We describe a method for obtaining specific and reproducible deoxyribonuclease I (DNase I) typing from liquid semen. Isoelectric focusing of the enzymes on polyacrylamide gel (IEF-PAGE, pH 3.5-5) was accomplished using a 0.5-mm thick gel. The separated isozymes were visualized by a new activity staining method, dried agarose film-overlay (DAFO). Pretreatment of semen samples with neuraminidase markedly enhanced the isozyme-band resolution and sensitivity. The method was simple and reliable, with high resolution and sensitivity. The DNase I types in semen samples were correlated with the types found in corresponding blood and urine samples. DNase I typing could therefore provide an additional discriminant characteristic in the forensic examination of semen. 相似文献

We have developed a new sensitive multiplex PCR system consisting of five male-specific and polymorphic tetranucleotide STRs--DYS441 (GDB: 10013873), DYS442 (GDB: 10030304), DYS443 (GDB: 10807127), DYS444 (GDB: 10807128), and DYS445 (GDB: 10807129) on the Y chromosome. Fifty pg DNA per 10 microL reaction volume was required for the correct typing of five STRs. Using this system, the five Y-STRs were correctly typed from blood and semen stains that had been stored for several years at room temperature. 相似文献

Hara‐kiri is a unique Japanese custom, primarily stemming from the manners and customs that a samurai held. The aim of the present study was to investigate the clinical features of individuals who attempted suicide by hara‐kiri. We enrolled 647 patients who had attempted suicide. Clinical features were compared between those who had employed hara‐kiri and those who had used other methods. 25 of the 647 subjects had attempted suicide by hara‐kiri. The ratio of men to women and the proportion of patients with mood disorders were significantly higher in the hara‐kiri group than in the other methods group. The average length of stay in either the hospital or in the intensive care unit was also longer in the hara‐kiri group than in the other methods group. Hara‐kiri is an original Japanese method of attempting suicide, and suicide attempts by hara‐kiri may be aimed at maintaining a reputation or taking responsibility. 相似文献

AbstractIn the 1970s much of the literature on the role of married Japanese women presented a picture that roles should be clearly demarcated and segregated by gender. According to this ideal a man's main role is to earn a living, whereas a married woman's primary role is to run the household, manage the family finances, and look after the welfare of the family. Fundamental to this gender role segregation was the notion that women had a natural aptitude for matters relating to the home and to the family, and that a woman's most important and rewarding role was that of nurturing her family. This view lies behind the complete interdependence and role complementarity that characterize the Japanese husband-wife relationship. 相似文献

This paper examines the domestic response to the Great East Japan Earthquake that occurred in March 2011. The demand to (re)build a new ‘community’ in Japan was triggered by the fatal destruction of towns by the earthquake and the tsunami, and the uncertain future of these towns – and of Japan – due to the subsequent prolonged crisis at the Fukushima Daiichi nuclear power plant. Drawing on the political theory of Jean-Luc Nancy, the paper examines what kinds of communities were realised in the aftermath of the disaster. It argues that two distinctively different types of communities emerged. The first type of community was based on the inside/outside logic, whereby Japan was imagined as one unified entity surrounded by a boundary which separated the safe inside from the dangerous outside. The second type of community was based on sharing. Immigrants in Japan identified themselves with the survivors since both of them shared the experience of losing home and the same home (called Japan). In this way, the immigrants challenged the idea of community as one unified entity and realised a new form of community. 相似文献

Methodology is presented for the profiling of fentanyl in seized drugs using ultra high performance liquid chromatography in combination with tandem mass spectrometry (UHPLC-MS/MS). Target analysis was performed for 40 fentanyl processing impurities, several of which are markers for a specific synthetic route (Siegfried or Janssen). For the separation of these solutes, an Acquity BEH C18 1.7 μm particle column (150 mm × 2.1 mm) with a binary 1% formic acid (pH 2.0)/acetonitrile gradient was used. For MS/MS detection, an atmospheric pressure positive electrospray source was employed with selected reaction monitoring (SRM). The coupling of the high separation power of UHPLC with the highly selective and sensitive detection of MS/MS is amenable to the determination of synthetic route and linking of drug seizures. The technology is also applicable to exhibits containing trace levels of fentanyl in the presence of significantly excess amounts of heroin and/or adulterants. 相似文献

Abstract. This article presents a definitional and historical summary of 'civil society' as a western concept, and then traces how it was used in Japanese political theory and practice. 'Civil society' discourse became a familiar term amongst post–war Japanese historians and social scientists in the period 1945–1970, and thus preceded the recent international proliferation of 'civil society' literature in the 'west' from the 1970s onwards. 'Civil society' discourse was politicised in practice in Japan in the 1950s and 1960s, principally in opposition movements wanting to encourage political participation by ordinary citizens. While some 'civil society' discourse in Japan was based on an idealisation of 'western' ideas and practice, it is also the case that the individualism and democratisation implied in 'civil society' discourse has been very differently understood in 'western' countries and very unevenly instituted in their political practice. A simple East–West frame makes significant similarities between Japanese ideas and practice and 'western' concepts and politics disappear, and additionally causes significant differences within the 'west' regarding individualism and democratisation to become invisible. 相似文献

A slim and yet powerful novella, Chichi to ran (Breasts and Eggs, 2008) helped the author, Kawakami Mieko, launch her career in the Japanese literary establishment by winning her the Akutagawa prize. The novella revolves around a middle-aged single-mother, struggling to eke out a living for herself and her teenage daughter as a bar hostess in Osaka. The bond between them is severely tested under the pressure of the precarious living conditions in the post-bubble, neo-liberalist Japan of the 2000s. This essay explores the absorbing and affective aspect of the novella by drawing on Rita Felski's ‘positive aesthetics’ and Bruno Latour's concept of ‘nonhuman actors’. With a focus on the movement of affect/feelings, the analysis traces how ‘non-human actors’ of all kinds and shapes in the novella, from the female characters, chatty style of speech in Osaka dialect, the kanji used in them, the protagonist's obsession with breasts, images of Higuchi Ichiyô, to a carton of eggs, interact and connect in a way that make a difference, facilitating innovative life-adjustments as the narrative unfolds – the reading which should, in turn, enhance our understanding and appreciation of the text. 相似文献

The aim of this study was to investigate the relationship between the clinical features of suicide attempts and the Great East Japan earthquake in Kanagawa. We enrolled 592 patients who attempted suicide and were hospitalized for inpatient treatment. Clinical features were compared between before the earthquake (A) and after the earthquake (B) groups. The number of suicide attempts increased from 286 to 306. The rate of increase in suicide attempts in men was the highest in the age group of 40–59 years. The frequency of joblessness, family psychiatric histories, precipitating event, and alcohol intake at suicide attempt was significantly higher in the B group than in the A group. The number of patients in serious condition in the B group was significantly higher than that in the A group. Other clinical features related to suicide in people who were indirectly affected by the earthquake disaster should be investigated to develop targeted prevention strategies. 相似文献
