沂蒙黑山羊发情周期不同阶段卵巢中FSHR和LHR基因表达规律的研究

摘    要:选取健康未孕的成年沂蒙黑山羊,按发情周期(间情期、发情前期、发情期、发情后期)宰杀取卵巢,采用实时荧光定量差异显示PCR(real-time qRT-PCR)技术,以GAPDH为持家基因,对处于发情周期不同阶段的沂蒙黑山羊卵巢组织内的卵泡刺激素受体(FSHR)、黄体生成素受体(LHR)基因进行了mRNA水平上的相对定量分析。发现FSHR mRNA、LHR mRNA在黑山羊发情周期的卵巢中都有表达。FSHRmRNA的相对表达量依次为0.618 7±0.294 42、1.290 2±0.290 24、2.575 9±0.458 80、0.790 8±0.261 93,其中发情期最高,间情期最低,两者差异极显著(P<0.01)。LHR mRNA的相对表达量依次为2.284 0±0.146 22、0.508 9±0.0789 9、0.340 3±0.045 38、2.859 2±0.205 66,其中发情后期相对表达量最高,发情期最低,两者差异极显著(P<0.01)。结果表明,FSH、LH在黑山羊的整个发情周期中是相互作用、相互影响的。
