绵羊肺炎支原体免疫蛋白质组学的初步研究

摘    要:利用双向电泳及免疫印迹方法对绵羊肺炎支原体MoGH3-3分离株的抗原蛋白进行筛选,并采用基质辅助激光解吸离子化飞行时间质谱对其进行初步鉴定。结果,共筛选到MoGH3-3株3个主要抗原蛋白点,其中2个被鉴定为延伸因子(elongation factor Tu,EF-Tu),1个为丙酮酸脱氢酶E1-a亚单位(pyruvate dehydrogenase E1-alpha subunit,PDHA)。结果表明,EF-Tu及PDHA是绵羊肺炎支原体主要的抗原蛋白,这为下一步绵羊支原体肺炎基因工程疫苗的研制提供了靶标。

Abstract:The proteins of MoGH3-3 strain of Mycoplasma ovipneumoniae(Mo) were extracted and separated by two-dimensional electrophoresis(2-DE),then subjected to immunoblotting with antiserum of Mo.The screened major immunoreactive proteins were identified by Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry(MALDI-TOF-MS)and analyzed with bioinformatics methods. Three major immnunoreactive protein plots were screened with two of them identified as the elongation factor Tu(EF-Tu) and the rest one as the pyruvate dehydrogenase E1-alpha subunit(PDHA). The results showed that EF-Tu and PDHA are the major immnunogenic proteins of Mo. This study provided antigen candidates for the development of novel vaccines against disease caused by Mo.
