摘    要:目的 建立人脑脊液中利多卡因的高效液相色谱(HPLC)分析方法。方法 以空白人脑脊液添加标准利多卡因进行HPLC分析,并系统考察实验中的色谱条件、样品处理方法、线性关系、精密度及回收率。结果 所建方法的线性范围是1.0~20.0μg·ml-1(r=0.9993),最低检测浓度为1.0μg·ml-1(S/N≥3),日内、日间检测的相对标准偏差≤3.0%,回收率为98.3%~102.7%。结论 所建方法定量检测人脑脊液中的利多卡,灵敏、准确、快速。

Abstract:Objective For the purpose of forensic toxicological analysis, a RPHPLC method was established for determining lidocaine in human spinal fluid. Method Analysis column was YWG-C18 (φ4.6 X 150mm). Mobile phase was 0.05 mol·1-1 NaH2PO4 :CH3CN = 9:1 (v/v) and wavelength of detection was 210 nm. The method of sample treatment, detection conditions, linear range, precision and recovery of the method were comparatively investigated by using blank spinal fluid spiked with standard lidocaine. Results The linear range was 1.0 - 20. 0μg· ml-1 (r = 0.9993). The limit of detection was 1.0μg· ml-1 (S/N>=3) in spinal fluid. The intra and inter-day relative standard deviation of analysis were less than 3.0 % (n = 5). The recovery of lidocaine was between 98.3 % - 102.7 % . Lidocaine assay was carried out in a medical case by using the method established. Conclusion Spinal fluid is suitable for assay lidocain in forensic toxicological analysis and other medical studies by using the HPLC method which is sensitive, rapid and accurate.
