伊维菌素对内毒血症小鼠生存率和细胞因子分泌的影响

摘    要:为研究伊维菌素对脂多糖所致内毒血症小鼠的保护率及对小鼠血清中细胞因子的影响,给C57BL/6小鼠灌服伊维菌素(1、2和4mg/kg)或等体积蒸馏水,2h后分别腹腔注射脂多糖32mg/kg或等体积PBS,观察7d内小鼠的死亡情况;采用夹心-ELISA法测定注射脂多糖后不同时间点小鼠血清中的TNF-α、IL-1β和IL-6水平,硝酸还原法测定小鼠血清中NO的水平。结果显示,伊维菌素能显著(P<0.05)提高脂多糖诱导的内毒血症小鼠生存率,降低小鼠血清中的TNF-α、IL-1β、IL-6和NO水平。表明,伊维菌素可通过抑制脂多糖诱导的小鼠体内炎性因子和介质释放提高内毒血症小鼠的生存率。

Abstract:To study the effects of ivermectin on survival rate and serum cytokine secretions in lipopolysaccharide(LPS)-induced endotoxemic mice,C57BL/6 mice were given intraperitoneally with LPS at a lethal dose of 32 mg/kg,and ivermectin of different doses(1,2 and 4 mg/kg) were pre-treated with an orallyadministration 2 h before LPS challenge.Survival of mice was monitored for 7 days twice a day following administration of LPS.The levels of tumour necrosis factor-α(TNF-α),interleukin-1β(IL-1β) and interleukin-6(IL-6) in serum of the mice were assessed by sandwich ELISA method,and nitric oxide(NO) levels were measured by nitric acid reduction method at different time following LPS administration.The results showed that ivermectin significantly increased the survival rate in LPS challenged mice,decreased serum levels of TNF-α,IL-1β,IL-6 and NO after LPS challenge.These observations indicated that ivermectin can improve survival rate of endotoxemia mice induced by LPS,which may be mediated by the inhibition of inflammatory cytokine and mediator production.
