摘    要:以5株同一血清型的鸭沙门菌为研究对象,分别提取基因组DNA后,用20条随机引物对其进行RAPD分析。结果,20条随机引物中不能扩出任何条带的引物有2条,能扩出条带但扩增图谱中没有特异性条带出现的引物有10条,能扩出条带且扩增图谱具有多态性的引物有8条。对筛选到的具有多态性的8条引物进行分析,共扩增出46个DNA片段,片段大小为0.2~3.2kb,其中5个菌株共有的片段有13条,显示多态性的片段有33条。

Abstract:Twenty random primers 10 bp in length were used in this experiment to study genetic (diversity) among five same serotype Salmonella strains from ducks by means of the random amplified polymorphic DNA(RAPD) method. Results showed that the primers were divided into three groups, i.e. Group 1 with 2 primers having no amplification products, Group 2 with 10 primers having no characteristic amplification products and Group 3 with 8 primers having characteristic amplification products with total 46 RAPD bands from 0.2 kb to 3.2 kb in length, 13 bands of which are common and 33 bands of which are polymorphic (polymorphic rate is 71.7%). The results mentioned above suggested that the RAPD method can be used for identification of the Salmonella strains mentioned above.
