Abstract: | Objective To analyze differentially expressed genes at different time points after skin lesions usingbioinformatics approaches and to provide theoretical basis for wound age estimation in forensic medicine. Methods GSE23006 was downloaded from the gene expression omnibus(GEO), DEGs of skin lesions group at 1 d, 3 d, 5 d, 7 d and 10 d after lesions were screened by. online analsis tool GEO2 R, and the hub genes were screened by Cytoscape software,and protein network interaction analysis, GO and KEGG enrichment analysis were carried out on the selected hub genes. Results Compared with the control group, the hub genes of group 1 d were Cd44,Mmp9, Stat1, Ptprc, Il6, Tlr4, Tnf, Tlr2, Il1β;the hub genes of group 3 d were Mmp9, Stat1, Ptprc, Il6 and Il1β;the hub genes of group 5 d were Crp, Serpine1, Fos, Stat1, Jun, Mmp9, Cxcl10, Vcam1, Mmp3, Cxcl5, Spp1, Timp1 and Fn1;the hub genes of group 7 d were Krt83, Col1 a1 and Timp1;the hub genes of group 10 d were Crp, Fos, Ctgf, Il1 f6, Cxcl1, Il6, Actc1, Mmp13, Timp1, Cxcl2, Eln, Atf3 and Il1β. The GO function analysis of hub genes showed that it was associated with inflammatory response, immune regulation, tissue repair and tissue remodeling, while KEGG analysis showed that the pathways were related to apoptosis, inflammatory mediator regulation, and immune cell receptor signaling pathways. Hub genes play an important role in different stages of skin healing process and are correlated with wound age. Conclusion The identified hub genes at different time pionts could be applied as specific expressed genes at corresponding time pionts, which provides the basis and ideas for further research on skin lesions and the estimation of the lesion time. |
