摘    要:<正> 乌头碱是乌头属植物中所含的二萜双酯型生物碱类中的一种,成人口服纯品致死量仅约5mg,由乌头属植物引起的中毒与死亡屡见报导但一直缺乏较好的检测方法,使中毒后体内乌头碱分布与分解规律的研究难以进行,也给这类中毒案件的法医学鉴定带来一定困难。近年来应用HPLC分离检测生药中乌头碱类生物碱的方法已有报导。本文应用HPLC

Abstract:A HPLC method for the determination of aconitine in biological samples has been established. Column:C18 ODS 10μm,4.6mmlD×30cm with guard column. Mobile phase: methanol; acetonitrile: 0.02mol (NH_4)_2CO_3, 53: 35: 12. Variable-wave-length UV detector is used, measured at 235nm. Internal standard is Teteracaine. Samples preparation:Blood or homogenisedtissues extracted with e ther and purified by acid-base extraction,then condenced under reduced pressure with N_2 flow, De-tectable limit 10~(-8)g/ml. Only 1 ml. blood or homogenised tissues is required and corrupted samples can also be determined. Thismethod has been used to study the distribution of aconitine in liver, kindney, heart, and blood of rats orally poisoned by aconi-tine. The largest aconitine content is found in liver. The method is also used for study on decreasing of aconitine in liver at aperiod of 14 days after death under situation temperature 16±4℃ and the decreasing rate is found to be conformable with dc/dt= KC~2; the value of K determined in this paper is --0.043 ml·μg~(-1)·day~(-1).
