The solution parameters associated with gradient elution reversed phase high-performance liquid chromatography (HPLC) were characterized by evaluating the electrospray ionization mass spectrometry (ESIMS) response of selected smokeless powder additives. Using direct liquid infusion, the positive ion ESIMS responses were determined for three general classes of powder constituents: dialkylphthalate acid esters, N,N'-dialkyl-N,N'-diphenyl urea based stabilizers and nitroso-, nitro-, and dinitro- derivatives of diphenylamine. The relative ESIMS intensities of the powder components were investigated as a function of three solution parameters: ammonium acetate concentration, pH, and percent methanol. The effect of the ammonium acetate concentration demonstrates that the electrolyte is required for efficient ionization and the ESIMS intensity was optimal at a concentration of 2mM for the selected compounds, except 2,4'-dinitrodiphenylamine. The aqueous solution pH, corresponding to the available protons in solution, did not have a significant effect on the ESIMS intensity of the analytes. The percent methanol was evaluated with both decreasing and constant electrolyte concentrations to demonstrate the effects of droplet stability and ion transfer into the gas phase. These findings were applied to the analysis of the selected smokeless powder additives using HPLC-ESIMS to illustrate increased sensitivity for the protonated molecules.  相似文献

Cross-lagged panel correlation technique is used to examine whether self-esteem has a greater effect on delinquency than delinquency has on self-esteem. Analysis of a nationwide study of tenth-grade boys shows that self-esteem is the more powerful causal factor, even when initial levels of delinquency are held constant. This result, however, is found to be stronger in the lower class than in the upper class. These data are interpreted as lending greater support to Kaplan's theory of the self-enhancing nature of delinquent behavior than to the idea of reflected appraisals.The research reported in this article was funded by a grant from the National Institute of Mental Health (MH27747), which is gratefully acknowledged. Based on interviews conducted with the Chinese elite, ranging from intellectuals to ex-army officers, this article examines Chinese society between 1978 and 1988, the 1988–89 period of crisis, and China's future prospects. The author analyzes the relative performance of the agricultural, rural-industrial, and heavy industrial sectors as well as the special economic zones over this period of time. China's debt problem, recent splits within the People's Liberation Army (PLA), the Communist Party's loss of legitimacy, and the rise of new interest groups are examined. The author concludes that present socio-economic trends indicate China with return to a program of economic liberation and political decentralization after the dealth of Deng. In this paper we have adapted a technique, previously used to determine the presence of urea in aqueous samples of wine and urine, to detect trace levels of urea nitrate explosives. The procedure involves the reaction of the uronium ion (protonated urea) with a fluorophore, xanthydrol. By modification of the procedure to utilize non-aqueous reagents, in neutral conditions, it can be made specific to the presence of the urea nitrate ion pair. The procedure includes selective detection of derivatization products by UV and fluorescence following separation by High-Performance Liquid Chromatography (HPLC). Analytical method development included optimization of HPLC conditions (solvent, gradient), UV and fluorescence wavelengths, and derivatization parameters (xanthydrol amount, reaction times, temperature). The extraction of urea nitrate from surfaces was also investigated and optimized. For best quantification, it was shown that an internal standard was required; this resulted in a quantification limit around 0.17mM (21mg/L). The entire procedure could be performed in less than 30min per sample and potential interferences such as ammonium, nitrate, and urea did not produce a response under standard conditions.  相似文献

The recent emergence of a new class of piperazine-type compounds has brought about the need for laboratory screening methods for both seized drugs and toxicological samples. These piperazine compounds, which include 1-benzylpiperazine (BZP) and 1-(3-trifluoromethylphenyl)piperazine (TFMPP), exhibit comparable physiological effects and can be substituted for the classic amphetamine-type drugs. We have optimized a chiral capillary electrophoresis (CE) separation that detects a set of 6 piperazine and 4 chiral amphetamine compounds in under 23 min using a 200 mM phosphate buffer at a pH = 2.8 with 20 mM hydroxypropyl- beta-cyclodextrin (HPbeta3CD). In addition to the above compounds, a series of "clandestine" BZP diHCl samples were also analyzed using this method to assess the ruggedness of the procedure. The novel CE separation was tailored to simultaneously detect these piperzine compounds in addition to amphetamine-type drugs. Distinct migration time and UV-spectral data were obtained for all compounds of interest.  相似文献

In capillary gel electrophoresis (CE), changing run conditions such as temperature can result in minor variations in the size determination of an allele. These effects are caused by secondary structure differences that can occur between the amplified sample and the internal standard. The type of method chosen to generate the sizing curve in STR analysis can influence the relationship between estimated allele size and temperature. To better understand the effects of temperature and sizing method on the reproducibility of DNA migration, two fluorescently labeled allelic ladders, CTTv and Y-PLEX 6 were analyzed using the ABI Prism 310 Genetic Analyzer. The default method on the Genetic Analyzer utilizes an electrophoretic temperature of 60 degrees C and a Local Southern method to generate a sizing curve from the fragment migration times of the internal lane standard. In this work, electrophoresis was conducted at 35-70 degrees C using the commercially available POP 4 buffer at pH 8 and two sizing methods, Global Southern and Local Southern, were compared. The slopes of the regression line between estimated allele size and temperature, using either sizing method, were measured in order to demonstrate the temperature sensitivity of migration time and the importance of the operator-chosen method. Our results indicate that the Global Southern method is a better choice in situations where temperature fluctuations can occur. In addition, the temperature dependence of the DNA size estimates using the POP 4 system were compared to results obtained using an experimental buffer consisting of 3% hydroxyethylcellulose at pH 11. These results demonstrate that secondary structure effects are minimized at an elevated pH, increasing the precision of size estimates obtained.  相似文献

Experiments were performed to determine the extent of cross‐contamination of DNA resulting from secondary transfer due to fingerprint brushes used on multiple items of evidence. Analysis of both standard and low copy number (LCN) STR was performed. Two different procedures were used to enhance sensitivity, post‐PCR cleanup and increased cycle number. Under standard STR typing procedures, some additional alleles were produced that were not present in the controls or blanks; however, there was insufficient data to include the contaminant donor as a contributor. Inclusion of the contaminant donor did occur for one sample using post‐PCR cleanup. Detection of the contaminant donor occurred for every replicate of the 31 cycle amplifications; however, using LCN interpretation recommendations for consensus profiles, only one sample would include the contaminant donor. Our results indicate that detection of secondary transfer of DNA can occur through fingerprint brush contamination and is enhanced using LCN‐DNA methods.  相似文献
