Turkey and Russia in a shifting global order: cooperation,conflict and asymmetric interdependence in a turbulent region

The current global political economy is characterised by the intensifying economic interaction of BRICS and ‘near BRICS’ economies, with emerging powers increasing their influence in neighbouring regions. The growing partnership between Turkey and Russia constitutes a useful case study for examining this transformation, in which Western supremacy and US hegemony are under increasing challenge. Turkish–Russian relations shed light on broader themes in global political economy. First, significant economic interdependence may be generated among states with different political outlooks, in the form of loose regional integration schemes driven by bilateral relations between key states and supporting private actors or interests. Second, growing economic interdependence may coexist with continued political conflict and geopolitical rivalry, as indicated by the Syrian and Ukrainian crises. An important strategy that emerges is the tendency to compartmentalise economic issues and geopolitical rivalries in order to avoid negative spill-over effects. This facilitates the coexistence of extensive competition with deepening cooperation, as reflected in relations in the field of energy.     

An ambiguous sequence-based allele of SE33

SE33 was a well-known autosomal short tandem repeat (STR) marker that was high polymorphic and therefore was high discrimination power. The sequence structure of STR markers has been increasingly explored with next-generation sequencing (NGS) technology. The sequencing resulted in the development of a new locus designation and allele nomenclature that was also backward compatible with the conventional capillary electrophoresis. SE33 was one of the STR markers that had been coamplified by Forenseq™ Signature Prep Kit (Verogen) but were not analyzed and illustrated in the Universal Analysis Software (UAS) (Verogen). This study reported an ambiguous sequence-based allele 16.3 of the SE33 locus. This allele was observed while analyzed by STRait Razor 3.0. The configuration file was modified from the previous studies to include 15 bp of 5′ flanking region and 24 bp of 3′ flanking region. The ambiguous allele was called 16.3 (106 bp) with a read count of 2070. However, the sequence of the repeat region cannot be designated as allele 16.3. Several possible scenarios for allele designation were presented and discussed.     

Soil sample metagenome NGS data management for forensic investigation

Soil analysis is a valuable resource in forensic investigation. Classical forensic soil analysis involves examination of its physical characteristics and chemical composition, such as soil type, colour, particle size, shape, pH, elemental, mineral and organic content. However the limited variability of these parameters is not always allowing adequate discrimination between soil samples. As soil supports extreme diversity of microorganisms and eukaryotic communities, microbiological approaches have been proposed. Several molecular approaches for microbial DNA profiling are available; however there is a lack of published data of implementation of the next generation sequencing (NGS) approaches for forensic soil analysis.The aim of the current study was elaboration of criteria for soil metagenome data management and database searching. We used our previously sequenced collection of 11 samples collected from different environments (forests, fields, grasslands, urban park) with different flora. The single sample collection includes 9 soil samples per one sampling area (30 m × 30 m) spaced by 15 m. In the current study we concentrated mainly on 18S rRNA gene V2-V3 region for fungi however SSU rRNA region for arbuscular mycorrhizal (AMF) fungi and V2-V3 hypervariable region of 16S rRNA gene for bacterial communities were taken into account. The sequencing was performed by Roche/454 platform. For data analysis OTU based approach on mothur software and NCBI BLASTN search were used. NCBI BLASTN analysis revealed altogether 2983 AMF matches and 8997 18S matches as well as 25477 OTUs (16S) were determined. Several data filtration approaches were used for data management. We found that 18S marker results could be used to create and run a filtered database that is computationally much more efficient and flexible. Our results have broad impact; however more samples have to be analysed, additional studies performed and cooperation between soil scientists and forensic scientists is required to be able to implement these novel techniques into the routine forensic practice.     

Digital Britain – the final report

The final Digital Britain report was published in June as to set out the Government's final proposals and updating an interim version that has been published in January this year. This article builds upon an earlier article in [2009] 25 CLSR 263, which analysed the interim report in depth. The intention is not to repeat that earlier analysis, but rather update it and draw the reader's attention to those key areas of change.

The return of public investment in telecommunications: Assessing the early challenges of the national broadband network policy in Australia

Against a current trend of investing in the next generation networks (NGNs) by using public funds, the Australian government has initiated a so-called National Broadband Network (NBN) project to invest up to AUD$36 billion tax payer's money on building a national wide fibre broadband network aiming to cover 93 per cent Australian by 2020. As being the most costly infrastructure-building project in Australian history, the NBN project promises to deliver super-fast broadband services, create jobs and promote the country's economy at large. This article will critically analyse the NBN project in Australia and highlight the challenges that are coming alone at this early stage of the deployment, so the Australia's experience of pubic investment in broadband networks can be shared and lessons can be learnt.

Bad Piano? Bad Music? A Response to Comments on Lowe and Pemberton,The Official History of the British Civil Service,Volume II: 1982–1997

The reforms made to the civil service during the premierships of Margaret Thatcher and John Major were unparalleled in scope in peacetime. Undertaken in the name of efficiency and better management, they served to Balkanise the service, shrink the number of civil servants, subordinate them to the will of ministers, and effectively privatise a swathe of public services. Their legacy, however, was a relatively weak centre struggling to cope with a fragmented and extremely complex governmental machine, an overly managerialist senior civil service sometimes side‐lined from policy making, and a system lacking slack as a consequence of the quest for ‘efficiency’ and dependent on private contractors of sometimes dubious worth. Over the past five years that legacy left the country poorly placed to triumph over the challenges of Brexit and then of Covid‐19.     

Association between copy number variations in the OCA2-HERC2 locus and human eye colour

Human eye colour variation is strongly associated with single nucleotide polymorphisms (SNPs) in the OCA2-HERC2 locus, especially rs12913832 that is found in an enhancer element of OCA2. In a previous study we found that 43 out of 166 individuals in a Norwegian population with the brown eye colour genotype HERC2 rs12913832:AA or AG, did not have the expected brown eye colour. To investigate if duplications or deletions in the OCA2-HERC2 locus could explain the blue eye colour in these individuals, we analysed massively parallel sequencing (MPS) data for copy number variations (CNVs) in the OCA2-HERC2 region. The ∼500 kb long OCA2-HERC2 locus was sequenced in 94 individuals with the rs12913832:AG and AA genotypes. Of these, 43 were observed to have blue eye colour and 51 were observed to have brown eye colour. CNVs were analysed using R and the R-package panelcn.MOPS - CNV detection tool for targeted NGS panel data. In rs12913832:AG individuals, CNVs in 32 regions were significantly associated with blue eye colour (Benjamini-Hochberg adjusted p-value ≤ 0.05). In rs12913832:AA individuals, CNVs in 14 regions were associated with blue eye colour using raw p-values (p ≤ 0.05). The functional effects of these CNVs on OCA2 expression are yet to be investigated. However, this study suggests that CNVs in the OCA2-HERC2 locus might explain why some of the rs12913832:AG and AA individuals have unexpectedly blue eyes.
