犊牛腹泻大肠杆菌O15菌蜕的制备及免疫原性研究

摘    要:为了制备犊牛腹泻大肠杆菌O15菌株菌蜕并研究其免疫原性,克隆噬菌体PhiX174裂解基因E,并构建温控溶菌表达载体。制备HBTN3菌蜕,并研究其免疫原性。结果显示:成功构建了温控溶菌表达载体pBVE,并制备了HBTN3菌蜕。最大细菌裂解率是99.87%,剩余的活菌被冷冻干燥并失活。安全性试验结果证实是安全的。用扫描电子显微镜观察到菌蜕的形态完整且内容物排出。小鼠接种菌蜕会产生良好的免疫反应和保护,保护率为80%。该研究结果为防控犊牛腹泻大肠杆菌疾病奠定了基础。

Abstract:In order to prepare the diarrheal calves Escherichia coli O15 strain ghost and study its immunogenicity,the phage Phi X174 lyse gene E was cloned,and a temperature-controlled lysate expression vector was constructed.The HBTN3 bacterium was prepared and its immunogenicity was studied.The results showed that the temperature-controlled lysate expression vector p BVE was successfully constructed,and the HBTN3 bacterial ghosts were prepared.The maximum bacterial lysis rate was 99.87%,and the remaining viable bacteria were freeze-dried and inactivated.The results of the safety experiments proved to be safe.Scanning electron microscopy showed that the morphology of the bacteria was complete and the contents were discharged.Mice inoculated with the ghosts will have a good immune response and protection,the protection rate was 80%.The results of this study lay the foundation for the prevention and control of diarrheal calves E.coli disease.
