冬凌草甲素诱导多药耐药细胞系K562／A02凋亡,逆转耐药性？…

目的:探讨冬凌草甲素诱导多药耐药细胞系K562/A02凋亡、逆转耐药性的作用.方法:以不同浓度的冬凌草甲素处理K562/A02及敏感株K562/S细胞,流式细胞仪检测细胞凋亡率,四甲基偶氮唑盐(MTT)比色法检测半数抑制率(IC50),分析冬凌草甲素对两种细胞柔红霉素(DNR)、高三尖杉酯碱(HHT)敏感性的影响,流式细胞仪检测冬凌草甲素对两种细胞内DNR浓度的影响.结果:冬凌草甲素可诱导两种细胞凋亡,两者在各浓度、各时点无显著差异;可显著逆转K562/A02细胞对DNR、HHT的耐药性,提高该细胞内DNR的浓度.结论:冬凌草甲素对K562/A02细胞有诱导凋亡、逆转耐药性的作用,是一种很有希望的抗白血病中药.

EFFECTSOF ORIDONIN ON INDUCING THE MULTI-DRUG RESISTANT CELL LINE K562/A02 TO APOPTOSIS ANDREVERSING THEIR MULTI-DRUG RESISTANCE

Objective:To study the effects of oridonin on inducing the multi drug resistant cell line K562/A02 to apoptosis and reversing their multi drug resistance.Method:K562/A02 and sensitive K562/S cells were treated with oridonin on a variety of concentration.The apoptosis rates of the two cell lines were detected by flow cytometry,IC 50 values of the two chemotherapy drugs (DNR and HHT) were analyzed by MTT assay to analyze the reversal effects;DNR concentration in the cells were detected by flow cytometry.Result:Oridonin can induce the two kinds of cells to apoptosis and the apoptosis rates of the two cell lines were not significantly different,the apoptosis rates were increased with the increasing of dose or time,IC 50 values of DNR and HHT of K562/A02 cells were reduced significantly,and DNR concentration in K562/A02 cells was upregulated.As to K562/S cells,however,this compound couldn''t affect them.Conclusion:In K562/A02 cells,oridonin can induce cell apoptosis,reverse multi drug resistance,so this compound is a kind of very promising anti leukemia traditional chinese drug.
