A novel method for cyanide quantification in human whole blood using ion chromatography with amperometric detection and its application to cyanide intoxication cases

Cyanide is a highly toxic agent that has been frequently used for suicide in South Korea. It is also used in various industrial fields, such as metal plating, in which many accidental cyanide intoxications have occurred. To overcome the disadvantages of conventional cyanide analysis methods, a simple and fast method for the analysis of cyanide in whole blood using ion chromatography (IC) with amperometric detection was developed in this study. Whole blood samples were deproteinized, diluted, and analyzed using an IC–amperometric detection system. The limits of detection and quantitation were 0.1 and 0.2 mg/L, respectively. The method showed good linearity in the range of 0.2 to 50 mg/L with R² > 0.99. The intra- and inter-assay precision and accuracy values were <10%. The established method was successfully applied to analyze whole blood samples from three cyanide intoxication cases.     

中华鳖全血呼吸爆发化学发光检测方法的建立

为进一步研究中华鳖的先天免疫能力及其机理,以酵母多糖为吞噬底物,通过对鲁米诺浓度、全血稀释度和检测温度等条件的优化,建立了中华鳖全血呼吸爆发的化学发光检测方法,并用此方法对放置不同时间的全血和稀释血液进行了呼吸爆发检测效果的比较。结果表明,鲁米诺浓度为1×10-4mol/L,检测温度为31.5℃时,以0.4~0.8μL/mL的中华鳖全血进行检测可获得理想效果。保存6 d的全血仍可检测到呼吸爆发,稀释全血放置30 h后不再产生呼吸爆发。     

电针对血管性痴呆大鼠学习记忆能力及脑血流量的影响

目的：观察电针对血管性痴呆(vascular dementia，VD）模型大鼠学习记忆能力和局部服血流量（regional cerebral blood flow,rCBF )的影响。方法：用双侧颈总动脉反复缺血－再灌注结合腹腔注射硝普钠法，并配合行为学测试确定模型。术后7d电针百会、大椎、足三里（双）治疗。用跳台法评价各组大鼠不同时期的学习记忆能力，治疗后测定各组大鼠顶叶有海马rCBF。结果：VD大鼠在电针治疗后的行为学测试成绩明显提高（P＜0．01），其低下的大脑顶叶及海马rCBF水平明显改善（P＜0．01）。结论：电针对VD大鼠的学习记忆能力有显的改善作用；电针能促使机体低灌流的缺血服组织灌流水平恢复，可能有助于与学习记忆相关的传导环路的修复。     

血液样品中乙醇稳定性的实验研究

目的考察在不同存放条件下含乙醇的血液样品中乙醇浓度的变化情况。方法采用顶空-气相色谱法,以异丙醇为内标,对存放条件不同的血液样品中乙醇进行检测。结果冷冻(-10℃)条件存放1至30天,血液样品中乙醇含量无显著变化;冷藏(4℃)条件存放1至30天,血液样品中乙醇含量变化不显著;室温(28℃~33℃)条件存放1至30天,血液样品中乙醇含量显著改变。结论含有乙醇的血液样品,在冷冻、冷藏条件下可较稳定的存放30天;在室温条件下存放30天血液样品中乙醇浓度发生显著变化,不可在此条件下存放。     

复合扩增mtDNA-HVI和Cyt b片段鉴定混合血痕种属

目的探讨mtDNA-HVI和Cyt b片段复合扩增法鉴定人与动物混合血痕种属的应用价值。方法用chelex-100法从人、牛、猪、狗、兔、鱼、鸡和鼠血痕中提取DNA,复合扩增mtDNA-HVI片段和Cyt b片段,琼脂糖凝胶电泳检测。结果人类在mtDNA-HVI区和Cyt b区分别出现279bp和358bp各一条带,且279bp条带亮于358bp;动物均只有358bp一条带。人与7种动物血痕的检测灵敏度均为3.13ng。检测人与动物混合DNA,灵敏度仍为3.13ng,但358bp条带亮于279bp条带。结论当358bp带明显强于279bp带时,提示检材为人与动物的混合。

The Contribution of Donor Support Programs to Decentralized Development: Learning from the District Level in Ghana

Donor aid has over the years turned out to be a very decisive development component for most developing nations, but aid is often criticized for ineffectiveness. This article seeks to share perspectives on the relevance of direct project or program support and the need to refrain from homogeneity in aid research by assessing the contribution of the Government of Ghana/European Union (GoG/EU) Micro-Projects Programme in the Nadowli District of Ghana from 1997–2009. The findings acknowledge positive impacts of the program in the district. There are sector consistencies between the donor interest and local development priorities in health, education and the water and sanitation sectors. However, selectivity of project within and among sectors brings potential conflicts of interest and tradeoffs between the program and local community needs. The results imply a reexamination of decentralized development through a lens of new partnership between donors’ interest and local needs. Stepping up community awareness to ensure full participation in project design, execution, operation, and maintenance could serve as a start-up platform for new partnerships. It is imperative to align donor interest with local realities, which prompts a rethinking of adoption of new project approaches such as co-development, design, and implementation.

The Stability of 4-Chloromethcathinone in Blood and Vitreous Humor

We present results of our study on the stability of 4-chloromethcathinone (4-CMC) in authentic postmortem peripheral blood and vitreous humor samples. The stability of 4-CMC was determined in postmortem blood samples (for a period of 90 days) and vitreous humor (30 days) at three different temperatures: −15°C, +4°C, and + 23°C. The analyses were carried out using ultra-high-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (UHPLC-QqQ-MS/MS). In both materials, the lowest 4-CMC stability was demonstrated at room temperature. The blood samples stored in a freezer (−15°C) showed stability for the entire study period (90 days), while in the case of the vitreous humor sample stored at the same temperature the concentration of the substance decreased by 53% after 30 days. The study carried out in authentic postmortem blood and vitreous humor samples confirms the previous reports of 4-CMC instability in biological material. Authors suggest that the biological material should be stored frozen until analyses are carried out as soon as possible after collection of the material.

Strenuous efforts by donors and lenders over four decades turned Mozambique from a socialist success story into a neoliberal capitalist one. The private sector dominates; a domestic elite dependent on foreign companies has been created. But a secret US$2.2 billion arms and fishing boat deal involving Swiss and Russian banks and Mozambican purchases from France, Germany, and Israel, with large profits on all sides, was a step too far down the donor’s capitalist road. The International Monetary Fund cut off its programme and western donors ended budget support.
