A new individualization marker of semen: deoxyribonuclease I (DNase I) polymorphism.

We describe a method for obtaining specific and reproducible deoxyribonuclease I (DNase I) typing from liquid semen. Isoelectric focusing of the enzymes on polyacrylamide gel (IEF-PAGE, pH 3.5-5) was accomplished using a 0.5-mm thick gel. The separated isozymes were visualized by a new activity staining method, dried agarose film-overlay (DAFO). Pretreatment of semen samples with neuraminidase markedly enhanced the isozyme-band resolution and sensitivity. The method was simple and reliable, with high resolution and sensitivity. The DNase I types in semen samples were correlated with the types found in corresponding blood and urine samples. DNase I typing could therefore provide an additional discriminant characteristic in the forensic examination of semen.     

A novel multiplex PCR system consisting of Y-STRs DYS441, DYS442, DYS443, DYS444, and DYS445

We have developed a new sensitive multiplex PCR system consisting of five male-specific and polymorphic tetranucleotide STRs--DYS441 (GDB: 10013873), DYS442 (GDB: 10030304), DYS443 (GDB: 10807127), DYS444 (GDB: 10807128), and DYS445 (GDB: 10807129) on the Y chromosome. Fifty pg DNA per 10 microL reaction volume was required for the correct typing of five STRs. Using this system, the five Y-STRs were correctly typed from blood and semen stains that had been stored for several years at room temperature.     

Transferrin polymorphism detected in human urine using isoelectric focusing followed by immunoblotting

Genetic polymorphism of transferrin (TF) was revealed in human urine by isoelectric focusing and immunoblotting on thin-layer polyacrylamide gels. Using this technique more than 300 urine samples were examined, and correct TF typing from a small volume of urine (approx. 0.5 ml) was achieved, in comparison with the results of direct grouping for plasma. Three common phenotypes, TF C1, C2-1 and C2, were differentiated. In addition, the rare types TF C1D, C2D, and C1B were observed. The frequencies of the TF alleles in our samples were found to be: TF*C1 = 0.7265, TF*C2 = 0.2624, TF*D = 0.0083 and TF*B = 0.0028.     

Regulatory capitalism and its discontents: Bilateral interdependence and the adaptability of regulatory styles

The expansion of global trade has produced new challenges for the effective governance of product safety. We argue that many of these challenges arise at the bilateral level from the interaction of more or less adaptable national regulatory styles. When regulatory styles are unadaptable they produce gaps in risk management, slow and contested resolutions to crises, and limited regulatory cooperation. To examine these claims empirically, we study bilateral food safety regulation in four major exporter–importer dyads: China–Japan; Canada–United States (US), China–European Union (EU), and the US–Japan. The China–Japan dyad is the most adaptable, combining China's “export segmentation” regulatory style with Japan's strongly “risk‐averse, interventionist” style. The Canada–US dyad operates effectively, bringing together Canada's “global market–conforming” regulatory style with the US strategy of “sovereign regulator.” The China–EU dyad is less adaptable because the EU's “harmonization” regulatory style makes it difficult for the EU to adapt to the weaknesses of the Chinese food safety system. Finally, the US's sovereign regulator style clashes with Japan's interventionist style, making them the least adaptable of the four dyads. The paper concludes with a discussion of the broader relevance of our findings for the development of regulatory capitalism.     

Strategic Patience in a Turbulent World: The Obama Doctrine and its Approach to the World

The Obama administration is often criticized for not having a strategic vision. After more than six years in office, a high expectation that was widely shared around the globe seems to have nearly evaporated. Many see the “disorder” the world is facing today as partly a result of a lack of vision from the White House. You could make a case that the emergence of the Islamic State (IS) and other “violent extremism” in the Middle East and beyond, Russia's aggression in Ukraine, chaos in Syria, China's assertive moves in the East China Sea and South China Sea, and so forth, are results, direct and indirect, of a perceived vacuum created by “American withdrawal.” However, the Obama administration's understanding of the world has been surprisingly positive and constant. This paper will argue that Obama administration in fact has a core world view, based on a new self-image of the United States, and that the issue is not whether the administration lacks a vision; rather the focus should be on the vision itself and the execution of that vision.     

Accuracy and Usefulness of the AVOXimeter 4000 as Routine Analysis of Carboxyhemoglobin

The measurement of blood carboxyhemoglobin (CO‐Hb) is important to determine the cause of death. The AVOXimeter 4000 (AVOX), a portable CO‐oximeter, has the advantages of a low purchase price and operating cost, ease of operation, and rapid results. Little information is available on the usefulness of AVOX in the forensic sample, and the previous study investigated only six samples. Therefore, in this study, we confirmed the usefulness of the AVOX through a comparison of its re s ults with data previously obtained using the double wavelength spectrophotometric method in autopsies. Regression analysis was performed between CO‐Hb levels measured by the AVOX and those measured by the conventional double wavelength spectrophotometric method in postmortem blood samples: a significant correlation was observed. This study suggests the usefulness of the AVOX to analyze postmortem blood, and the AVOX is suitable for routine forensic analysis and can be applied at the crime scene.
