摘    要:综述了原始串联亲和纯化方法的原理及其存在的缺陷和各种最新构建的串联亲和纯化标签组合及其从哺乳动物细胞中纯化蛋白复合物时的蛋白得率。与单步纯化方法相比,串联亲和纯化方法显著地减少了非特异性背景,可分离到高纯度的蛋白复合物。这一优势极大地简化了蛋白质鉴定及其相互作用关系的验证工作。

Abstract:The paper reviews the principle and limitations in the original tandem affinity purification(TAP) method,the newly developed TAP tags and their high recovery of protein complexes from mammalian cells.Compared with single-step purification,TAP method greatly reduces non-specific background and isolates protein complexes with higher purity.This dominant feature greatly simplifies identification and validation of the isolated proteins as true interacting partners.
