摘    要:利用DNA重组技术,由pUC19-MYO质粒中获得MYO小卫星DNA探针片段,将其插入到具有RNA聚合酶启动子的pGEM-4Z质粒的多克隆位点,从而构建了pGEM-4Z一MYO亚克隆,为制备RNA探针并将其应用于DNA指纹检测,提高DNA指纹的敏感性和小卫星DNA多态性的检出率奠定了基础。

Abstract:By means of DNA recombinant techniques, the fragment of MYO minisatellite DNA probe 0b-tained from plasmid pUC19 - MYO was inserted into plamsid pGEM - 4Z containing RNA poly-merase promotor, thus a sub-clone pGEM- 4Z-MYO was constructed then. It provided the RNAprobe for the DNA fingerprinting and laid a foundation for raising the sensitivity of detecting the poly-morphism of the minisatel1ite DNA.
