PR8流感病毒突变株的制备及其在鸡胚上生长特性的研究

摘    要:在前期发现流感病毒WSN株的NS2或NP蛋白突变使病毒在细胞上增殖能力明显提高的基础上,研究了这些突变是否会使PR8病毒效价在鸡胚上进一步提高的可行性,通过反向遗传操作技术获得了NS2E67S和NPP277A两株PR8突变病毒。经过比较突变病毒与野生病毒在鸡胚中的增殖能力,发现NS2E67S突变病毒在鸡胚上增殖后的效价与HA抗原表达量均明显提高,而NPP277A突变病毒在鸡胚上的增殖能力不及野生PR8病毒。该研究结果将对进一步研究病毒基因变异与宿主互作机制奠定基础,有可能通过NS2E67S突变病毒对改良流感疫苗具有重要的应用价值。

Abstract:In previous studies,we found that some A/WS/33(WSN) mutants on NS2 and NP proteins showed increasing replication abilities on MDCK cells.In order to characterize the amplification of PR8 mutants on these proteins in embryonated chicken eggs,the mutants were obtained by PCR-mutation and influenza virus reverse genetic technology.After being replicated in chicken embryonated eggs,the viral titer and HA titer of mutant NS2 E67S were higher than that of wt PR8,while these titers of NP P277A were lower than that...
