高浓度口蹄疫病毒抗原的稀释试验

摘    要:用预备试验初选的 4种稀释剂a、b、c、d ,对经浓缩培养法繁殖的牛、猪O型口蹄疫病毒进行稀释 ,使其与普通毒 (对照 )浓度相当。然后将其与普通毒一起在不同温度下放置不同时间后 ,测定TCID50 ,并对测定结果进行方差分析和多重比较。结果显示 ,4种稀释剂稀释的病毒TCID50 存在显著差异 ;a和b稀释的病毒TCID50 之间无显著差异 ,但均显著高于c和d稀释的病毒TCID50 ;a和b稀释的病毒与普通对照毒的TCID50 也无显著差异。表明 ,a和b均可用于稀释口蹄疫病毒抗原 ,考虑到成本 ,宜选用b。

Abstract:A method of dilution of concentrated foot-and-mouth disease (FMD) virus antigen was (developed) in this experiment . The concentrated virus antigens of the vaccine strains from bovine and pig,which were produced in large-scale monolayer BHK-21 cell systems by the concentrating propagation (respectively), were diluted with 4 diluents (a,b,c and d) selected by preliminary test so as to make them be the same concentration as the antigens prepared by the routine propagation. After the concentrated virus antigens were preserved for a period at different temperatures, TCID_(50) of them were measured and analyzed statistically by the variance analysis and the multiple comparison. The results were as follows: (1) TCID_(50) of the antigen diluted with a was not significantly different from that of the antigen diluted with b (P>(0.05)). (2) TCID_(50) of the antigens diluted with a or b were significantly higher than those of the antigens diluted with c or d, and were not significantly different from that of the control antigen. It was concluded from the above-(mentioned) results that the diluents a and b can be used to dilute FMD virus antigen, but b is more suitable than a for the sake of low cost.
