| 摘 要： | 采用三步反应合成了己烯雌酚人工抗原 ,为己烯雌酚酶免疫检测技术的进一步研究奠定基础。试验结果证实 ,己烯雌酚及其衍生物存在着顺反异构转化现象 ,酸碱处理是降低交联前衍生物中顺式体比例的有效方法 ;经三步合成 ,成功地制备出了人工抗原 ;人工免疫原及包被原的最适结合比例分别为 1∶2 8和 1∶7。

| Abstract: | Antigenic conjugates and coating conjugates were synthesized by the chemical approach in which one phonetic group of DES was covalently bonded through an ether linkage to the free amine of BSA (or OVA). The synthesis was completed by three steps. First, DES mono ethylbutyrate ether (DES MEBE) was synthesized from DES and ethyl 4 bromobytyate and prepared by TLC. Secondly, DES MEBE was hydrolyzed by 20% KOH methnol to obtain DES mono carboxyproxyl ether (DES MCPE). Thirdly the conjugates of DES MCPE with protein were prepared by the mixed anhydride method. Analytical techniques such as TLC, HPLC, MS and H 1 NMR were inducted to screen reactions and identification of the target compounds. |
