Here we report a case where a discrete section of the cytological slide preparation of a female individual was obviously contaminated with pleura liquid of a female tumor patient. Analysis of the cancerous pleura liquid and the healthy cells of the slide preparation showed different DNA profiles, indicating that the material originated from two different female individuals. The DNA profile of the cell mixture revealed a heterogenous pattern whereby the alleles could be assigned to the healthy and the tumor patient. Loss of heterozygosity (LOH) was observed in four of eight short tandem repeat systems for the pleura liquid and the cell mixture. Despite the low amount of DNA on the slide preparation and the occurrence of LOH, it was possible to clarify the case and to support the assumption that a drop of cancerous pleura liquid contaminated the cytological slide.  相似文献

摘要:目的:观察虫对慢性血瘀型大鼠血液流变性和红细胞免疫功能的影响。方法:模拟阴虚火旺复制慢性血瘀大鼠模型,用毛细血管法测定不同切速下的全血比粘度、血浆比粘度,常规法测定红细胞压积。参照郭氏法测定红细胞免疫粘附功能指标,并将其与血液流变性有关指标作一相关分析。结果:虫能明显降低瘀血大鼠的全血比粘度、血浆比粘度和红细胞压积;提高瘀血大鼠红细胞C3b受体花环率,使红细胞免疫复合物花环率下降,血清红细胞免疫粘附促进因子活性增强,而抑制因子减弱;血液粘度与红细胞C3b受体花环率的相关分析呈显著负相关。结论:虫不仅有活血化瘀作用,还具有促进红细胞免疫功能的功效。  相似文献

目的 通过观察益气健脾通便方对慢传输型便秘(slow transit constipation,STC)大鼠Cajal间质细胞(Cajal interstitial cells,ICC)和一氧化氮(nitric oxide, NO)、一氧化氮合酶(nitric oxide synthase,NOS)的影响,探讨其作用机制。方法 将60只SPF级雄性8周龄Wistar大鼠随机分为正常组,模型组,益气健脾通便方低、中、高剂量组和莫沙必利组,每组10只。采用“复方地芬诺酯灌胃法”复制STC大鼠模型。模型复制后,各组大鼠连续给药4周,观察每组大鼠的一般情况,测定粪便粒数、粪便含水率、肠道推进率和结肠组织ICC表达量及NO、NOS含量。结果 益气健脾通便方能改善大鼠的一般情况,提高大鼠的粪便粒数、粪便含水率和肠道推进率(P<0.05),提高结肠组织ICC表达量(P<0.05),减少神经递质NO、NOS的含量(P<0.05)。结论 STC的发病机制可能与ICC表达量降低及NO、NOS水平升高有关。  相似文献

目的:探讨蜂毒素诱导人T淋巴细胞白血病细胞株6T-CEM的凋亡作用。方法:利用DNA断裂点标记法(TUNEL)、DNA凝胶电泳、活细胞拒染以及光镜方法研究6T-CEM细胞的凋亡。结果:5μg/ml蜂毒素作用4h及4μg/ml蜂毒素作用24h诱导6T-CEM出现明显的凋亡特征。结论:蜂毒素在杀伤6T-CEM的同时能明显诱导细胞凋亡。  相似文献

为了研究羊口疮病毒(ORFV)F1L基因,以该病毒基因组DNA为模板,应用聚合酶链式反应(PCR)扩增得到1011bp的F1L基因,将其插入pMD18-TEasy克隆载体,构建了F1L基因克隆重组质粒;再将该基因片段插入pGEX-6P-1表达载体,通过PCR、限制性内切酶双酶切和测序鉴定,得到F1L基因插入方向和读码框结构均正确的阳性质粒,表明,成功构建了羊口疮病毒F1L基因的重组表达载体。通过对F1L基因序列进行生物信息学分析,预测发现F1L蛋白亲水、无信号肽,B细胞表位主要位于第4~8位、第16~22位、第32~53位、第59~73位、第82~99位和第123~133位氨基酸。这将为建立ORFV诊断方法、制备单克隆抗体及合成肽疫苗奠定基础。  相似文献

为了探讨褪黑激素(Mel)对犬脂肪干细胞(ADSCs)向雪旺细胞(SCs)诱导分化过程中凋亡发生的影响。本实验将犬ADSCs诱导分化为SCs,并使用免疫荧光法进行鉴定。采用CCK8法选取Mel的最佳浓度。应用q RT-PCR法和Western-blot法检测无Mel组和Mel处理组中ADSCs向SCs诱导过程中细胞凋亡相关基因Caspase-3、Caspase-8、Bcl-2和Bax的m RNA和蛋白表达水平。结果显示,50 nmol/L Mel处理过的细胞活力最高;ADSCs向SCs诱导后S-100和GFAP呈阳性表达,并且观察到Mel的添加并不会影响到SCs的表型特征。ADSCs向SCs诱导后Bcl-2表达量明显低于对照组,Caspase-3、Caspase-8和Bax的表达量显著高于对照组,细胞凋亡较多;在加入Mel后Bcl-2表达量显著升高,Caspase-3、Caspase-8和Bax的表达量显著下降。结果说明Mel可以显著抑制犬ADSCs向SCs诱导分化过程中的凋亡发生。  相似文献

Infants born to diabetic mothers are at increased risk for symptomatic hypoglycemia and death after birth. A 36-year-old G4P3 mother with a history of gestational diabetes and newly diagnosed type II insulin-dependent diabetes gave birth at home, in the care of a midwife, to a macrosomic infant girl (10 lbs.). Several hours after birth, the infant became lethargic and was found to be hypoglycemic (blood sugar: 28 mg/dL). Glucose and sugar water were administered by the midwife; however, the infant continued to decompensate. Emergency medical services were called, and the infant was transported to the hospital where, despite resuscitative efforts, she died. An autopsy and review of the literature was performed. At autopsy, characteristic features of maternal–fetal glucose dysregulation were identified, including fetal macrosomia, cardiomegaly, hepatomegaly, and severe pancreatic islet cell hypertrophy/hyperplasia. Developmental abnormalities and other potential causes of death were not identified. Although deaths due to hypoglycemia cannot be reliably diagnosed postmortem using vitreous glucose levels, a clinical history of maternal glucose dysregulation in combination with certain gross and histologic findings should prompt a pathologist to consider maternal–fetal glucose dysregulation as a diagnosis of exclusion and cause of death.  相似文献

A 26‐year‐old Caucasian sailor, with no past medical history aside from headache for the last 1 week, was found dead in his cabin. The body was stored in a refrigerator on board and disembarked for autopsy 3 days later. Autopsy showed a large, nodular, necrotic and hemorrhagic retroperitoneal mass, and smaller hemorrhagic nodules in the brain, lungs, liver, and left kidney, with the brain being markedly edematous. Both testes were descended and normal. Histologically, the retroperitoneal mass showed a malignant mixed germ cell tumor comprising choriocarcinoma, embryonal carcinoma, and teratoma components. Retroperitoneal extragonadal germ cell tumors are uncommon, and this case of a young male who presented with headache and sudden death due to metastases is extremely rare.  相似文献
