—2型自动化提取、加样工作站,采用磁珠法及Chelex-100法对各类涉案生物检材进行DNA提取、PCR扩增、毛细管电泳检测其STR分型,进行比较测试。在“全国公安机关DNA数据库应用系统”中建立并应用实验室信息管理系统(LIMS)模拟实施规范化DNA检案。结果1552份各类检材,采用工作站-磁珠法提取DNA效果最佳,STR检测成功率为95%,工作站-Chelex法为88%;二者分别与其手工提取法比较,成功率无明显差异。92个样本同期检测,自动化工作站较手工操作DNA检案时间可缩减1.25倍。结论工作站域珠法提取涉案检材DNA,可获得满意的STR分型结果。应用LIMS管控,可有效防控污染,明显提高检案效率及鉴定质量。 相似文献

目的研究不同稀释度胍盐裂解液对自动化提取工作站DNA回收效率的影响。方法制备不同浓度的DNA样本,并将EQ1000法医DNA提取试剂盒中的胍盐裂解液梯度稀释,在本实验室配置的自动化提取工作站上,运行"工作站磁珠法"提取程序,AB-7500型荧光定量PCR仪检测提取前后的样本DNA浓度。选取2~7号梯度组DNA回收效率数据进行统计学检验,比较不同稀释度胍盐裂解液的处理对样品DNA回收效率的影响。结果 90%~60%稀释度胍盐裂解液组样品回收效率在66%左右;50%~20%稀释度组样品DNA回收效率在41%左右;稀释度在50%以下与60%以上组样品回收效率之间差异极显著。结论应用"工作站磁珠法"提取生物检材DNA,其DNA回收效率受胍盐裂解液浓度的影响。 相似文献

业净增。研究建议:应积极推进新业态就业的综合配套保障,提高技术型群体零工经济就业的能力;大力加强
相关制度体制保障,支持基于新技术的“副业创新”;多方鼓励产业交叉,加大自动化就业与消费升级相结合

Abstract The present study examined the effects of automating the Relevant–Irrelevant (RI) psychophysiological detection of deception test within a mock-screening paradigm. Eighty participants, recruited from the local community, took part in the study. Experimental design was a 2 (truthful/deceptive) by 2 (human/automation) factorial. Participants in the deceptive conditions attempted deception on two items of an employment application. Examinations conducted with the automated polygraph examination were significantly more accurate than examinations conducted by the human polygraph examiner. Statistical analyses revealed different patterns of physiological responses to deceptive items depending upon the automation condition. Those results have potentially interesting theoretical implications. The results of the present study are clearly supportive of additional efforts to develop a field application of an automated polygraph examination. 相似文献

Advances in information technologies over the last two decades have offered the promise of revolutionizing workplaces and other settings by expanding the capabilities of individuals working in various professions. Enabled by high-speed computing systems and marked by an intensification of command and control properties, new technologies have expanded, and in some cases intentionally surpassed, the scope of the senses. Technologies such as drones, surveillance cameras, and airport and border security systems promise the seemingly infinite catchment and infinitesimal account of human actions, while computer processing software provides real-time data on large-scale transactional processes occurring in monetary systems and security markets. Charting, tracking, and mapping functions open up new vistas for quantifying social behaviors and detecting misbehaviors, creating new frontiers for criminal justice, the military, and economic activity. The papers in this special issue reveal the processes guiding the rapid transformation and flexible adaptation of institutional structures, economic and financial systems, and threat surveillance and military/police mobilization in the near future. 相似文献

Abstract: Laboratory automation has the ability to increase the throughput and efficiency of laboratory processes to keep pace with current backlogs and requests for analysis. This paper addresses the specific studies employed to properly evaluate an automated method for DNA quantitation setup using Applied Biosystems Quantifiler? Human DNA Quantification kit on a Biomek® 3000. The calibration of robotic pipetting as well as comparison with manually performed steps confirmed the accuracy of the automated methods used. Reproducibility studies evaluated differences between robotic and manually prepared human DNA standard curves. Additional studies examined DNA samples of known quantities, extract storage formats, sensitivity, and an assessment of contamination. The Biomek® 3000 not only demonstrated reproducibility and accuracy that equaled or surpassed the manual method but also revealed a contamination‐free method to replace the multiple pipetting steps required during quantitation setup. 相似文献

We have implemented and validated automated methods for washing FTA Card punches containing buccal samples and subsequent PCR setup using a Sias Xantus automated liquid handler. The automated methods were controlled by worklists generated by our LabWare Laboratory Information Management System (LIMS). The protocols were validated according to EN/ISO 17025 for use with STR amplifications kits AmpF?STR® Identifiler® and Y-filer® (Applied Biosystems). 相似文献
