摘 要: | 目的 探究桃红四物汤(Taohong Siwu Decoction, THSWD)干预后的血小板外泌体(platelet-derived exosomes, PLT-Exos)对脑缺血再灌注大鼠的血管新生作用。方法 采用超高速差速离心法提取PLT-Exos,透射电子显微镜观察外泌体形态,纳米微粒示踪分析检测外泌体粒径分布,蛋白质免疫印迹法检测外泌体标志蛋白表达水平;建立大鼠脑缺血再灌注模型,将所提取的经THSWD干预后的PLT-Exos通过尾静脉注射到模型大鼠体内,再通过脑血流量测定、苏木精-伊红染色、免疫荧光染色,验证THSWD-PLT-Exos对大鼠脑缺血再灌注模型血管新生的治疗作用。结果 提取的PLT-Exos表征符合典型。THSWD-PLT-Exos可显著提升脑缺血大鼠脑部的血流量,改善海马区损伤,具有良好的血管生成和神经保护作用。结论 THSWD-PLT-Exos对脑缺血再灌注大鼠损伤有良好的血管新生作用。

Abstract: | Objective To investigate the effect of platelet-derived exosomes (PLT-Exos) after Taohong Siwu Decoction (THSWD) intervention on a rat model of cerebral ischemia/reperfusion. Methods PLT-Exos were extracted by ultra-high speed differential centrifugation; a transmission electron microscope was used to observe the morphology of exosomes; nanoparticle tracing was used to observe the particle size distribution of exosomes; Western blot was used to measure the expression of exosome biomarkers. A rat model of cerebral ischemia/reperfusion was established, and PLT-Exos extracted after THSWD intervention (THSWD-PLT-Exos) were injected into the model rats via the caudal vein; cerebral blood flow measurement, HE staining, and immunofluorescent staining were used to verify the therapeutic effect of THSWD-PLT-Exos on angiogenesis in a rat model of cerebral ischemia/reperfusion. Results The PLT-Exos extracted had typical characteristics. THSWD-PLT-Exos significantly increased cerebral blood flow, improved hippocampal injury, and had good angiogenesis and neuroprotective effects. Conclusion THSWD-PLT-Exos has a good angiogenesis effect on a rat model of cerebral ischemia/reperfusion injury. |
