Testing Times Ahead: Non‐Invasive Prenatal Testing and the Kind of Community We Want to Be

This article reviews the Nuffield Council on Bioethics’ report on Non‐Invasive Prenatal Testing (NIPT); and introduces two general questions provoked by the report – concerning, respectively, the nature and extent of the informational interests that are to be recognised in today's ‘information societies’ and the membership of today's ‘genetic societies’. The article also considers the role and nature of the Nuffield Council. While the Council's report identifies a range of individual and collective interests that are relevant to determining the legitimate uses of NIPT, we argue that it should put these interests into an order of importance; we sketch how this might be done; and we suggest that, failing such a prioritisation of interests, the Council should present its reflections in a way that engages public debate around a number of options rather than making firm recommendations.     

An Incidental Diffuse Midline Glioma Found at Autopsy

This case shows an unexpected midline glioma found at autopsy. Two siblings were riding on a single bicycle on the side of a road. The 13‐year‐old brother was seated and steering the bicycle, while the 14‐year‐old sister held onto the back. The bicycle veered left into traffic and was struck by a vehicle. The siblings were admitted to the local Level 1 trauma center, but both later succumbed to injuries. Autopsies were performed on the children, including brains for neuropathologic evaluation. The brother was found to have an infiltrating astrocytoma located in the left middle cerebellar peduncle, with extension to the pons and medulla. His hospital course included several imaging studies using CT and MRI modalities. However, this lesion was not identified until the postmortem neuropathologic examination. This rare case shows the continued need for postmortem autopsy and the current limitations of medical imaging.     

Detection and Identification of Psilocybe cubensis DNA Using a Real‐Time Polymerase Chain Reaction High Resolution Melt Assay,

Psilocybe cubensis, or “magic mushroom,” is the most common species of fungus with psychedelic characteristics. Two primer sets were designed to target Psilocybe DNA using web‐based software and NBCI gene sequences. DNA was extracted from eighteen samples, including twelve mushroom species, using the Qiagen DNeasy^® Plant Mini Kit. The DNA was amplified by the polymerase chain reaction (PCR) using the primers and a master mix containing either a SYBR^® Green I, Radiant? Green, or LCGreen Plus^® intercalating dye; amplicon size was determined using agarose gel electrophoresis. The PCR assays were tested for amplifiability, specificity, reproducibility, robustness, sensitivity, and multiplexing with primers that target marijuana. The observed high resolution melt (HRM) temperatures for primer sets 1 and 7 were 78.85 ± 0.31°C and 73.22 ± 0.61°C, respectively, using SYBR^® Green I dye and 81.67 ± 0.06°C and 76.04 ± 0.11°C, respectively, using Radiant? Green dye.     

5F-UR-144的热不稳定性研究

目的利用气相色谱-质谱法(GC-MS)、液相色谱-四级杆-飞行时间质谱(LC-Q-TOF/MS)和核磁共振光谱法(NMR)研究合成大麻素5F-UR-144遇热分解的具体变化情况。方法对照品用无水乙醇定容稀释后,经GC-MS和LC-Q-TOF/MS检测得到对应的色谱图和质谱图;对照品分别于常温和280℃密封加热后用无水乙醇定容稀释,经LC-Q-TOF/MS和NMR检测得到对应的质谱图、¹H和¹³C核磁共振波谱;对照品分别于常温逐步提升至300℃密封加热后用无水乙醇定容稀释,经LC-Q-TOF/MS检测得到对应的色谱图。结果5F-UR-144在高温下会开环产生新的物质;5F-UR-144从130℃开始分解,随着温度升高分解程度提升,240℃时分解率达到98%;随着温度继续升高,超过260℃,分解产物会碳化。结论基于5F-UR-144的热不稳定性,在检测时应考虑若通过烫食方式吸食5F-UR-144,其进入人体的成分会发生变化;气相色谱或气相色谱-质谱法不适合定量检测5F-UR-144。     

In Search of the Holy Grail in Public Service: A Study on the Mediating Effect of Public Service Motivation on Organizational Politics and Outcomes

ABSTRACT

There appears to be a research gap in terms of examining whether PSM will hold fast even in the midst of a politically charged work environment. This article investigates how highly motivated government employees would react to the potentially adverse effects of political perception (POP) on organizational commitment, job satisfaction and workplace stress. The findings indicated that PSM exists among the respondent municipal social workers in the Philippines and that this motivation directly translates to positive outcomes. However, PSM did not have the expected mediating effect which is a departure from the commonly held view about its buffering power and mitigating role.

Morphometric Investigation of Death by Asphyxia

Abstract:  The aim of this study was to investigate the possibility of distinguishing deaths by asphyxia from those due to natural causes by comparing morphometric measurements in lungs. The study population comprised 27 subjects: 14 cases of death by asphyxia (hanging or drowning) and 13 cases of sudden natural death. Eighty parenchyma sections were used for each subject. Data were collected by computerized image analysis. Measurements aimed at quantifying, as percentages, pulmonary parenchyma (mean values of positive-fraction areas [PFA] and their standard deviations). Subjects who died of natural causes and of drowning showed a relative internal homogeneity compared to those who hanged. Results show significant discrimination between drowned subjects and those dying of natural causes (mean of PFA p  = 0.01) and between hanged and drowned subjects (SD of PFA p  = 0.04). Attention must be paid to the possible role played by senile emphysema. The method is proposed as a complementary tool in forensic cases.
