The microscopical examination of original surfaces on casework sized glass particles using an interference objective and the interpretation of their interferograms is discussed. To reduce subjectivity, a numerical criterion was devised to assess the curvature of glass particles and this was then applied to a survey of window, container, tableware and vehicle glasses. Objects with curved surfaces produced particles that were distinctly curved while those from surfaces of both modern and old windows were clearly identifiable as flat. Apparently flat surfaces from tableware and containers almost always yielded specimens that were detectably curved. In addition, there was no evidence that a fracture surface would be confused with an original surface from a plain flat glass.In a ‘blind’ trial to assess the interferometric routines, three examiners correctly identified flat and curved surfaces on 15 particles derived from a selection of glass objects.  相似文献

目的 比较新疆与甘肃地区甘草的性状、鉴别特征和有效成分含量,研究在不同生长条件下我国甘草主产区的药材和饮片质量,为甘草的后续开发与利用提供理论依据。方法 采用性状鉴别法对性状进行描述,采用显微鉴别法展现甘草粉末和切片的结构特征,采用高效液相色谱法测定有效成分含量。结果 在性状方面,新疆甘草药材比甘肃甘草药材平均直径略大;在显微方面,两个产地具有基本相同的结构,但新疆家种甘草的淀粉粒数量略多,且平均粒径也略大;在含量方面,新疆甘草中甘草酸平均含量约为甘肃甘草酸平均含量的1.5倍。结论 新疆、甘肃地区甘草的鉴别和质量评价为甘草药材的质量标准研究及甘草药材的后续开发提供理论依据。  相似文献

4种硅藻检验方法的比较研究   总被引:3,自引:2,他引:1  
目的比较硝酸乙醇法、酶消化法、“破机罐”法和Soluene-350法在检验硅藻中各自的优缺点,为法医实际办案提供理论依据。方法取家兔肝、肾和骨髓3种组织各2g,分别用上述4种方法消化,比较4种不同方法分别将组织这些完全消化所需要的时间、消化能力、对硅藻的检出率以及对硅藻的破坏程度。结果消化时间以Solu-ene-350法耗时最长,其次是酶消化法,硝酸乙醇法和破机罐法耗时最短。4种方法消化能力的强弱依次是“破机罐”法、硝酸乙醇法、酶消化法、Soluene-350法。对梅尼小环藻(淡水)和桥弯藻(淡水),酶消化法的检出率最高,硝酸乙醇法其次,“破机罐”法和Soluene-350法最低。对舟形藻(海水),酶消化法的检出率高,其余3种方法几乎没检测到。扫描电镜观察:酶消化法对硅藻结构几乎没有破坏。其余3种方法对硅藻均有不同程度的破坏。结论从降低成本、提高检测效率角度,应选择“破机罐”法和硝酸乙醇法;从安全性和环保角度,或可疑水样为海水及硅藻密度较小时,应选择酶消化法;Soluene-350法因耗时长、成本高且硅藻检出率低,不适于在基层法医中推广。  相似文献

Interference filters were investigated as alternatives to sodium vapour lamps to establish their suitability for making refractive index measurements using a hot stage microscope. Full DNA profiles can be generated from just a few cells; however these profiles can be contaminated from other cell types present at the crime scene. We report here on the development of an immunofluorescent technique to spatially locate human-specific blood in situ and also on the ability of this technique to detect individual leukocytes and the DNA contained within them. Four monoclonal mouse anti-human antibodies were evaluated; anti-glycophorin A to detect erythrocytes and anti-CD45, anti-myeloperoxidase (MPO) and anti-histone H1 to detect the nucleated leukocytes. Each antibody was labeled with either Alexa Fluor 488 or 568 for direct application to blood smears which allowed the simultaneous detection of erythrocytes and leukocytes. Furthermore, because histones are DNA binding proteins, the application of anti-histone H1 allowed the detection of DNA within a blood smear. Importantly it was found that full DNA profiles could be achieved after using this method with similar peak area ratios compared to untreated cells. The fluorescent antibodies were found to be human-specific with the exception of anti-histone H1 due to its conserved sequence. However, used in combination with anti-CD45 or anti-MPO the location of DNA from human-specific leukocytes could be detected. The technique was also tested on older blood stains and was still found to be sensitive and cell-specific after 4 months. Following the optimization of the methodology, the fluorescent antibodies were applied to short lengths of black cotton fibres covered with human blood spots. Although the background fluorescence from the cotton was found to be high, erythrocytes and even individual leukocytes could easily be detected, indicating that this technique could be used to detect extremely minute amounts of blood. Used in combination with laser capture microdissection (LCM), this method could be used to pick off individual leukocytes for LCN DNA techniques.  相似文献

In the second part of this survey, the ability of micro‐Raman spectroscopy to discriminate 180 fiber samples of blue, black, and red cottons, wools, and acrylics was compared to that gathered with the traditional methods for the examination of textile fibers in a forensic context (including light microscopy methods, UV‐vis microspectrophotometry and thin‐layer chromatography). This study shows that the Raman technique plays a complementary and useful role to obtain further discriminations after the application of light microscopy methods and UV‐vis microspectrophotometry and assure the nondestructive nature of the analytical sequence. These additional discriminations were observed despite the lower discriminating powers of Raman data considered individually, compared to those of light microscopy and UV‐vis MSP. This study also confirms that an instrument equipped with several laser lines is necessary for an efficient use as applied to the examination of textile fibers in a forensic setting.  相似文献
