孕酮对大鼠小肠黏膜结构及其相关细胞的影响

摘    要:为了研究孕酮对小肠黏膜屏障结构的影响,将24只Wistar健康雌性大鼠进行同期发情处理后,于发情间情期随机分为4组。对照组:只做假手术;OVX组:双侧卵巢摘除;OVX P1组:双侧卵巢摘除 孕酮2 mg/kg;OVX P2组:双侧卵巢摘除 孕酮10 mg/kg,连续用药7 d后宰杀,观察小肠黏膜上皮细胞、上皮内淋巴细胞、杯状细胞形态结构和数量的变化。结果表明,和对照组相比,OVX组大鼠的小肠黏膜结构有明显损伤,绒毛萎缩,绒毛高度和隐窝深度的比值(V/C)明显降低,上皮内淋巴细胞数量和杯状细胞的数量减少;补充2种剂量的孕酮后,各项指标比OVX组有不同程度的改善,其中OVX P1组与OVX组差异显著,且好于OVX P2组。证实,卵巢摘除对大鼠小肠黏膜屏障结构有严重影响,补充适量孕酮可以在一定程度上减轻对黏膜屏障结构的影响。

Abstract:To explore the effect of progesterone on the small intestinal mucosal barrier function of rat,twenty-four female Wistar rats were induced to oestrous synchronization and then were divided into four groups during the dioestrus: Control group(sham operation),OVX group(bilateral ovariectomy),OVX P1 group(bilateral ovariectomy and injected with 2 mg/kg progesterone,once a day),OVX P1 group(bilateral ovariectomy and injected with 10 mg/kg progesterone,once a day).A week later,the rats in each group were sacrificed and their morphology of mucous epithelium cells and the number of intraepithelial lymphocytes(IELs) and goblet cells(GCs) were observed by optical microscope.In the OVX group,the villus was atrophic,the ratio of villus height to depth of crypt was significantly decreased,and the counts of IELs and GCs were also decreased.There was significant difference between the control group and the OVX group,or between the OVX group and the OVX P1 group.There was difference between the OVX group and the OVX P2 group,but some indexes were not significant.The results indicated that the small intestinal mucosal barrier function was damaged greatly in the ovariectomy rats,and proper dosage of progesterone(2 mg/kg each day for 7 days) would improve the small intestinal mucosal barrier function in the rats removed bilateral ovariectomy to a certain extent.
