摘    要:目的:观察中药复方消核冲剂对实验性乳腺增生大鼠血清性激素水平和增生乳腺组织中增殖细胞核抗原(PCNA)表达的影响,探讨该药物治疗乳腺增生病的可能机制.方法:Wistar雌性大鼠60只,随机分为空白对照组(对照组)、模型对照组(模型组)、消核冲剂组(中药组)、他莫昔芬组(西药组),每组15只.肌注苯甲酸雌二醇(E2)、黄体酮复制大鼠乳腺增生模型.模型复制后灌服药物,化学发光免疫法测定血清性激素水平,免疫组化法观察乳腺组织PCNA的表达.结果:中药组和西药组血清E2、孕酮(P)水平均低于模型组,差异有显著性;PCNA阳性细胞数目和阳性细胞面积中药组和西药组均明显减少.中药组各项指标的改变与西药组相比均无明显差异.结论:中药复方消核冲剂对乳腺增生大鼠血清性激素水平具有调节作用,同时也能够下调PCNA阳性表达.

Abstract:Objective:To explore the therapeutic mechanism of Xiaohe granules in treating hyperplasia of mammary glands, and observe the effect of Xiaohe granules on the level of serum hormone and on the expression level of proliferating cell nuclear antigen (PCNA) in hyperplastic mammary glands of rats. Methods: Sixty female Wister rats were randomly divided into four groups: Control group, Model group, Xiaohe group and Tamoxifen group. The control group was injected with normal saline, while the other three groups were injected with both estradiol and progesterone to induce hyperplasia of mammary glands. The sex hormones such as estradiol(E2), pregnendione(P) and prolactin(PRL) were determined by immunochemiluminescent. The expression level of PCNA in mammary cells was measured by immunohistochemistry. Results: The levels of E2 and P in Xiaohe group and Tamoxifen group were lower than those in model group (P<0.05). Compared with those in control group, the number and total area of PCNA positive cells in model group,Xiaohe group and Tamoxifen group decreased significantly. There was no significant difference in above-mentioned indexes between Xiaohe group and Tamoxifen group. Conclusion: Xiaohe granules could regulate the level of sex hormones, and decrease the expression level of PCNA in rats with hyperplasia of mammary gland.
