Fast PCR amplification using AmpFlSTR Identifiler: Second report

At ISFG2007, an earlier conference, we presented reports on two fast PCR cycling methods using AmpFlSTR Identifiler. In our current study, which involved PCR amplification using AmpliTaq Gold Fast PCR Master Mix, UP, and a standard PCR thermal cycler rather than the Fast PCR thermal cycler, we succeeded in allele typing while reducing PCR running times by half (to approximately 90 min).     

Case study of dissolving allele dropout

Allele typing performed using the original components of the AmpFlSTR Identifiler kit led to unbalanced peak heights and allele dropouts. However, allele typing was completely successful when substituting the QIAGEN Multiplex PCR Master Mix for the AmpFlSTR PCR Reaction Mix of the Identifiler kit. The successful range for allele typing was 250 pg to 1.5 ng of DNA. The running times for PCR amplification were reduced by about 30 min from the original protocol.     

Hippocampal Agenesis in an Individual who Engaged in Violent Criminal Behaviors After Discontinuing Carbamazepine and Paroxetine Treatment

Antidepressant discontinuation syndrome (ADS) occurs after abrupt discontinuation of an antidepressant medication. A 23‐year‐old man with right hippocampal agenesis demonstrated sexual crime (hypersexuality) since the age of eight and had been successfully treated with carbamazepine since the age of 13. He had required increased doses of paroxetine and carbamazepine owing to the development of an unstable affect after quitting his job. He abruptly stopped taking his medication for 3 days and his criminal behaviors re‐emerged. We examined changes in brain structure and activity before and after medication cessation, using MRI and functional MRI (fMRI). The image of a girl in a swimsuit increased activity in the thalamus only after medication discontinuation. The alteration in thalamic activity might induce hypersexuality. We conclude that a primary hypersexuality had been suppressed with carbamazepine and paroxetine treatment, and the discontinuation of the medication caused the hypersexuality.     

Fast PCR amplification of AmpFlSTR Identifiler

To increase throughput for DNA typing, we examined fast PCR cycling using AmpFlSTR Identifiler by three methods. In this study, we reduced PCR running times by 1/3 to 2/3 (approximately 1-2 h). This means DNA typing, including PCR reactions, can be completed within a timeframe ranging from 90 min to 2 h and 30 min.     

Providing credibility around the world: effective devices of the Cape Town Convention

This paper analyzes the characteristics and effects of the Cape Town Convention, which provides legal protection for investors in relation to asset-based aircraft financing. Some countries lack credibility for investors, who believe that the protection level of creditor rights in the country differs before and after investment. This time inconsistency problem results in credit rationing. Airlines in such countries cannot access the credit market, particularly the international capital market. This paper first examines why developing countries have been suffering from a lack of credibility and from credit rationing, and then explores how the Convention resolves this problem. We focus on two devices: the white list system and the system of declarations. The former gives the contracting states incentive not to break the Convention. The latter is considered to promote the ratification of the Convention by potential contracting states. These devices enable the Convention to attract a number of states.     

Proposal for internet-based Digital Dental Chart for personal dental identification in forensics

A dental chart is very useful as a standard source of evidence in the personal identification of bodies. However, the kind of dental chart available will often vary as a number of types of odontogram have been developed where the visual representation of dental conditions has relied on hand-drawn representation. We propose the Digital Dental Chart (DDC) as a new style of dental chart, especially for open investigations aimed at establishing the identity of unknown bodies. Each DDC is constructed using actual oral digital images and dental data, and is easy to upload onto an Internet website. The DDC is a more useful forensic resource than the standard types of dental chart in current use as it has several advantages, among which are its ability to carry a large volume of information and reproduce dental conditions clearly and in detail on a cost-effective basis.     

Regulatory Enforcement in Local Government in Japan

The Japanese inspectors usually adopt an "informal measure-oriented" style in enforcing violations of regulatory statutes. They prefer administrative guidance with no legally binding effect to compliance orders or criminal prosecution. These formal measures, they think, would erode the cooperative relationship with the regulatee, which is believed essential for cost-effective enforcement. In contrast to that in the United States, the Japanese enforcement process lacks citizen participation and information disclosure, this enabling the inspectors to deal with violations at their own pace.
